Type II integral membrane protein, TM of J paramyxovirus promotes cell-to-cell fusion.

Paramyxoviruses include many important animal and human pathogens. Most paramyxoviruses have two integral membrane proteins: fusion protein (F) and attachment proteins hemagglutinin, hemagglutinin-neuraminidase, or glycoprotein (G), which are critical for viral entry into cells. J paramyxovirus (JPV) encodes four integral membrane proteins: F, G, SH, and transmembrane (TM). The function of TM is not known. In this work, we have generated a viable JPV lacking TM (JPV∆TM). JPV∆TM formed opaque plaques compared with JPV. Quantitative syncytia assays showed that JPV∆TM was defective in promoting c

Structure of a flavivirus envelope glycoprotein in its low-pH-induced membrane fusion conformation.

Enveloped viruses enter cells via a membrane fusion reaction driven by conformational changes of specific viral envelope proteins. We report here the structure of the ectodomain of the tick-borne encephalitis virus envelope glycoprotein, E, a prototypical class II fusion protein, in its trimeric low-pH-induced conformation. We show that, in the conformational transition, the three domains of the neutral-pH form are maintained but their relative orientation is altered. Similar to the postfusion class I proteins, the subunits rearrange such that the fusion peptide loops cluster at one end of an …

Role of metastability and acidic pH in membrane fusion by tick-borne encephalitis virus.

The envelope protein E of the flavivirus tick-borne encephalitis (TBE) virus is, like the alphavirus E1 protein, a class II viral fusion pro … d with influenza virus HA, this transition could not be triggered by input of heat energy alone and membrane fusion could be induced only when the virus was exposed to an acidic pH. In a previous study we showed that the dimer-to-trimer transition appears to be a two-step process involving a reversible dissociation of the dimer followed by an irreversible trimeri …

Acidic pH triggers LCMV membrane fusion activity and conformational change in the glycoprotein spike.

Membrane fusion activity of the glycoprotein (GP) complex of LCMV was determined using the R18 fluorescent dequenching assay. Utilization of an endosomal entry route by LCMV and acidic activation of the membrane fusion activity were indicated by the inhibition of LCMV infection at an early stage by the lysosomal weak base chloroquine and the ionophore monensin. When LCMV was mixed with R18-labeled liposomes with a lipid composition mimicking that of the endosome, dequenching occurred only at acidic pH (< or = 6.0). The measured dequenching was inhibited by protease treatment of the LCMV, indic…

IFITM3 at the viral fusion-pore checkpoint: membrane energetics and entry-route-dependent outcomes.

Interferon-induced transmembrane proteins (IFITMs) are small interferon-stimulated membrane proteins that can limit viral entry, but their effects depend on the virus, entry route, and experimental context. This Mini Review focuses on IFITM3 at the viral fusion-pore checkpoint, defined here as the transition at which hemifusion either proceeds to fusion-pore opening and viral-content release or stalls before productive entry. Work on influenza virus supports a model in which IFITM3 remodels late-endosomal membranes, prolongs or kinetically stabilizes hemifusion-like intermediates, and raises t

Fusion peptide-functionalized amniotic membrane scaffold promotes diabetic burn repair by modulating neurovascular coupling.

Diabetic wounds are a severe complication of diabetes, characterized by impaired neurovascular regeneration and chronic inflammation, which leads to high rates of amputation and a diminished quality of life. To overcome this challenge, we engineered a biofunctional composite scaffold by immobilizing a rationally designed fusion peptide (FP) onto a decellularized amniotic membrane (dAM) matrix. The FP integrates the neuroactive IKVAV sequence and the angiogenic QK motif through a specific collagen-binding domain (CBD), enabling stable and uniform anchoring within the dAM's collagen network. Predictive modeling and fragment-based design of triterpenoid-nucleoside conjugates targeting coronavirus membrane fusion.

Triterpenoids are known for their broad biological activities, yet their potential against SARS-CoV-2 has not been fully elucidated. In this study, using an antiviral dataset generated from our experimental evaluation of triterpenoid-nucleoside derivatives that disrupt SARS-CoV-2 pseudovirus entry by targeting the fusion peptide-heptad repeat 1 (FP-HR1) interface of Spike, we developed a stacking ensemble model with strong predictive performance (accuracy = 74.17 %, AUC = 0.85) to facilitate efficient active-compound identification. Guided by the machine-learning model and a fragment-based ass

A Short Peptide Inhibitor of Measles Virus Fusion Protein that Exhibits Passive Membrane Permeability.

In this study, a passively membrane-permeable short peptide inhibitor targeting the measles virus fusion protein (MeV-F) is reported. Measles virus (MeV) is highly contagious, yet no approved antiviral drugs are currently available. MeV-F plays a crucial role in viral infection, making it an attractive target for drug development. The fusion inhibitor peptide (FIP) is a well-known short peptide that binds to MeV-F and prevents its structural rearrangement. However, improving both inhibitory activity and passive membrane permeability is essential for developing orally available MeV-F inhibitors

Salt Gradient-Induced Phoresis of Vesicles and Enhanced Membrane Fusion in a Crowded Milieu.

Phoresis of biocolloidal objects in response to chemical gradients is a matter of interest among diverse scientific disciplines owing to their importance in the spatiotemporal orchestration of biochemical processes. Although there are reports of soft matter transport/phoresis in the gradient of ions or salts in the aqueous system, their phoretic behavior in the presence of macromolecular crowder is largely unexplored. Notably, cellular cytoplasm is illustrated as a crowded milieu and thereby understanding biomolecular phoresis in the presence of polymeric macromolecules would endorse phoretic

Engineering extracellular vesicles with macrophage membrane fusion for ameliorating imiquimod-induced psoriatic skin inflammation.

INTRODUCTION: Herein, we developed an engineered extracellular vehicle (EV)-based method for ameliorating inflammatory responses in psoriasis. METHODS: EVs, derived from annexin A1 (ANXA1) overexpressing T cells, were co-extruded with M2 macrophage membrane to obtain engineered EVs. In vitro, the effect of engineered EVs on macrophage polarization was evaluated by real-time PCR. In imiquimod (IMQ)-induced psoriasis-like mouse model, the efficacy of engineered EVs in ameliorating psoriatic inflammation was evaluated by Psoriasis Area and Severity Index (PASI) score and immunohistochemistry sta

CADM1 and CADM2 Trigger Neuropathogenic Measles Virus-Mediated Membrane Fusion by Acting in cis.

Measles virus (MeV), an enveloped RNA virus in the family Paramyxoviridae, is still an important cause of childhood morbidity and mortality … ynCAM1) and CADM2 (also known as IGSF4D, Necl-3, SynCAM2) are host factors that enable MeV to cause membrane fusion in cells lacking the known receptors and to spread between neurons. During enveloped virus entry, a cellular receptor generally interacts in trans with the attachment protein on the envelope. However, CADM1 and CADM2 interact in cis with the MeV att …

Two coiled-coil domains of Chlamydia trachomatis IncA affect membrane fusion events during infection.

Chlamydia trachomatis replicates in a parasitophorous membrane-bound compartment called an inclusion. The inclusions corrupt host vesicle trafficking networks to avoid the degradative endolysosomal pathway but promote fusion with each other in order to sustain higher bacterial loads in a process known as homotypic fusion. The Chlamydia protein IncA (Inclusion protein A) appears to play central roles in both these processes as it participates to homotypic fusion and inhibits endocytic SNARE-mediated membrane fusion. How IncA selectively inhibits or activates membrane fusion remains poorly under…

Membrane interactions of the tick-borne encephalitis virus fusion protein E at low pH.

Membrane fusion of the flavivirus tick-borne encephalitis virus is triggered by the mildly acidic pH of the endosome and is mediated by envelope protein E, a class II viral fusion protein. The low-pH trigger induces an oligomeric rearrangement in which the subunits of the native E homodimers dissociate and the monomeric subunits then reassociate into homotrimers. Here we provide evidence that membrane binding is mediated by the intermediate monomeric form of E, generated by low-pH-induced dissociation of the dimer. Liposome coflotation experiments revealed that association with target membrane…

Roles of Cholesterol in Early and Late Steps of the Nipah Virus Membrane Fusion Cascade.

Cholesterol has been implicated in various viral life cycle steps for different enveloped viruses, including viral entry into host cells, cell-cell fusion, and viral budding from infected cells. Enveloped viruses acquire their membranes from their host cells. Although cholesterol has been associated with the binding and entry of various enveloped viruses into cells, cholesterol's exact function in the viral-cell membrane fusion process remains largely elusive, particularly for the paramyxoviruses. Furthermore, paramyxoviral fusion occurs at the host cell membrane and is essential for both viru…

Receptor binding and priming of the spike protein of SARS-CoV-2 for membrane fusion.

Infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is initiated by virus binding to the ACE2 cell-surface receptors1-4, followed by fusion of the virus and cell membranes to release the virus genome into the cell. Both receptor binding and membrane fusion activities are mediated by the virus spike glycoprotein5-7. As with other class-I membrane-fusion proteins, the spike protein is post-translationally cleaved, in this case by furin, into the S1 and S2 components that remain associated after cleavage8-10. Fusion activation after receptor binding is proposed to involve …

Huntingtin is required for ER-to-Golgi transport and for secretory vesicle fusion at the plasma membrane.

Huntingtin is a large membrane-associated scaffolding protein that associates with endocytic and exocytic vesicles and modulates their trafficking along cytoskeletal tracks. Although the progression of Huntington's disease is linked to toxic accumulation of mutant huntingtin protein, loss of wild-type huntingtin function might also contribute to neuronal cell death, but its precise function is not well understood. Therefore, we investigated the molecular role of huntingtin in exocytosis and observed that huntingtin knockdown in HeLa cells causes a delay in endoplasmic reticulum (ER)-to-Golgi t

Structures of the atlastin GTPase provide insight into homotypic fusion of endoplasmic reticulum membranes.

The generation of the tubular network of the endoplasmic reticulum (ER) requires homotypic membrane fusion that is mediated by the dynamin-like, membrane-bound GTPase atlastin (ATL). Here, we have determined crystal structures of the cytosolic segment of human ATL1, which give insight into the mechanism of membrane fusion. The structures reveal a GTPase domain and athree-helix bundle, connected by a linker region. One structure corresponds to a prefusion state, in which ATL molecules in apposing membranes interact through their GTPase domains to form a dimer with the nucleotides bound at the i…

[The process of the pH-dependent fusion of the tick-borne encephalitis virus with artificial membranes].

Fusion of TBE virus with liposomes was distinctly determined at pH 7.0 or lower, the maximum degree of fusion being observed at pH 6.4. Disorders in the native structure of TBE virus envelope protein E prevented virus-membrane fusion. Pre-incubation of viral preparations at pH 6.0 completely inhibited the fusion process, while rupture of disulfide bonds in protein E reduced the degree of fusion approximately 2-fold. Reduction of TBE virus infectivity upon changes in the native conformation of protein E as a consequence of disorders in the process of fusion of virions with cell membranes is dis…

Microscopic insight into HIV fusion peptide-mediated dehydration and packing regulation in membranes.

Human immunodeficiency virus (HIV) infection is believed to occur through the entry of the virion into mammalian cells engineered by the interaction of its fusion peptide (FP), in particular gp41, with the plasma membranes. Despite having a significant understanding of the biochemical pathways of HIV infection, a viable remedy is yet to be achieved. This necessitates the evaluation of peptide-induced microscopic biophysical changes of the host membrane that support viral entry. In this report, we present the first, to our knowledge, detailed microscopic insights into the mechanisms of gp41-med
