Pseudorabies virus (PRV) is an alphaherpesvirus that causes a neurological disease in many wild and domestic animals. The neuropathology elicited by PRV is quite consistent regardless of the host with the only exception of mink, in which it is characterized by a vasculopathy rather than by an encephalitis. In this study, we aimed to investigate the underlying pathogenic mechanism(s) of PRV infection in mink by using immunohistochemistry and laser capture microdissection (LCM) on material from naturally and experimentally infected animals. The inflammatory reaction induced by PRV was minimal or

Comparative genomics were used to assess genetic differences between Staphylococcus aureus strains derived from infected animals versus colonized or infected humans. A total of 77 veterinary isolates were genetically characterized by high-throughput amplified fragment length polymorphism (AFLP). Bacterial genotypes were introduced in a large AFLP database containing similar information for 1,056 human S. aureus strains. All S. aureus strains isolated from animals in close contact with humans (e.g., pet animals) were predominantly classified in one of the five main clusters of the AFLP database

Pulmonary effects, lung clearance, and tissue retention of blood-borne Pseudomonas aeruginosa were compared in dogs (n = 5) and pigs (n = 5) during continuous 6-hour intravenous infusion of 1.2(10(9)) bacteria/min/20 kg. Control pigs received an equal volume of sterile saline. In contrast to controls, experimental pigs developed pulmonary artery (PA) hypertension (mean, 30 +/- SE 3; baseline, 17 mm Hg) and pulmonary failure manifested by hypoxemia (mean PaO2, 49 +/- 4; baseline, 78 +/- 2 mm Hg; p less than 0.001), increased intrapulmonary shunting (40 to 50%), noncardiogenic pulmonary edema, a

Microbial symbionts can affect host phenotypes and, thereby, ecosystem functioning. The microbiome is increasingly being recognized as an important player in the tripartite interaction between parasitic flatworms, snail intermediate hosts, and the snail microbiome. In order to better understand these interactions, transplant experiments are needed, which rely on the development of a reliable and reproducible protocol to obtain microbiome-disturbed snails. Here, we report on the first successful snail bacteriome transplants, which indicate that Biomphalaria glabrata can accrue novel bacterial a

Factors that drive parasite specificity and differences in infection dynamics among alternative host species are important for ecology and evolution of host-parasite interactions, but still often poorly known in natural systems. Here, we investigated spatiotemporal dynamics of infection, host susceptibility and parasite-induced changes in host phenotype in a rarely explored host-parasite system, the Australapatemon sp. trematode infecting two sympatric species of freshwater leeches, Erpobdella octoculata and Helobdella stagnalis. We show significant variation in infection abundance between the

Although the presence of cryptic host specificity has been documented in Blastocystis, differences in infection rates and high genetic polymorphism within and between populations of some subtypes (ST) have impeded the clarification of the generalist or specialist specificity of this parasite. We assessed the genetic variability and host specificity of Blastocystis spp. in wild howler monkeys from two rainforest areas in the southeastern region of Mexico. Fecal samples of 225 Alouatta palliata (59) and A. pigra (166) monkeys, belonging to 16 sylvatic sites, were analyzed for infection with Blas…

Human E-cadherin promotes entry of the bacterial pathogen Listeria monocytogenes into mammalian cells by interacting with internalin (InlA), a bacterial surface protein. Here we show that mouse E-cadherin, although very similar to human E-cadherin (85% identity), is not a receptor for internalin. By a series of domain-swapping and mutagenesis experiments, we identify Pro16 of E-cadherin as a residue critical for specificity: a Pro-->Glu substitution in human E-cadherin totally abrogates interaction, whereas a Glu-->Pro substitution in mouse E-cadherin results in a complete gain of function. Fimbrial adhesins are surface-associated bacterial proteins that contribute to colonization, tissue tropism, and biofilm formation. Genomic analysis of the avian pathogenic Escherichia coli (APEC) strain QT598 identified a plasmid-localized fimbrial operon, termed plf which encodes P-like (PL) fimbriae. Herein, we investigated the role of P-like (PL) fimbriae, encoded on the ColV plasmid of APEC strain QT598, in a natural host turkey respiratory infection model. We determined that deletion of the plf genes reduced colonization in the lungs of turkeys. The PlfG class II fimbrial adhesin from AP

We investigated the phenological variation of three genera of blood parasites of lizards (Schellackia, Karyolysus, and Lankesterella) using samples of 275 adult Psammodromus algirus and 138 Acanthodactylus erythrurus collected from April to September in 2021 and 2022. Both years experienced heatwaves, and 2022 had the warmest summer on historical record for the region. We captured lizards in two close areas that differed in vegetation quality due to differential human pressure. We found that Schellackia and Karyolysus more frequently infected P. algirus, whereas Lankesterella infected A. eryth

In North America, crown rust has emerged as a disease of Kentucky bluegrass (Poa pratensis) and may cause significant damage. After observing crown rust on Poa spp. in Minnesota and on three East Asian buckthorns (Rhamnus crenata, R. davurica, and R. japonica) in North Dakota, we sequenced one to four loci of nine samples for identification and to determine their placement within Puccinia series Coronata. Earlier studies had shown that the P. coronata var. avenae f. sp. avenae clade caused crown rust of Kentucky bluegrass in North America, but phylogenetic analysis identified our samples as a

Scale insects (Hemiptera: Coccomorpha) are important sap-feeding herbivores that influence plant physiology and contribute to forest ecosystem functioning. Despite their ecological importance, their host specificity and infestation dynamics remain insufficiently documented in Mediterranean semi-arid forests. This study investigated the distribution and ecological specialization of scale insects in Belezma National Park (Algeria), focusing on host tree species, plant communities, and infested plant parts. Significant variations (p = 0.006) in infestation rates were observed among host tree spec…

Freshwater parasitic copepods appear to exhibit great taxonomic diversity. However, little is known about gene flow between species or whether there is incongruence between morphological and phylogenetic species definitions. Additionally, little is known about what evolutionary factors may contribute to speciation across various lineages. The copepod genus Salmincola, which includes common ectoparasites of fishes in the family Salmonidae, is distributed throughout the northern hemisphere and is a good model to demonstrate limited taxonomic understanding. Much of the regular scholarly output re

The hepatitis C virus (HCV) is a major causative agent of hepatitis that may also lead to liver cancer and lymphomas. Chronic hepatitis C affects an estimated 2.4 million people in the USA alone. As the sole member of the genus Hepacivirus within the Flaviviridae family, HCV encodes a single-stranded positive-sense RNA genome that is translated into a single large polypeptide, which is then proteolytically processed to yield the individual viral proteins, all of which are necessary for optimal viral infection. However, cellular innate immunity, such as type-I interferon (IFN), promptly thwarts

Segmented filamentous bacteria (SFB) are known modulators of the mammalian immune system. Currently, the technology for investigating SFB culture in vitro is immature, and as a result, the mechanisms of SFB colonization and immune regulation are not yet fully elucidated. In this study, we investigated the gene diversity and host specificity of SFB flagellin genes. The fliC1 and fliC2 genes are relatively conserved, while the fliC3 and fliC4 genes are more variable, especially at the central and C-terminal regions. Host specificity analysis demonstrated that the fliC1 genes do not cluster toget…

Tsetse flies Glossina palpalis gambiensis and G. tachinoides are among the major vectors of sleeping sickness (Human African Trypanosomiasis-HAT) and nagana (African Animal Trypanosomiasis - AAT) in West Africa. Both riparian species occur sympatrically in gallery forests of south west Burkina Faso, but little is known of their interspecies relationships although different authors think there may be some competition between them. The aim of this study was to check if sympatric species have different strategies when approaching a host. A man placed in a sticky cube (1 m × 1 m × 1 m) and a stick

The mechanisms responsible for skin lesions during acute graft-vs.-host disease (aGVHD) after allogeneic bone marrow transplantation (BMT) are poorly understood. The exact role of various effector cell populations and "major" (particularly HLA-DP) or "minor" antigens as target molecules is not known. To investigate the nature of cells responsible for tissue injury, we cultured T cells from skin biopsy first with interleukin 2 (IL-2) alone and then in polyclonal activation conditions to avoid in vitro antigenic sensitization before specificity testing. We applied this method to two biopsies per
