Histidine-derived N-heterocyclic Carbenes in Metal Peptide Conjugates for Abiotic Catalysis.

Histidinium (Hum), a N,N'-dimethylated variant of histidine, serves as a non-canonical amino acid that provides an N-heterocyclic carbene (NHC) binding site for metal coordination. This amino acid therefore allows for combining peptides and proteins with NHC metal complexes, one of the most prolific scaffolds of homogeneous catalysis of the recent past. Here, we review our efforts in combining these two areas by constructing chemically synthesized tetrapeptide NHC rhodium conjugates for hydrosilylation and larger peptide metal systems obtained from solid phase peptide synthesis for abiotic iri

Reduced Histidine Metabolism Is Associated with Early Allograft Dysfunction Following Liver Transplantation.

Background/Objectives: Early allograft dysfunction (EAD) is a common complication after liver transplantation and is associated with inferior graft survival. While normothermic machine perfusion (NMP) has reduced EAD incidence, the prediction of early graft performance prior to implantation remains elusive. We aimed to correlate the peri-transplant energetic and metabolic profile of liver grafts with post-transplant outcome in a cohort that included grafts preserved with NMP. Methods: Sequential biopsies were taken from 20 transplanted livers (10 immediate graft function [IGF] and 10 EAD), pre

Conserved histidine residues and the control of tick-borne encephalitis virus maturation.

The maturation and infectivity of orthoflaviviruses is driven by interactions and structural changes of the two envelope proteins, prM and E, which are controlled by protonation and deprotonation events during the viral life cycle when the virus encounters acidic and neutral pH environments inside and outside the cell. Histidine residues act as sensors for these pH-induced conformational transitions, which differ between mosquito-borne and tick-borne orthoflaviviruses in some aspects. We identified six histidine residues that are conserved among the envelope proteins of mammalian tick-borne or

Tweaking Polybia-MP1: How a Lysine-Histidine Swap Redefines Its Surface Properties.

Background/Objectives: Polybia-MP1 (MP1) exhibits antimicrobial and anticancer properties. To improve selectivity toward acidic tumor microenvironments, we designed HMP1, a histidine-substituted analog of MP1, aiming to introduce pH-responsive behavior within physiological and pathological pH ranges. Methods: HMP1 was synthesized by replacing all lysine residues in MP1 with histidines. We characterized its surfactant properties and interactions with lipid monolayers composed of DPPC under varying pH and ionic strength conditions. Langmuir monolayer experiments were used to evaluate peptide-ind

Histidine-rich glycoprotein ameliorated lung ischemia-reperfusion injury in a mouse model.

BACKGROUND: Histidine-rich glycoprotein (HRG) is a multifunctional plasma glycoprotein involved in the regulation of various biological processes. Decreased plasma levels of HRG at 72 h after lung transplantation have been shown to be associated with severe primary graft dysfunction and poor transplant outcomes. In this study, we examined the effect of HRG supplementation on lung ischemia-reperfusion injury (IRI) in mice. METHODS: The experimental mice were divided into a Sham group and three IRI groups (n=5 each) that received phosphate-buffered saline, human serum albumin, or HRG (HRG/IRI g

The VanS sensor histidine kinase from type-B VRE recognizes vancomycin directly.

UNLABELLED: V ancomycin- r esistant e nterococci (VRE) are among the most common causes of nosocomial infections and have been prioritized as targets for new therapeutic development. Many genetically distinct types of VRE have been identified; however, they all share a common suite of resistance genes that function together to confer resistance to vancomycin. Expression of the resistance phenotype is controlled by the VanRS two-component system. This system senses the presence of the antibiotic, and responds by initiating transcription of resistance genes. VanS is a transmembrane sensor histid

Slow clearance of histidine-rich protein-2 in Gabonese with uncomplicated malaria.

UNLABELLED: Malaria rapid diagnostic tests (RDTs), which detect Plasmodium falciparum (Pf)-specific histidine-rich protein-2 (HRP2), have increasing importance for the diagnosis and control of malaria, especially also in regions where routine diagnosis by microscopy is not available. HRP2-based RDTs have a similar sensitivity to expert microscopy, but their reported low specificity can lead to high false positivity rates, particularly in high-endemic areas. Despite the widespread use of RDTs, models investigating the dynamics of HRP2 clearance following Pf treatment focus rather on short-term

Histidine Metabolic Pathway Contributes to Clozapine-Induced Sialorrhea Based on Nontargeted Metabolomics.

INTRODUCTION: Clozapine-induced sialorrhea (CIS) is one of the most common side effects of clozapine use, while the mechanism remains unclear. METHODS: A total of 51 schizophrenia patients taking clozapine were selected. Among them, 32 had sialorrhea, and 19 had no sialorrhea. Saliva metabolites were identified using ultra-high-performance liquid chromatography-MS/MS (UHPLC-MS/MS), and the differences in saliva metabolites in each group were analyzed through qualitatively searching HMDB, KEGG, and self-built databases, combined with multivariate statistics. After further evaluation by receive

Histidine triad nucleotide-binding protein 2: From basic science to clinical implications.

Histidine triad nucleotide-binding protein 2 (HINT2) is a dimeric protein that belongs to the histidine triad protein superfamily, predominantly expressed in the liver, pancreas, and adrenal gland, and localised to the mitochondrion. HINT2 binds nucleotides and catalyses the hydrolysis of nucleotidyl substrates. Moreover, HINT2 has been identified as a key regulator of multiple biological processes, including mitochondria-dependent apoptosis, mitochondrial protein acetylation, and steroidogenesis. Genetic manipulation has provided new insights into the physiological roles of HINT2 in several p

Harnessing Aromatic-Histidine Interactions through Synergistic Backbone Extension and Side Chain Modification.

Peptide engineering efforts have delivered drugs for diverse human diseases. Side chain alteration is among the most common approaches to designing new peptides for specific applications. The peptide backbone can be modified as well, but this strategy has received relatively little attention. Here we show that new and favorable contacts between a His side chain on a target protein and an aromatic side chain on a synthetic peptide ligand can be engineered by rational and coordinated side chain modification and backbone extension. Side chain modification alone was unsuccessful. Binding measureme

Discovery of SARS-CoV-2 Inhibitors Featuring Novel Histidine α-Nitrile Motif.

As COVID-19 infection caused severe public health concerns recently, the development of novel antivirals has become the need of the hour. Main protease (Mpro ) has been an attractive target for antiviral drugs since it plays a vital role in polyprotein processing and virus maturation. Herein we report the discovery of a novel class of inhibitors against the SARS-CoV-2, bearing histidine α-nitrile motif embedded on a simple dipeptide framework. In-vitro and in-silico studies revealed that the histidine α-nitrile motif envisioned to target the Mpro contributes to the inhibitory activity. Among a

A simple protein histidine kinase activity assay for high-throughput inhibitor screening.

Bacterial two-component systems (TCSs), which typically consist of a sensor histidine kinase (HK) and a response regulator (RR), have been investigated as attractive antibacterial drug targets. Unfortunately, current HK activity assays based on the quantification of autophosphorylated HKs are hampered by the instability of the phosphohistidine (pHis) product, rendering them ill-suited for high-throughput screenings. To address this challenge, we developed a simple HK activity assay using readily available reagents, which we have termed AUDECY (AUtophosphorylation-DEphosphorylation CYcle assay)

Intracerebroventricular administration of histidine reduces kainic acid-induced convulsive seizures in mice.

Kainic acid (KA)-induced seizures and other experimental models of epilepsy have been proven to be instrumental in identifying novel targets that could be responsible for human icto- and epileptogenesis. We have previously shown that the ablation of pharmacoresistant voltage-gated Ca2+ channels with Cav2.3 as central ion-conducting pore (R-type Ca2+ channel) reduces the sensitivity towards KA-induced epilepsy in mice. In vivo, Cav2.3 channels are thought to be under tight allosteric control by endogenous loosely bound trace metal cations (Zn2+ and Cu2+) that suppress channel gating via a high-

Histidine decarboxylase deficiency causes tourette syndrome: parallel findings in humans and mice.

Tourette syndrome (TS) is characterized by tics, sensorimotor gating deficiencies, and abnormalities of cortico-basal ganglia circuits. A mutation in histidine decarboxylase (Hdc), the key enzyme for the biosynthesis of histamine (HA), has been implicated as a rare genetic cause. Hdc knockout mice exhibited potentiated tic-like stereotypies, recapitulating core phenomenology of TS; these were mitigated by the dopamine (DA) D2 antagonist haloperidol, a proven pharmacotherapy, and by HA infusion into the brain. Prepulse inhibition was impaired in both mice and humans carrying Hdc mutations. HA i

Clinicopathological significance of fragile histidine triad transcription protein expression in endometrial carcinomas.

Abnormalities in structure and expression of the fragile histidine triad transcription (FHIT) gene have been reported in a variety of cancers, including endometrial cancers. A good correlation between FHIT gene alteration and loss of Fhit expression was observed in endometrial cancers, although those are the selected cases. Therefore, we investigated the association of Fhit expression with clinicopathological features in 111 cases of endometrial cancer. Loss of Fhit expression was associated with high malignant potential, including extensive muscular invasion, advanced surgical stage, high his

Comparison between activation of ornithine decarboxylase and histidine decarboxylase in rat stomach.

We compared the responses of rat stomach ornithine decarboxylase (ODC) and histidine decarboxylase (HDC) to food intake, oral treatment with antisecretagogues, NaHCO3, and hypertonic NaCl, antrectomy, intravenous infusion of gastrin-17, the selective cholecystokinin (CCK)-B/gastrin receptor antagonist L-365,260, and the somatostatin analogue RC-160. The serum gastrin concentration and oxyntic mucosal ODC and HDC activities were higher in freely fed rats than in fasted rats. Food intake in fasted rats raised the serum gastrin concentration and the ODC and HDC activities. Ranitidine, omeprazole,

Novel bacterial P-type ATPases with histidine-rich heavy-metal-associated sequences.

Menkes disease and Wilson disease are human disorders of copper metabolism. It has recently been shown that both are due to mutations in P-type ATPase copper transport molecules. Related heavy metal transporting ATPases have been described in several strains of bacteria. In an effort to isolate other mammalian metal transporters, we screened a human small intestine library with probes homologous to conserved sequences in the known proteins. Two novel cDNAs were isolated, which encode new members of this family. Surprisingly, they were both of bacterial origin, most likely derived from E. coli
