Re-evaluating archival specimens as a pathologic diagnostic resource for neuronal intranuclear inclusion disease.

OBJECTIVES: Neuronal intranuclear inclusion disease (NIID) is a multisystem neurodegenerative disorder caused by GGC repeat expansions in the NOTCH2NLC gene. Although genetic testing has improved diagnostic efficiency, histopathologic confirmation remains essential. We investigated whether routine archival surgical specimens could serve as an alternative pathologic resource for NIID diagnosis. METHODS: We retrospectively analyzed archival formalin-fixed, paraffin-embedded specimens from patients with genetically confirmed NIID, including gastric antrum, colon, gallbladder, prostate, and kidne

Neuronal intranuclear inclusion disease with subclinical peripheral neuropathy: A case report.

RATIONALE: Neuronal intranuclear inclusion disease (NIID) is a slowly progressing neurodegenerative disease with various manifestations and high heterogeneity. Clinical characteristics, imaging, skin biopsy, and genetic testing are necessary for its diagnosis. Electromyography may also be a useful tool for diagnosing NIID. In this study, we report a patient with motor and sensory nerve demyelination changes accompanied by axonal damage. PATIENT CONCERNS: A 64-year-old woman was admitted to our department with gradually worsening forgetfulness for over a year. After 6 years of follow-up, the s

Neurons with Cat's Eyes: A Synthetic Strain of α-Synuclein Fibrils Seeding Neuronal Intranuclear Inclusions.

The distinct neuropathological features of the different α-Synucleinopathies, as well as the diversity of the α-Synuclein (α-Syn) intracellular inclusion bodies observed in post mortem brain sections, are thought to reflect the strain diversity characterizing invasive α-Syn amyloids. However, this "one strain, one disease" view is still hypothetical, and to date, a possible disease-specific contribution of non-amyloid factors has not been ruled out. In Multiple System Atrophy (MSA), the buildup of α-Syn inclusions in oligodendrocytes seems to result from the terminal storage of α-Syn amyloid a

Assessing the NOTCH2NLC GGC repeat expansion in Taiwanese patients with hereditary spastic paraplegia.

We investigated 98 Taiwanese patients with molecularly unassigned hereditary spastic paraplegia (HSP) and found none of them had the NOTCH2NLC GGC repeat expansion, which is the cause of neuronal intranuclear inclusion disease (NIID). Our findings suggest that the NOTCH2NLC GGC repeat expansion may not contribute to HSP.

NOTCH2NLC-related repeat expansion disorders: an expanding group of neurodegenerative disorders.

The NOTCH2NLC gene 5' untranslated region (UTR) GGC repeat expansion mutations were identified as a genetic contributor of neuronal intranuclear inclusion disease (NIID) in 2019. Since then, the number of reported cases with NOTCH2NLC GGC repeat expansion in Asian and European populations has increased rapidly, indicating that the expanded mutation not only leads to the onset or progression of the NIID, but also may play an important role in multiple progressive neurological disorders, including Parkinson's disease, essential tremor, multiple system atrophy, Alzheimer's disease, frontotemporal

Urodynamic Assessment of Neuronal Intranuclear Inclusion Disease.

Neuronal intranuclear inclusion disease (NIID) is a disease that causes leukoencephalopathy (dementia) and peripheral neuropathy (variable manifestation including bladder dysfunction). This is the first urodynamic report to show that bladder dysfunction in NIID is a combination of detrusor overactivity, decreased bladder sensation, large post-void residual, and neurogenic changes in the sphincter electromyogram. This report will help managing bladder dysfunction in NIID.

Pathological background of subcortical hyperintensities on diffusion-weighted images in a case of neuronal intranuclear inclusion disease.

AIMS: Neuronal intranuclear inclusion disease (NIID) is a rare neurodegenerative disorder with eosinophilic intranuclear inclusion bodies. The variable symptoms of NIID increase the difficulty in an antemortem diagnosis. NIID shows leukoencephalopathy on brain magnetic resonance imaging MRI, but the significance of the radiological findings have not been clarified. METHODS: We examined an autopsied case of NIID with subcortical linear hyperintensities on diffusion weighted imaging (DWI) and leukoencephalopathy on fluid attenuation inversion recovery (FLAIR) imaging. Semiquantitative analysis …

Intranuclear inclusions in a case of Creutzfeldt-Jakob disease: an ultrastructural study.

Two types of intranuclear inclusions were described in a brain biopsy from a patient with Creutzfeldt-Jakob disease. The first type of intranculear inclusion was papova virus-like and was observed within 20% of the nuclei of all astrocytes and neurons examined. The particles measured 32 nm in diameter and consisted of a dense core surrounded by a multi-layered shell. The second type of intranuclear inculsion was a granulo-fibrillar nuclear body found within the nuclei of astrocytes and neurons. The possible significance of these inculsions to the etiology of Creutzfeldt-Jakob disease is discus

[Acute herpesvirus-gastritis in a cat].

Gastritis in cats is caused, among other things, by infectious agents, like bacteria, metazoic parasites or viruses. Herpesvirus-gastritis has not as yet been documented in cats. Therefore in this paper such a case will be described. In this case the mucous membrane of the stomach shows multifocal acute necroses with evidence of intranuclear inclusion bodies in epithelial cells of the gastric glands. By means of electron microscopy the causative virus can be specified as herpesvirus.

The abnormal dermis in pyogenic granuloma. Histochemical and ultrastructural observations.

The connective and vascular tissues of pyogenic granulomas have been studied in lesions from forty-six patients. Twenty were studied histochemically, and five were examined ultrastructurally. Both tissues showed a wide range of maturity and cellular activity. The fibroblasts in particular showed evidence of increased synthetic activity. Intranuclear inclusion bodies, suggestive of altered synthetic activity, were found in the endothelial cells of four lesions. It is suggested that pyogenic granuloma arises from disordered growth of papillary dermis.

Hepatitis and hydropericardium syndrome associated with adenovirus infection in goslings.

Two outbreaks of severe acute disease characterised by hepatitis and hydropericardium were observed in young goslings on large-scale farms in Hungary. Histological examination revealed multifocal necrotic areas and two types of intranuclear inclusion bodies adjacent to necrotic areas in the liver. The most prominent type of inclusion bodies showed strong basophilic staining and completely filled the enlarged nucleus. The other type was eosinophilic and occupied the centre of the nucleus, which had margination of chromatin. In the heart, haemorrhage was associated with multifocal necrosis in th…

Parvovirus as a differential diagnosis of hydrops fetalis in the first trimester.

We report a case of fetal parvovirus B19 infection which appears to have resulted in hydrops in the first trimester. An ultrasound scan performed at the booking visit of a woman in the first trimester showed generalised oedema. Karyotyping to exclude a fetal abnormality was normal. Maternal radioimmunoassay serological investigations showed parvovirus B19 IgG and IgM. Post-mortem analysis revealed intranuclear inclusion bodies typical of parvovirus in the erythroid cells in the liver and in the myocardium.

Discovery of myeloid zinc finger (MZF) 1 nuclear bodies.

Myeloid zinc finger 1 (MZF1) is a multifaceted transcription factor that can act either as a transcriptional activator or a gene repressor. We examined its production of nuclear bodies (NBs) and subcellular localization. Proteomic and protein-protein interaction analysis were used to identify its cofactors and interactions. These revealed the presence of MZF1-NBs (intranuclear oligomers containing MZF1). MZF-NBs are similar to some other nuclear bodies, notably promyelocytic leukemia (PML) -NBs in terms of size and morphology. However the two structures appear to be different. MZF-NBs and PML-

Neuronal intranuclear inclusion disease in patients with adult-onset non-vascular leukoencephalopathy.

Neuronal intranuclear inclusion disease (NIID), caused by an expansion of GGC repeats in the 5'-untranslated region of NOTCH2NLC, is an important but underdiagnosed cause of adult-onset leukoencephalopathies. The present study aimed to investigate the prevalence, clinical spectrum and brain MRI characteristics of NIID in adult-onset nonvascular leukoencephalopathies and assess the diagnostic performance of neuroimaging features. One hundred and sixty-one unrelated Taiwanese patients with genetically undetermined nonvascular leukoencephalopathies were screened for the NOTCH2NLC GGC repeat expan

Adenovirus infection of a renal allograft.

One month after renal transplantation, a 60-year-old man developed acute allograft dysfunction associated with gross hematuria and dysuria. Urinary cytological examination showed viral inclusion-bearing epithelial cells. A renal transplant biopsy specimen showed granulomatous interstitial nephritis, tubular necrosis, and ground glass-like intranuclear viral inclusion bodies in tubular cells caused by an adenovirus (ADV) infection. A reduction in baseline immunosuppressive therapy resulted in rapid normalization of allograft function and ultimately viral clearance. We report this case not only

Nuclear inclusions mimicking poly(A)-binding protein nuclear 1 inclusions in a case of inclusion body myopathy associated with Paget disease of bone and frontotemporal dementia with a novel mutation in the valosin-containing protein gene.

A middle-aged Japanese man presented with slowly progressive asymmetric weakness of legs and arm but had neither ptosis nor dysphagia. He had a family history of similar condition suggestive of autosomal dominant inheritance. A muscle biopsy showed mixture of neurogenic atrophy and myopathy with rimmed vacuoles. Furthermore we found intranuclear inclusions that had a fine structure mimicking that of inclusions reported in oculopharyngeal muscular dystrophy (OPMD). Immunohistochemical staining for polyadenylate-binding nuclear protein 1, which is identified within the nuclear inclusions of OPMD

Inclusion body disease (herpesvirus infection) of falcons (IBDF).

Inclusion body disease of falcons (IBDF) is caused by a herpesvirus. The clinical course is short, 24 to 72 hours in duration, and is characterized by mild to severe depression and weakness often accompanied by anorexia. The disease is invariably fatal. The virus has a marked affinity for the reticuloendothelial system and hepatocytes, producing focal to diffuse necrosis of infected tissues accompanied by the formation of intranuclear inclusion bodies. The virus is pathogenic for American kestrels (Falco sparverius) and great horned owls (Bubo virginianus) in which typical lesions of IBDF are r…
