The medial septal-medial habenula cholinergic circuit: A new mechanism of exercise improving cognitive function in AD mice.

BACKGROUND: Alzheimer's disease (AD) is a progressively and incurably neurodegenerative disorder with cognitive dysfunction (e.g., memory de … al activity. Against this background, we aimed to investigate whether the medial septum (MS)-medial habenula (MHb) cholinergic circuit mediated the effects of combined (aerobic and resistance) exercise intervention on cognitive function in AD mice. METHODS: Six-week-old male C57BL/6 J wild-type (WT) mice were randomly divided into the sham-operated contro …

Effects of predator odor stress on alcohol-induced conditioned taste aversion and lateral habenula cFos expression in rats.

Stress-related disorders and alcohol use disorder (AUD) frequently co-occur, yet the mechanisms linking stress coping styles to alcohol sensitivity remain unclear. Here, we tested whether acute stress exposure alters sensitivity to alcohol's aversive effects, an important factor in regulating drinking, and alcohol-induced neural activation in the lateral habenula (LHb), a brain region implicated in negative valence processing. Male and female rats were exposed to acute predator odor stress and classified as Avoiders or Non-Avoiders based on post-stress avoidance behavior. Alcohol-induced condi…

Dysregulation of kappa opioid receptor neuromodulation of lateral habenula synaptic function following a repetitive mild traumatic brain injury.

Mild traumatic brain injury (mTBI) increases the risk of affective disorders, anxiety and substance use disorder. The lateral habenula (LHb) plays an important role in pathophysiology of psychiatric disorders. Recently, we demonstrated a causal link between mTBI-induced LHb hyperactivity due to excitation/inhibition (E/I) imbalance and motivational deficits in male mice using a repetitive closed head injury mTBI model. A major neuromodulatory system that is responsive to traumatic brain injuries, influences affective states and also modulates LHb activity is the dynorphin/kappa opioid receptor…

Epigenetic regulation of immediate-early gene Nr4a2/Nurr1 in the medial habenula during reinstatement of cocaine-associated behavior.

Propensity to relapse following long periods of abstinence is a key feature of substance use disorder. Drugs of abuse, such as cocaine, caus … netic function and behavior, we investigated HDAC3-mediated regulation of Nr4a2/Nurr1 in the medial habenula, an understudied pathway in cocaine-associated behaviors. Nr4a2, a transcription factor critical in cocaine-associated behaviors and necessary for MHb development, is enriched in the cholinergic cell-population of the MHb; yet, the role of NR4A2 wit …

Tacr3 in the lateral habenula differentially regulates orofacial allodynia and anxiety-like behaviors in a mouse model of trigeminal neuralgia.

Trigeminal neuralgia (TN) is debilitating and is usually accompanied by mood disorders. The lateral habenula (LHb) is considered to be involved in the modulation of pain and mood disorders, and the present study aimed to determine if and how the LHb participates in the development of pain and anxiety in TN. To address this issue, a mouse model of partial transection of the infraorbital nerve (pT-ION) was established. pT-ION induced stable and long-lasting primary and secondary orofacial allodynia and anxiety-like behaviors that correlated with the increased excitability of LHb neurons. Adeno-a…

Distinct and shared structural covariance reductions of the habenula in first-episode drug-naive and recurrent major depressive disorder.

Emerging evidence suggests that habenula (Hb) dysfunction plays an important role in major depressive disorder (MDD), with prior functional and structural studies implicating Hb-related networks in emotional and reward processing. However, it remains unclear whether structural covariance of Hb differs between first-episode drug-naive (FEDN) and recurrent MDD (RMDD). T1-weighted structural MRI data were obtained from the REST-meta-MDD consortium, including 839 MDD patients (346 FEDN and 261 RMDD) and 788 healthy controls. Single-subject structural covariance networks were constructed via Kullba…

Foxg1 Modulation of the Prkcd Gene in the Lateral Habenula Mediates Trigeminal Neuralgia-Associated Anxiety-Like Behaviors in Mice.

Trigeminal Neuralgia (TN) is a debilitating disorder frequently accompanied by mood complications such as depression and anxiety. The curren … bilateral microinjection of adeno-associated virus harboring Foxg1-specific shRNA into the lateral habenula (LHb), resulted in a significant attenuation of both orofacial pain and anxiety-like behaviors. Subsequent RNA sequencing implicated Prkcd as a downstream effector gene modulated by Foxg1. Pharmacological inhibition of protein kinase C delta, encode …

Neuronal IL-18 Signaling in the Medial Habenula Contributes to Schizophrenia-Like Behavioral Deficits via ERK-Associated BDNF Suppression.

Current antipsychotics for schizophrenia (SCZ) are notably ineffective against the debilitating cognitive and negative symptoms, creating a major therapeutic gap. Neuroinflammation in the medial habenula (MHb) is implicated in SCZ; however, the underlying cell-type-specific mechanisms remain unknown. This study aimed to determine whether pathologically elevated neuronal interleukin-18 (IL-18) in the MHb contributes to schizophrenia-like behavioral abnormalities through ERK-associated downregulation of brain-derived neurotrophic factor (BDNF). Utilizing an MK-801-treated rat model, we adopted a…

Lateral habenula lesions improve the behavioral response in depressed rats via increasing the serotonin level in dorsal raphe nucleus.

The dorsal raphe nucleus (DRN)-serotonin (5-HT) system plays a key role in stress-related psychiatric disorders such as anxiety and depression. The habenular nucleus (Hb) is closely connected with the DRN both morphologically and functionally. Here, we used two types of depressive animal models by exposing rats to chronic mild stress (CMS) and by chronically administering the tricyclic antidepressant clomipramine (CLI) in the rat during the neonatal state of life to produce adult depressed rats. We investigated the effects of lateral habenular nucleus (LHb) lesions on the behavioral response a…

Effects of Repetitive Mild Traumatic Brain Injury on Corticotropin-Releasing Factor Modulation of Lateral Habenula Excitability and Motivated Behavior.

Mild traumatic brain injury (mTBI) is a significant health burden due to mTBI-related chronic debilitating cognitive and psychiatric morbidities. Recent evidence from our laboratory suggests a possible dysregulation within reward/motivational circuit function at the level of a subcortical structure, the lateral habenula (LHb), where we demonstrated a causal role for hyperactive LHb in mTBI-induced motivational deficits in self-care grooming behavior in young adult male mice when exposed to mTBI during late adolescence (at ∼8 weeks old). In this study, we extended this observation by further ch…

Disrupted functional connectivity of the habenula links psychomotor retardation and deficit of verbal fluency and working memory in late-life depression.

BACKGROUND: Functional abnormalities of the habenula in patients with depression have been demonstrated in an increasing number of studies, and the habenula is involved in cognitive processing. However, whether patients with late-life depression (LLD) exhibit disrupted habenular functional connectivity (FC) and whether habenular FC mediates the relationship between depressive symptoms and cognitive impairment remain unclear. METHODS: Overall, 127 patients with LLD and 75 healthy controls were recruited. The static and dynamic FC between the habenula and the whole brain was compared between LL…

Morphometric analysis of the cerebral expression of ATP-binding cassette transporter protein ABCB1 in chronic schizophrenia: Circumscribed deficits in the habenula.

There is increasing evidence that microvascular abnormalities and malfunction of the blood-brain barrier (BBB) significantly contribute to s … xpressing capillaries between all patients with schizophrenia and control cases. The left and right habenular complex, however, showed significantly reduced capillary densities in schizophrenia patients. In addition, we found a significantly reduced density of ABCB1-expressing neurons in the left habenula. Reduced ABCB1 expression in habenular capillaries …

Tongmai Yishen Formula alleviates post-stroke depression by restoring neuronal homeostasis in the lateral habenula via the ITPKA signaling pathway.

BACKGROUND: Post-stroke depression (PSD) is a common and debilitating complication of stroke, characterized by persistent abnormalities in neuronal excitability and synaptic plasticity. The lateral habenula (LHb) has emerged as a key regulatory hub in the pathophysiology of PSD. Tongmai Yishen Formula (TMYSF) has demonstrated clinical efficacy in alleviating PSD symptoms, yet its mechanism in restoring the excitability-plasticity balance remains unclear. PURPOSE: We aimed to determine whether TMYSF alleviates PSD by modulating the inositol-trisphosphate 3-kinase A (ITPKA)-mediated beta isofor…

Lateral Habenula 5-HT₂A Receptors Mediate Social Deficits in a Valproic Acid-Induced Rodent Model of Autism Spectrum Disorder.

Autism Spectrum Disorder (ASD) is characterized by social deficits, yet underlying neural mechanisms remain poorly understood. Using a valproic acid (VPA)-induced rodent model of ASD, we demonstrate that hyperactivity of lateral habenula (LHb) neurons, driven by upregulated 5-HT₂A receptors, mediates social impairments. VPA-exposed rats showed significant reductions in sociability, increased anxiety-like behaviors, and elevated repetitive actions. These behavioral changes coincided with enhanced LHb neuronal activity, including increased c-Fos expression and spontaneous firing rates. Transcrip…

Functional connectivity between the habenula and posterior default mode network contributes to the response of the duloxetine effect in major depressive disorder.

This study aims to investigate the functional connectivity (FC) changes of the habenula (Hb) among patients with major depressive disorder (MDD) after 12 weeks of duloxetine treatment (MDD12). Patients who were diagnosed with MDD for the first time and were drug-naïve were recruited at baseline as cases. Healthy controls (HCs) matched for sex, age, and education level were also recruited at the same time. At baseline, all participants underwent resting-state functional MRI. FC analyses were performed using the Hb seed region of interest, and three groups including HCs, MDD group and MDD12 grou…

Mediation of lateral hypothalamus orexin input to lateral habenula in the inhibitory effects of mechanical stimulation on psychomotor responses induced by cocaine.

INTRODUCTION: The lateral hypothalamus (LH) plays an important physiological role in brain function and also plays an important role in substance abuse. The neuropeptides called orexin (or hypocretins) have been identified as being located exclusively in the cell bodies of the LH. Our previous studies have demonstrated that mechanical stimulation (MS) of the ulnar nerve produces strong inhibitory effects on cocaine addiction-like behaviors through activation of LH projection to the lateral habenula (LHb). METHODS: Therefore, the present study hypothesized that ulnar MS would suppress the psyc…

Chemogenetic inhibition of lateral habenula projections to the dorsal raphe nucleus reduces passive coping and perseverative reward seeking in rats.

The lateral habenula (LHb) processes information about aversive experiences that contributes to the symptoms of stress disorders. Previously, we found that chemogenetic inhibition of rat LHb neurons reduced immobility in the forced swim test, but the downstream target of these neurons was not known. Using an intersectional viral vector strategy, we selectively transduced three different output pathways from the LHb by injecting AAV8-DIO-hM4Di into the LHb and CAV2-CRE (a retrograde viral vector) into one of the three target areas as follows: dorsal raphe nucleus (DRN), ventral tegmental area (…
