CD4+ T-cell epitope-based heterologous prime-boost vaccination potentiates anti-tumor immunity and PD-1/PD-L1 immunotherapy.

BACKGROUND: Antitumor therapeutic vaccines are generally based on antigenic epitopes presented by major histocompatibility complex (MHC-I) molecules to induce tumor-specific CD8+ T cells. Paradoxically, continuous T cell receptor (TCR) stimulation from tumor-derived CD8+ T-cell epitopes can drive the functional exhaustion of tumor-specific CD8+ T cells. Tumor-specific type-I helper CD4+ T (TH1) cells play an important role in the population maintenance and cytotoxic function of exhausted tumor-specific CD8+ T cells in the tumor microenvironment. Nonetheless, whether the vaccination strategy ta

Cellular immune response following homologous and heterologous BNT162b2 booster vaccination after primary immunization with BNT162b2 and CoronaVac respectively.

Several vaccines targeting SARS-CoV-2 were developed following the COVID-19 outbreak. However, subsequent studies have indicated that humoral immune responses wane over time, particularly with the emergence of new SARS-CoV-2 variants. There is limited understanding of the resilience of T cell-mediated immunity induced by COVID-19 vaccines. This prospective cohort study involved a population who completed two primary doses of either the Pfizer BNT162b2 mRNA vaccine (PP) or the Sinovac Coronavac inactivated virus vaccine (SS) followed by one homologous (PPP) or heterologous (SSP) booster dose of

Differential functional immune recall across homologous and heterologous COVID-19 vaccination regimens.

INTRODUCTION: Understanding how different COVID-19 vaccine combinations shape long-term immunity is essential for improving durability and guiding booster strategies. Despite extensive characterization of neutralizing antibodies, long-term memory B and T cell responses after homologous and heterologous vaccination regimens remain poorly understood. METHODS: In this study, peripheral blood mononuclear cells (PBMCs) from 50 individuals were analyzed 12 months after completion of a homologous two-dose primary COVID-19 vaccination series and had received a single booster dose (12-month follow-up)

Comprehensive analysis of immune responses in CLL patients after heterologous COVID-19 vaccination.

Patients with chronic lymphocytic leukemia (CLL) treated with B-cell pathway inhibitors and anti-CD20 antibodies exhibit low humoral response rate (RR) following SARS-CoV-2 vaccination. To investigate the relationship between the initial transcriptional response to vaccination with ensuing B and T cell immune responses, we performed a comprehensive immune transcriptome analysis flanked by antibody and T cell assays in peripheral blood prospectively collected from 15 CLL/SLL patients vaccinated with heterologous BNT162b2/ChAdOx1 with follow up at a single institution. The two-dose antibody RR w

Immune unresponsiveness to secondary heterologous bacterial infection after sepsis induction is TRAIL dependent.

Sepsis is the leading cause of death in most intensive care units, and patients who survive the hyperinflammation that develops early during sepsis later display severely compromised immunity. Not only is there apoptosis of lymphoid and myeloid cells during sepsis that depletes these critical cellular components of the immune system, but also the remaining immune cells show decreased function. Using a cecal-ligation and puncture (CLP) model to induce intra-abdominal polymicrobial peritonitis, we recently established a link between the apoptotic cells generated during sepsis and induction of se

Intranasal immunization with heterologously expressed polysaccharide protects against multiple Pseudomonas aeruginosa infections.

Surface-expressed bacterial polysaccharides are often immunodominant, protective antigens. However, these antigens are chemically and serologically highly heterogeneous, and conjugation to protein carriers is often necessary to enhance their immunogenicity. Here we show the efficacy of intranasal immunization of mice with attenuated Salmonella enterica serovar Typhimurium expressing the O antigen portion of Pseudomonas aeruginosa lipopolysaccharide. P. aeruginosa is an ideal model system because it can cause a myriad of localized and systemic infections. In particular, this bacterium is a lead

Studies on heterologous immunity in schistosomiasis. 6. Observations on cross-immunity to Ornithobilharzia turkestanicum, Schistosoma bovis, S. mansoni, and S. haematobium in mice, sheep, and cattle in Iran.

Experiments were carried out in mice, cattle, and sheep to investigate the possibility that heterologous immune reactions may occur between the schistosomes prevalent in man and domestic animals in Iran. Immunization with Ornithobilharzia turkestanicum from cattle produced a considerable degree of immunity in mice against challenge with Schistosoma bovis, S. haematobium, and S. mansoni. The results of immunizing cattle with O. turkestanicum, S. bovis, and S. haematobium were even more striking; there was a reduction of 30-40% in the number of adult worms and a proportionally greater reduction

Heterologous prime-boost immunization of AAV-based neoantigen cancer vaccine induces anti-tumor immunity to inhibit tumor growth and relapse.

Therapeutic cancer vaccines, which induce anti-tumor immunity by targeting specific antigens, constitute a promising approach to cancer therapy. Our previous work developed a novel engineered adenovirus-associated virus 2 (AAV2)-based tumor-specific (neoantigen) cancer vaccine to boost antitumor immunity in combination with radiotherapy, resulting in tumor regression and less distant metastasis. However, the therapeutic efficacy of constitutive vector-based vaccination may be limited by poor immunogenicity, pre-existing vector-specific immunity, and vaccine-induced vector-specific immunity. Saponin derivative and MPLA-adjuvanted intranasal protein nanoparticle vaccines induce potent mucosal immunity and durable heterologous influenza protection in mice.

Mucosal immunity is essential for defenses against respiratory infections, such as influenza. Identifying adjuvant formulations that enhance mucosal immune responses in the respiratory tract will facilitate the development of effective mucosal vaccines. In this study, we explored the mucosal application of a novel semisynthetic saponin derivative, VSA-2. An intranasal vaccine formulation, PMV, containing influenza double-layered protein nanoparticles (PNPs) with monophosphoryl lipid A (MPLA) and VSA-2 was compared with PNPs alone or PNPs adjuvanted with MPLA or VSA-2 in mice. Two doses of PMV

Disparate epitopes mediating protective heterologous immunity to unrelated viruses share peptide-MHC structural features recognized by cross-reactive T cells.

Closely related peptide epitopes can be recognized by the same T cells and contribute to the immune response against pathogens encoding those epitopes, but sometimes cross-reactive epitopes share little homology. The degree of structural homology required for such disparate ligands to be recognized by cross-reactive TCRs remains unclear. In this study, we examined the mechanistic basis for cross-reactive T cell responses between epitopes from unrelated and pathogenic viruses, lymphocytic choriomeningitis virus (LCMV) and vaccinia virus. Our results show that the LCMV cross-reactive T cell resp

PRRSV GP4 subunit vaccine combined with adenovirus heterologous prime-boost immunization strategy induced a significant immune response in mice.

BACKGROUND: The porcine reproductive and respiratory syndrome virus (PRRSV) continues to cause widespread infections in the pig industry worldwide. Currently, multiple PRRSV vaccine candidates are in preclinical or clinical trials, and each has different advantages and limitations. Glycoprotein 4 (GP4) is rich in epitopes, which can induce the body to produce neutralizing antibodies, plays a vital role in causing the host immune response, and is a key target for PRRSV vaccine development. In this study, we developed a novel candidate vaccine immunization strategy combining a subunit vaccine wi

Heterologous Prime-Boost Immunization with DNA Vaccine and Modified Recombinant Proteins Enhances Immune Response against Trueperella pyogenes in Mice.

Trueperella pyogenes (T. pyogenes) is a crucial opportunistic pathogen normally causing mastitis, abscesses and pneumonia in economically important ruminants. Although only one commercial vaccine of T. pyogenes is currently obtainable, its immunoprotective effect is limited. Pyolysin (PLO) is the most predominant virulence factor highly expressed in T. pyogenes and is an excellent target for the development of novel vaccines against T. pyogenes. In this study, we designed a heterologous prime-boost vaccination scheme combining a DNA vaccine pVAX1-PLO and a subunit vaccine His-PLO to maximize h

Deletion of the D345L gene attenuates ASFV and induces protection against homologous and heterologous challenge by enhancing host innate immunity.

African swine fever (ASF) is a highly contagious and lethal disease caused by African swine fever virus (ASFV), with a mortality rate approaching up to 100%. At present, the functions of multiple genes of ASFV remain to be elucidated. In this study, we demonstrate that pD345L is a late-expressed viral protein and localizes to the viral factory. The recombinant strain JX23-02ΔD345L was obtained from the genotype I/II strain JX23-02. In vitro assays demonstrated that the deletion of D345L remarkably impairs viral replication. Further analyses revealed that JX23-02ΔD345L significantly up-regulate

A heterologous prime-boost vaccination regimen using ORFF DNA and recombinant ORFF protein confers protective immunity against experimental visceral leishmaniasis.

OBJECTIVE: We describe the effectiveness of a prime-boost vaccination regimen using the open-reading frame (ORFF) gene from the LD1 locus of Leishmania donovani. METHODS: A group of BALB/c mice was immunized with the plasmid carrying the gene for ORFF (F/pcDNA 3.1) and given a booster dose of either the same DNA vaccine or a vaccine with a recombinant ORFF (rORFF) protein. Another group of BALB/c mice was immunized and given a booster dose of the rORFF protein vaccine. The protective efficacies of these vaccine formulations were compared after challenge with L. donovani stationary-phase proma

Heterologous SARS-CoV-2 Buccal Immunization with Oral Dissolving Films Generated a Strong Systemic and Mucosal Immunity in a Murine Model.

BACKGROUND: In response to the emergence of immune-evasive variants of SARS-CoV-2, this study explores a novel heterologous vaccination strategy using a microparticulate formulation approach that is delivered via oral dissolving film (ODF) formulations into the buccal cavity. Heterologous administration has the potential to generate cross-reactive antibodies, which can be especially beneficial against viruses with ever-mutating variants. Moreover, the microparticulate oral dissolving film-based vaccine approach is a non-invasive vaccine delivery platform. METHODS: The vaccine design incorpora

Heterologous Versus Homologous COVID-19 Boosters: Immune Response Outcomes in Renal Transplant Recipients.

We aimed to investigate the immune responses to homologous and heterologous COVID-19 booster vaccinations in renal transplant recipients (RTRs) and to identify factors affecting these responses. In this prospective multicenter observational study, we measured the antibody kinetics of 90 RTRs using the chemiluminescent microparticle immunoassay method. The mean age of participants was 45.2 ± 11.4 years, with 35.6% being female. On the 42nd day after the first vaccine dose, the median antibody level was 16.7 (IQR 2.5-249.5) AU/mL, and the seropositivity rate was 60% (n = 36). Mycophenolic acid (

Heterologous immunization strategy developed broadly reactive human monoclonal antibodies against the BK virus.

BK polyomavirus (BKV) causes polyomavirus-associated nephropathy (PyVAN) and polyomavirus-associated hemorrhagic cystitis (PyVHC) following kidney transplantation and allogeneic hematopoietic stem cell transplantation (HST). BKV strains fall into four distinct genotypes (BKV-I, -II, -III, and -IV) with more than 80% of individuals are seropositive against BKV-I genotype, while the seroprevalence of the other four genotypes is lower. PyVAN and PyVHC occurs in immunosuppressed (e.g. transplant recipients) or immunomodulated (e.g. pregnant women) individuals. In the case of transplant patients, t

Heterologous prime-boost immunization induces protection against dengue virus infection in cynomolgus macaques.

Currently licensed dengue vaccines do not induce long-term protection in children without previous exposure to dengue viruses in nature. These vaccines are based on selected attenuated strains of the four dengue serotypes and employed in combination for two or three consecutive doses. In our search for a better dengue vaccine candidate, live attenuated strains were followed by non-infectious virus-like particles or the plasmids that generate these particles upon injection into the body. This heterologous prime-boost immunization induced elevated levels of virus-specific antibodies and helped t

Heterologous immunization with BNT162b2 followed by mRNA-1273 in dialysis patients: seroconversion and presence of neutralizing antibodies.

INTRODUCTION: The vital renal replacement therapy makes it impossible for dialysis patients to distance themselves socially. This results in a high risk of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and developing coronavuris disease 2019, with excess mortality due to disease burden and immunosuppression. We determined the efficacy of a 100-µg booster of mRNA-1273 (Moderna, Cambridge, MA, USA) 6 months after two doses of BNT162b2 (BioNTech/Pfizer, Mainz, Germany/New York, USA) in 194 SARS-CoV-2-naïve dialysis patients.
