Anatomical evidence for glutamate and/or aspartate as neurotransmitters in the geniculo-, claustro-, and cortico-cortical pathways to the cat striate cortex.

Data obtained by using various experimental approaches suggest that in the mammalian brain, most neurons within the visual system projecting to the striate cortex employ excitatory amino acids as transmitters. In order to investigate further the neurotransmitter phenotype of the ipsilateral afferents to area 17 of the cat, we have injected D-[3H]-aspartate, a retrograde tracer which selectively reveals putative glutamatergic and/or aspartatergic pathways, into this area. Retrogradely labelled neurons were observed in the dorsal lateral geniculate nucleus, visual claustrum, cortical areas 18, 1

Thalamic interactions of cerebellum and basal ganglia.

Cerebellum and basal ganglia are reciprocally interconnected with the neocortex via oligosynaptic loops. The signal pathways of these loops predominantly converge in motor areas of the frontal cortex and are mainly segregated on subcortical level. Recent evidence, however, indicates subcortical interaction of these systems. We have reviewed literature that addresses the question whether, and to what extent, projections of main output nuclei of basal ganglia (reticular part of the substantia nigra, internal segment of the globus pallidus) and cerebellum (deep cerebellar nuclei) interact with ea

Participation of the thalamic CM-Pf complex in movement performance in patients with dystonia.

INTRODUCTION: The centrum medianum- parafascicular complex of the human thalamus has a critical influence on cortical activity and significantly influences somatosensory function, arousal, and attention. In addition to its cortical connections, this region of the intralaminar thalamic nuclei is also connected to motor areas of the basal ganglia and the brain stem. OBJECTIVE: The goal of this study was to identify movement-related neurons in the centrum medianum-parafascicular complex and analyze the changes in their activity during voluntary movements in patients with cervical dystonia. METH…

Stimulation of the central median thalamic nucleus for epilepsy.

Electrical stimulation of the centromedian thalamic nucleus (ESCM) has been used in cases of difficult to control seizures with multifocal onset in frontal and temporal lobes, as well as in cases of seizures with no evidence of focal onset, such as Lennox-Gastaut syndrome. The stimulation program consists of 1 min stimulation on one side, 4 min interval OFF stimulation and 1 min stimulation on the other side, alternating from one side to the other for 24 h, at 60-130 Hz, 2.5-5.0 V, 0.21-0.45 ms. 49 cases have been treated and followed for periods of 6 months to 15 years. Results indicate that

Calretinin-Positive Neurons in Central Medial Thalamus Drive Nociception and Anxiety via Projections to Anterior Insular Cortex in male mice.

The central medial thalamic nucleus (CM), a key component of the thalamic intralaminar nuclei, receives nociceptive inputs from the lateral parabrachial nucleus and is implicated in pain processing. However, the specific neuronal subtypes and downstream pathways that mediate these functions remain poorly defined. Here, we combined tract tracing, immunofluorescence, in situ hybridization, chemogenetics, fiber photometry, and behavioral tests to investigate the role of calretinin-positive (CR+) neurons in the CM (CMCR+) and their projections to the anterior insular cortex (aIC) in nociceptive tr

Organization of ascending spinal projections in Caiman crocodilus.

Ascending spinal projections in the caiman (Caiman crocodilus) were demonstrated with Nauta and Fink-Heimer methods following hemisections of the third spinal segment in a series of twelve animals. These results were compared with earlier data in the literature obtained from a turtle, a snake, and a lizard using the same experimental and histological procedures. The results show remarkable similarities considering that each species represents a different reptilian order with different evolutionary history and habitat. However, the caiman displays several important peculiarities. Although the d

Role of Thalamic CaV3.1 T-Channels in Fear Conditioning.

The potential contribution of the ion channels that control the excitability of the midline and intralaminar nuclei of the thalamus to the modulation of behaviors has not been well studied. In this study, we used both global genetic deletion (knock-out, KO) and thalamus-specific molecular knock-down (KD) approaches to investigate the role of thalamic CaV3.1 T-type calcium channels (T-channels) in fear learning and fear responses. Previously, we have shown that the dominant subtype of T-channels in the central medial nucleus of the thalamus (CMT) is the CaV3.1 isoform and that CMT neurons from

Medial thalamotomy with the Leksell Gamma Knife for treatment of chronic pain.

The authors describe 10 patients who underwent stereotactic medial thalamotomy with the Leksell Gamma Knife for treatment of chronic intractable pain. The pain was related to structural spinal disorders (4), postherpetic neuralgia (2), spinal cord injury (1), thalamic syndrome (1), anesthesia dolorosa of the face (1), and brainstem infarction (1). All patients had undergone extensive treatment with a variety of modalities prior to gamma thalamotomy. Nine patients underwent unilateral and one patient bilateral lesions. Magnetic resonance imaging (MRI) was used for target localization and the le

Distribution of androgen and estrogen receptor mRNA-containing cells in the rat brain: an in situ hybridization study.

The distribution of cells that express mRNA encoding the androgen (AR) and estrogen (ER) receptors was examined in adult male and female rat … nd in regions with widespread connections throughout the brain, including the lateral hypothalamus, intralaminar thalamic nuclei, and deep layers of the cerebral cortex, suggesting that AR and ER may modulate a wide variety of neural functions. Mild Traumatic Brain Injury Is Associated With Increased Thalamic Subregion Volume in the Subacute Period Following Injury.

Structural vulnerability of the thalamus remains underinvestigated in mild traumatic brain injury (mTBI), and few studies have addressed its constituent nuclei using robust segmentation methods. This study aimed to investigate thalamic subnuclei volume in the subacute period following mTBI. Trauma control (TC) and mTBI patients aged 18-60 years old completed a magnetic resonance imaging (MRI) protocol including both high resolution structural (T1w) and diffusion-weighted sequences at 6-12 weeks following injury (mean: 57 days; SD 11). Each thalamus was segmented into its constituent subnuclei,

Glutamatergic Projection from the Ventrolateral Periaqueductal Gray to the Parafascicular Nucleus of the Thalamus Contributes to the Modulation of Neuropathic Pain in Mice.

Chronic pain is frequently accompanied by increased pain sensitivity and affective disturbances. The ventrolateral periaqueductal gray (vlPAG) and the parafascicular nucleus of the thalamus (PF) are key nodes in pain modulation. However, the functional role of the vlPAG-PF neural pathway remains poorly understood. In this study, we employed an integrative approach combining neuroanatomical tracing and chemogenetic techniques to investigate the role of the vlPAGvGluT2-PF neural pathway in pain modulation. Our results demonstrate that vGluT2-positive neurons in the vlPAG project to the PF. Chemo

Modulation of neuronal activity in human centromedian nucleus during an auditory attention and working memory task.

Centromedian nucleus (CM) is one of several intralaminar nuclei of the thalamus and is thought to be involved in consciousness, arousal, and attention. CM has been suggested to play a key role in the control of attention, by regulating the flow of information to different brain regions such as the ascending reticular system, basal ganglia, and cortex. While the neurophysiology of attention in visual and auditory systems has been studied in animal models, combined single unit and LFP recordings in human have not, to our knowledge, been reported. Here, we recorded neuronal activity in the CM nuc

Characterising the Analgesic Effect of Different Targets for Deep Brain Stimulation in Trigeminal Anaesthesia Dolorosa.

BACKGROUND: Several deep brain stimulation (DBS) targets have been explored for the alleviation of trigeminal anaesthesia dolorosa. We aimed to characterise the analgesia produced from the periaqueductal grey (PAG) and centromedian-parafascicular (CmPf) nucleus using a within-subject design. METHOD: We report a case series of 3 subjects implanted with PAG and CmPf DBS systems for the treatment of anaesthesia dolorosa. At follow-up, testing of onset and offset times, magnitude, and thermal and mechanical sensitivity was performed. RESULTS: The mean pain score of the cohort was acutely reduced

The CM-aGPi Network in the Generation of Tics in Tourette Syndrome.

BACKGROUND: Tourette syndrome (TS) is a neurodevelopmental disorder characterized by intermittent involuntary, repetitive movements and vocalizations collectively referred to as tics. For participants with severe tics who are resistant to medication and behavioral therapy, deep brain stimulation (DBS) has emerged as a potential treatment. Suboptimal animal models and shortcomings in anatomical imaging have collectively hindered progress in understanding the neurophysiological basis of human tic generation. OBJECTIVES: This study investigates the mechanisms underlying tic generation across two

Evidence for Thalamic Responsive Neurostimulation in Treatment of Adult and Pediatric Epilepsy.

Responsive neurostimulation (RNS) has well-established efficacy in patients with identifiable seizure foci. Emerging evidence suggests the feasibility of expanding this treatment to patients with nonfocal or multifocal epileptic profiles with thalamic targeting. Our institution performed two successful implantations of thalamic RNS (tRNS) targeting the centromedian nucleus of the thalamus (CMT), and 1-year postoperative outcomes are provided. Additionally, a literature review of all reported tRNS was conducted. Publications were excluded if they did not include demographic data and/or epilepsy

Retinofugal projections in the mouse.

The laboratory mouse is increasingly a subject for visual system investigation, but there has been no comprehensive evaluation of this species' visual projections. Here, projections were visualized and mapped following intraocular injection of cholera toxin B subunit. Tissue was processed using standard procedures applied to 30 μm free-floating sections with diaminobenzidine as the chromogen. The mouse retina projects to ~46 brain regions, including 14 not previously described in this species. These include two amygdaloid nuclei, the horizontal limb of the diagonal band, the paraventricular hy

VIM deep brain stimulation does not improve pre-existing restless legs syndrome in patients with essential tremor.

The neuroanatomic substrate for restless legs syndrome (RLS) is not known. We implanted deep brain stimulators into the ventralis intermedius nucleus (VIM) of the thalamus in nine subjects for essential tremor (ET) whom all concurrently had RLS. Although the VIM DBS improved tremor, none of the subjects felt there was any meaningful effect on their RLS symptoms. The VIM thalamus, which is involved in ET and Parkinson's disease, does not seem to be primarily involved in RLS.

Effects of chlordiazepoxide and FG 7142 on a rat model of diencephalic amnesia as measured by delayed-matching-to-sample performance.

The intralaminar thalamic nuclei (ILn) have been implicated as a critical site of pathology in amnesia. Lesions of the ILn have been found to produce behavioral effects comparable to benzodiazepine (BDZ) receptor agonists. We compared the effects of chlordiazepoxide (CDP), a BDZ agonist, and FG 7142, a partial inverse agonist at the BDZ receptor, in rats with thalamic lesions and in unlesioned controls. Delayed matching-to sample (DMS) performances were studied during treatment with ascending doses of CDP, counterbalanced trials with 2.5 mg/kg CDP and saline, ascending doses of FG 7142, and (f…

Reticular thalamic responses to nociceptive inputs in anesthetized rats.

The present study compares nociceptive responses of neurons in the reticular thalamic nucleus (RT) to those of the ventroposterior lateral nucleus (VPL). Extracellular single-unit activities of cells in the RT and VPL were recorded in anesthetized rats. Only units with identified tactile receptive fields in the forepaw or hindpaw were studied. In the first series of experiments, RT and VPL responses to pinching with a small artery clamp were tested with the rats under pentobarbital, urethane, ketamine, or halothane anesthesia. Under all types of anesthesia, many RT units were inhibited. Second
