Impact-Resistant Hydrogels Via Quaternary Ammonium-Regulated Networks.

Hydrogels are increasingly explored as structural candidates for impact-mitigation and protection systems because of their biocompatibility, softness, and design versatility. However, conventional hydrogels typically suffer from inadequate mechanical properties and environmental instability, which severely hamper their practical applications. Here we report the use of quaternary ammonium-regulated networks to overcome these limitations, without the need for cumbersome postprocessing or high energy consumption. The approach enables controlled structural organization in polymers containing regul…

Multiscale Biophysical Characterization of Ultra-Short Peptide Hydrogels.

Ultra-short peptide (USP) hydrogels have emerged as a simple yet innovative class of biomaterial. Small peptide sequences (≤8 amino acid residues) self-assemble to form hierarchical nanofibrillar networks, which are characterized by minimalist design, biocompatibility, and the ability to mimic native extracellular matrix (ECM). These characteristics make USP hydrogels an excellent candidate for various biomedical applications, including 3D bioprinting, disease modeling, and targeted drug delivery. However, a thorough understanding of their hierarchical assembly and function is necessary to ful…

Pathological Microenvironment-Guided Smart Hydrogels for Periodontitis Therapy.

Periodontitis is a chronic inflammatory disease initiated by dental plaque biofilms and mediated by dysregulated host immune responses, frequently resulting in irreversible alveolar bone resorption. Although mechanical debridement, local drug delivery, and antimicrobial therapies are widely applied in clinical practice, their therapeutic efficacy remains limited by insufficient drug retention, nonspecific drug release, antibiotic resistance, and inadequate periodontal tissue regeneration. Recently, smart hydrogels have emerged as promising platforms for periodontal therapy owing to their ECM-m…

Bamboo-inspired ultra-strong nanofiber-reinforced composite hydrogels.

Biological materials, such as bamboo, are naturally optimized composites with exceptional mechanical properties. Inspired by such natural composites, traditional methods involve extracting nanofibers from natural sources and applying them in composite materials, which, however, often results in less ideal mechanical properties. To address this, this study develops a bottom-up nanofiber assembly strategy to create strong fiber-reinforced composite hydrogels inspired by the hierarchical assembly of bamboo. Self-assembled chitosan-sodium alginate nanofibers (CSNFs) are combined with tannic acid (…

Mesoscale Modeling of Hydrogels Under Frictional Shear Stress.

Hydrogels are three-dimensional networks of hydrophilic polymers often used as a simplified model of hydrated biological materials, from cartilaginous joints to the ocular tear film. However, the lubrication mechanisms of hydrogels remain poorly understood, partly due to their complex polymeric structure, which creates blurred interfaces during sliding that are challenging to study experimentally. In this study, we employ dissipative particle dynamics (DPD) to investigate the frictional behavior of a polymeric hydrogel network sliding against a solid wall in an explicit viscous solvent. This c…

Tuneable microfibrillar collagen structures within dense chitosan hydrogels.

Chitosan-type I collagen hydrogels are paradigms of polysaccharide-protein assemblies with applications as biomaterials. However, preparing physical hydrogels combining them at comparable, high concentrations (>20 mg mL-1) within interpenetrated networks remains challenging. Here, we could combine chitosan and collagen solutions at 25 mg mL-1 to prepare two different types of concentrated hydrogels. When neutralized under ammonia vapours, mixed solutions form composite hydrogels, where collagen fibers exhibiting an unusual, branched morphology occupy a chitosan network porosity. In contrast, n…

Synthetic hydrogels support robust and reproducible cardiomyocyte differentiation.

Cardiomyocyte manufacturing from human pluripotent stem cells is limited by the variability of differentiation efficiencies, partly attributed to the widespread use of the tumor-derived substrate Matrigel. Here, we describe a screening approach to identify fully-defined synthetic PEG hydrogels that support iPSC-derived cardiac progenitor cell (iPSC-CPC) adhesion, survival, and differentiation into iPSC-derived cardiomyocytes (iPSC-CMs). Our PEG hydrogels supported superior iPSC-CM differentiation efficiency, with a 24% increase in cTnT expression, and greater reproducibility when compared to c…

Protein-based double-network hydrogels mimicking oral mucosa.

The oral mucosa plays a critical role in protecting the body from external threats and serves as a key site for drug absorption. However, ethical concerns and the high costs associated with animal models traditionally used for oral mucosa research have increased the demand for reliable alternatives. In this study, we developed two types of protein-based double-network hydrogels to replicate the mechanical and structural properties of buccal mucosa and hard palate, respectively. By incorporating polyprotein into the rigid network and elastin-like peptides as the loose network, we fabricated hyd…

Tunable Viscoelasticity of Alginate Hydrogels via Serial Autoclaving.

Alginate hydrogels are widely used as biomaterials for cell culture and tissue engineering due to their biocompatibility and tunable mechanical properties. Reducing alginate molecular weight is an effective strategy for modulating hydrogel viscoelasticity and stress relaxation behavior, which can significantly impact cell spreading and fate. However, current methods like gamma irradiation to produce low molecular weight alginates suffer from high cost and limited accessibility. Here, a facile and cost-effective approach to reduce alginate molecular weight in a highly controlled manner using se…

Modeling Acute Myeloid Leukemia Using StarPEG-Heparin Hydrogels.

Biomimetic semi-synthetic hydrogels formed from a combination of star-shaped poly(ethylene glycol) (starPEG) and the glycosaminoglycan, heparin, allows for the three-dimensional (3D) culture of various cells and tissues. In this chapter, we describe methods for the use of starPEG-heparin hydrogels to cultivate primary and immortalized human acute myeloid leukemia (AML) cells. The resulting 3D culture models allow for the study of AML development and response to chemotherapeutic agents.

Disodium Cromoglycate Templates Anisotropic Short-Chain PEG Hydrogels.

Anisotropic hydrogels have found widespread applications in biomedical engineering, particularly as scaffolds for tissue engineering. However, it remains a challenge to produce them using conventional fabrication methods, without specialized synthesis or equipment, such as 3D printing and unidirectional stretching. In this study, we explore the self-assembly behaviors of polyethylene glycol diacrylate (PEGDA), using disodium cromoglycate (DSCG), a lyotropic chromonic liquid crystal, as a removable template. The affinity between short-chain PEGDA (Mn = 250) and DSCG allows polymerization to tak…

Cell-Material Interactions in Covalent Adaptable Thioester Hydrogels.

Covalent adaptable networks (CANs) are polymeric networks with cross-links that can break and reform in response to external stimuli, including pH, shear, and temperature, making them potential materials for use as injectable cell delivery vehicles. In the native niche, cells rearrange the extracellular matrix (ECM) to undergo basic functions including migration, spreading, and proliferation. Bond rearrangement enables these hydrogels to mimic viscoelastic properties of the native ECM which promote migration and delivery from the material to the native tissue. In this work, we characterize thi…
