Safety of BCG vaccination and revaccination in healthcare workers.

BCG vaccination and revaccination are increasingly being considered for the protection of adolescents and adults against tuberculosis and, more broadly, for the off-target protective immunological effects against other infectious and noninfectious diseases. Within an international randomized controlled trial of BCG vaccination in healthcare workers (the BRACE trial), we evaluated the incidence of local and serious adverse events, as well as the impact of previous BCG vaccination on local injection site reactions (BCG revaccination). Prospectively collected data from 99% (5351/5393) of particip…

[Wells' cellulitis and bacterial necrotizing cellulitis induced by anakinra].

BACKGROUND: Anakinra is a recombinant form of the naturally occurring human interleukin-1 receptor antagonist. It is used in the treatment of rheumatoid arthritis. The most frequent side effects are injection site reactions, which seem to have a toxic mechanism. PATIENTS AND METHODS: We report two unusual cases of injection site reactions with anakinra: a woman presented Wells' cellulitis of the thigh and a man developed serious bacterial cellulitis distinguished by deep necrosis at the site of the latest anakinra subcutaneous injection. DISCUSSION: The cases are examples of serious side eff…

Necrotizing skin lesion and radial nerve palsy in a patient treated with glatiramer acetate.

Glatiramer acetate (GA) is an approved and well tolerated drug for the treatment of relapsing-remitting multiple sclerosis. We report a case of a 52 year-old man with psoriasis and relapsing-remitting multiple sclerosis who developed, after 21 months of GA treatment, an injection-site cutaneous necrosis that involved both subcutaneous and muscular tissue with massive edema, followed, 3 days later, by radial nerve palsy. After few days another similar lesion appeared in another injection-site. We hypothesize that cutaneous necrosis could be due to a local dis-immune reaction and, probably, psor

Adverse reactions to BNT162b2 mRNA COVID-19 vaccine in medical staff with a history of allergy.

BACKGROUND: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2, COVID-19) vaccination is progressing globally. Several adverse reactions have been reported with vaccination against COVID-19. It is unknown whether adverse reactions to COVID-19 vaccination are severe in individuals with allergies. METHODS: We administered the COVID-19 vaccine to the medical staff at Yamagata University Hospital from March to August 2021. Subsequently, we conducted an online questionnaire-based survey to investigate the presence of allergy and adverse reactions after vaccination and examine the associat

Effect of muscle injection site on preanaesthetic sedation in dogs.

The demeanour of 122 dogs was assessed, and a mixture of 0.025 mg/kg acepromazine and 0.3 mg/kg morphine was injected intramuscularly into one of four randomly assigned muscle sites (cervical epaxial, triceps brachii, middle gluteal and quadriceps femoris) and the dogs' reactions to the injections were assessed. The development of sedation and the occurrence of side effects were assessed after 10, 20 and 30 minutes, and each dog's reaction to venous catheterisation was scored. All the dogs became similarly sedated after 30 minutes. The degree of sedation and the incidence of side effects were

Skin reaction to adalimumab.

The tumor necrosis factor alpha (TNFalpha) inhibitors etanercept and infliximab have shown good clinical results in the treatment of rheumatoid arthritis and other autoimmune disorders. With these novel fusion proteins, immune-mediated side effects, among them various cutaneous reactions, have been encountered. We report herein the case of an erythema multiforme-like skin reaction to treatment with the monoclonal anti-TNFalpha antibody adalimumab in a patient with rheumatoid arthritis. The reaction occurred after the sixth injection and affected the palms and soles as well as the injection sit

Pathology resulting from the administration of a live attenuated anti-Schistosoma haematobium vaccine in baboons.

Baboons (Papio anubis) were injected in the leg muscle with 18,000 20 Krad irradiated schistosomula of Schistosoma haematobium. Four protocols were followed: single, primary injection; single injection into animals primed by patent S. haematobium infection; secondary vaccine injection following an earlier injection; and single injection following praziquantel treatment of infected animals. Injection of the putative vaccine elicited localized mixed inflammatory infiltration at the site of injection which was both intense and prolonged. Three grades of tissue reaction were seen: the relatively m

[Additive activity of IgE and IgGa anaphylactic antibodies in rats (author's transl)].

Anaphylactic antibodies of two different classes are produced in rats: IgE and IgGa. When injected into the skin of normal animals, IgE remains at the site of injection for two weeks or more, while IgGa diffuses out within hours. In order to determine whether the anaphylactic reaction produced by one class of antibody is modified by the simultaneous presence of the other class of antibody, sites were prepared in normal rats with IgE antibody and 24 hours later IgGa antibody was injected into the same sites. The passive cutaneous anaphylactic reactions was elicited 2 hours later by intravenous

Carcinosarcoma mimicking a feline injection-site sarcoma in a cat.

A 15-year-old spayed female domestic short-haired cat with cutaneous/subcutaneous well-circumscribed, alopecic mass approximately 25 × 30 mm in diameter, localized to the left shoulder region was brought to the veterinary surgery department. Despite the suggestive location and macroscopic appearance, feline injection-site sarcoma was not suspected based on the cytologic examination of fine-needle aspirates. The tumor was surgically resected, and tissue sections were evaluated microscopically. The tumor was found to be nonencapsulated with a distinct border between the neoplastic parenchyma and

Desensitization of rabbit skin by repeated exposure to UV-visible light of sites injected with Rose Bengal.

We have shown in a previous paper that irradiation of rabbit skin sites injected with Rose Bengal (RB) produces immediate increase in vascular permeability and accumulation of PMNs. Studies on the development of temporary tolerance and the biological parameters related to the development of such tolerant state by repeated exposure to light of RB-injected sites are reported here. The increase in VP and PMN migration induced by RB (10 nmol) are of an immediate nature, i.e., occur within the first 3 h of irradiation, and the reaction subsides gradually after 24 h. When such moderate insult is rep

Studies on the effect of systemic administration of sensitizers to guinea-pigs with contact sensitivity to inorganic metal compounds. V. Studies on the mechanism of the 'flare up' reaction.

The mechanism of the `flare up' reaction of previous epicutaneous skin test sites in guinea-pigs sensitized to potassium dichromate seems to be similar to that of the Arthus phenomenon and different from that of contact skin sensitivity. No circulating antibodies were found in the serum of these animals but it was possible to transfer the capacity of developing `flare up' reactions by an intradermal injection of peritoneal exudate cells from sensitized donors to normal recipients in which a `flare up' could be elicited by intravenously injecting the hapten 4 days later. `Flare up' reactions co

Using a Safety Checklist Tool to Reduce Skin Reactions and Adverse Events After Cosmetic Dermal Filler Injections.

The rising popularity of dermal filler injections has highlighted the need for standardized safety protocols to minimize common skin reactions and reduce adverse events. This quality-improvement project implemented the Assessment Cosmetic Injection Safety Tool (ACIST), a preinjection safety checklist tool, at a suburban medical aesthetics clinic. The ACIST aims to reduce common skin reactions, such as bruising, swelling, and redness, and to identify high-risk cases that require treatment postponement, thereby reducing adverse events. A pre- and postintervention design was conducted over a 12-w

Parkinson's disease and skin.

Parkinson's disease is associated with a variety of dermatologic disorders and the study of skin may provide insights into pathophysiological mechanisms underlying this common neurodegenerative disorder. Skin disorders in patients with Parkinson's disease can be divided into two major groups: 1) non-iatrogenic disorders, including melanoma, seborrheic dermatitis, sweating disorders, bullous pemphigoid, and rosacea, and 2) iatrogenic disorders related either to systemic side effects of antiparkinsonian medications or to the delivery system of antiparkinsonian therapy, including primarily carbid
