Localized retroperitoneal fibromatosis causing intestinal obstruction in a cynomolgus monkey (Macaca fascicularis).

Gross examination of a 24-month-old, male cynomolgus monkey (Macaca fascicularis) revealed obstruction of the ileum by a mass that entrapped and compressed the ileocecal junction. The mass was well circumscribed, firm, and white on cut surface. Histologically, the mass consisted of spindle-shaped cells arranged in interweaving bundles or as narrow cords and individual cells widely separated by dense collagen. A diagnosis of localized retroperitoneal fibromatosis was made based on the characteristic gross and microscopic findings and isolation of type D simian retrovirus, serotype-2, from splee…

Adverse pregnancy outcome in the monkey (Macaca fascicularis) after chronic caffeine exposure.

Caffeine and the related methylxanthine theophylline are consumed regularly by pregnant women. In a study originally designed to assess the neurotoxic potential of caffeine in the infant, 40 female monkeys (Macaca fascicularis) were divided into three groups and administered caffeine in their drinking water at concentrations equivalent to 0, 10 to 15 or 25 to 35 mg/kg/day of caffeine 7 days a week. After a period of adaptation to caffeine these monkeys were mated with untreated males. Reproductive failure in the form of stillbirths and miscarriages was observed in the treated groups. Subsequen…

Climatic effects on the nasal complex: a CT imaging, comparative anatomical, and morphometric investigation of Macaca mulatta and Macaca fascicularis.

Previous studies exploring the effects of climate on the nasal region have largely focused on external craniofacial linear parameters, using dry crania of modern human populations. This investigation augments traditional craniofacial morphometrics with internal linear and volumetric measures of the anatomic units comprising the nasal complex (i.e., internal nasal cavity depth, maxillary sinus volumes). The study focuses on macaques (i.e., Macaca mulatta and Macaca fascicularis) living at high and low altitudes, rather than on humans, since the short residency of migratory human populations may…

Blood glucose levels in rhesus monkeys (Macaca mulatta) and cynomolgus macaques (Macaca fascicularis) under moderate stress and after recovery.

We evaluated normal blood glucose levels in two species of monkeys, namely rhesus monkeys (Macaca mulatta) and cynomolgus macaques (Macaca fascicularis) kept in the Breeding Center of the Institute of Medical Primatology. The animals were either under moderate stress due to catching or after recovery of defensive and orienting reactions by habituation to the blood sampling. Based on these data, seasonal, gender- and age-dependent variations in glucose levels were found. Glucose levels in rhesus monkeys accustomed to blood sampling procedures were shown to be substantially lower compared with t…

Cholinergic innervation of the primate hippocampal formation. I. Distribution of choline acetyltransferase immunoreactivity in the Macaca fascicularis and Macaca mulatta monkeys.

The cholinergic innervation of the hippocampal formation of Macaca fascicularis (cynomolgus) and Macaca mulatta (rhesus) monkeys was investigated by immunohistochemical procedures using a monoclonal antibody directed against choline acetyltransferase. The distribution of choline acetyltransferase in the monkey demonstrated both similarities and differences with the staining patterns observed in the rat or with acetylcholinesterase in the monkey. While both of these latter preparations demonstrated labeled cells, for example, no choline acetyltransferase labeled neurons were observed in the mon…

Phenotypic profiling of pristane-induced mimicking human systemic lupus erythematosus in Macaca fascicularis.

BACKGROUND: The development of nonhuman primate models that replicate human systemic lupus erythematosus (SLE) remains limited. This study aimed to develop a pristane-induced SLE model in Macaca fascicularis and evaluate its capacity to mimic human-like clinical and laboratory immunological alterations. METHODS: An experimental, single-arm investigation was performed using six female M. fascicularis (2-3 years old, 3-4 kg), which received a single intraperitoneal pristane injection (5 mL/kg body weight) to induce SLE and were monitored biweekly. RESULTS: Throughout the 24-week study period, …

Testing the patterning cascade model of cusp development in Macaca fascicularis mandibular molars.

OBJECTIVE: Molar crown configuration plays an important role in systematics, and functional and comparative morphology. In particular, the n … ng of later-forming cusps. DESIGN: Here we test whether variation in accessory cusp presence in 13 Macaca fascicularis mandibular second molars (M2s) is consistent with predictions of the PCM. Using micro-CT, we imaged these M2s and employed geometric morphometrics to examine whether shape variation in the enamel-dentine junction (EDJ) correlates with accessory cusp …

Optimal allogeneic islet dose for transplantation in insulin-dependent diabetic Macaca fascicularis monkeys.

Many groups are working to improve the results of clinical allogeneic islet transplantation in a primate model. However, few studies have focused on the optimal islet dose for achieving normal glycemia without exogenous insulin after transplantation in primate models or on the relationship between rejection and islet amyloid polypeptide (IAPP) expression. We evaluated the dose (10,000, 20,000, and > 25,000 islet equivalents (IEQ)/kg) needed to achieve normal glycemia without exogenous insulin after transplantation using eleven cynomolgus monkeys, and we analyzed the characteristics exhibited i

Clinical Trypanosoma cruzi Disease after Cardiac Transplantation in a Cynomolgus Macaque (Macaca fascicularis).

A cynomolgus macaque received a heterotopic cardiac allograft as part of a transplant study, with monoclonal antibodies targeted to specific immune costimulation molecules (CD154, CD28) but no traditional immunosuppressive therapy after surgery. Clinical anemia was detected on postoperative day (POD) 35 and had worsened (Hgb, 2.3 g/dL; Hct = 7.3%) by POD 47, despite type-matched whole-blood transfusions. After a total of 4 blood transfusions, hematologic parameters were improved (Hgb, 5.9 g/dL; Hct, 18.7%). On POD 50, a peripheral blood smear revealed trypomastigotes, and qualitative RT-PCR of

Echocardiographic diagnosis of muscular ventricular septal defect in a cynomolgus monkey (Macaca fascicularis).

There have been only a few case reports of heart disease in monkeys. In the case we present, a cardiac murmur was auscultated in a 26-month-old male cynomolgus monkey during a routine physical examination. Echocardiography of this monkey revealed features indicating blood flow from the left ventricle to the right ventricle through the ventricular septum. These findings clarified that the animal had a muscular interventricular septal defect.

Environmental enrichment-related injury in a macaque (Macaca fascicularis): intestinal linear foreign body.

A three-year old male cynomolgus macaque (Macaca fascicularis) presented with clinical signs of anorexia and depression that decreased over a 48-hour period. Results of abdominal radiography abdominocentesis, blood biochemical analysis and CBC suggested septic peritonitis. Exploratory laparotomy revealed multiple perforations along the mesenteric border of the small intestine. Necropsy revealed masses of fibrous material in the stomach and cecum. Multiple mucosal ulcerations, as well as linear fibrous material, were found in the small intestine. The ulceration, perforations, and septic periton…

Analysis of hindbrain neural crest migration in the long-tailed monkey (Macaca fascicularis).

Neural crest cells make a substantial contribution to normal craniofacial development. Despite advances made in identifying migrating neural crest cells in avian embryos and, more recently, rodent embryos, knowledge of crest cell migration in primates has been limited to what was obtained by conventional morphological techniques. In order to determine the degree to which the nonhuman primate fits the mammalian pattern, we studied the features of putative neural crest cell migration in the hindbrain of the long-tailed monkey (Macaca fascicularis) embryo. Cranial crest cells were identified on t…

Synaptic junctions between sympathetic axon terminals and pinealocytes in the monkey Macaca fascicularis.

The distribution of axon terminals in the pineal gland of monkeys was studied by electron microscopy. Numerous terminals bearing small pleomorphic agranular and dense-cored vesicles were localized in the perivascular space and among the pinealocytes in the parenchyma in normal monkeys. Following bilateral superior cervical ganglionectomy, they underwent degenerative changes, including the accumulation of glycogen masses, appearance of dense residual bodies and the displacement of synaptic vesicles. Some of these degenerating terminals showed synaptic contacts with the cell bodies of pinealocyt

Diagrammatic representation for chromosomal mutagenesis studies. II. Radiation-induced rearrangements in Macaca fascicularis.

A qualitative study is presented of chromosomal rearrangements induced in peripheral blood lymphocytes of Macaca fascicularis, after exposure to gamma-irradiation at 2 Gy and 3 Gy. The use of a new diagrammatic representation allowed us to compare, for each type of rearrangement, the distribution of the observed break-points with the theoretical random distribution. It was concluded that chromosomal mutagenesis does not occur at random: an excess of involvement of small chromosomes is found for dicentrics and reciprocal translocations; an excess of telomeric breaks exists in dicentrics and par…

Development of real-time PCR assays for the detection of Moraxella macacae associated with bloody nose syndrome in rhesus (Macaca mulatta) and cynomolgus (Macaca fascicularis) macaques.

BACKGROUND: Moraxella macacae is a recently described bacterial pathogen that causes epistaxis or so-called bloody nose syndrome in captive macaques. The aim of this study was to develop specific molecular diagnostic assays for M. macacae and to determine their performance characteristics. METHODS: We developed six real-time PCR assays on the Roche LightCycler. The accuracy, precision, selectivity, and limit of detection (LOD) were determined for each assay, in addition to further validation by testing nasal swabs from macaques presenting with epistaxis at the Tulane National Primate Research
