Unsaturated fatty acid ketones with αβ,γδ conjugation are susceptible to Michael addition of thiols, with unresolved issues on the site of adduction and precise structures of the conjugates. Herein we reacted 13-keto-octadecadienoic acid (13-oxo-ODE or 13-KODE) with glutathione (GSH), N-acetyl-cysteine, and β-mercaptoethanol and identified the adducts. The serum specimens of patients attending clinics and polyclinics of our hospital and suspected of having brucellosis were evaluated for B. abortus, B. canis and C. burnetti by Wright agglutination, 2ME. TAT (2 mercaptoethanol Tube Agglutination) and KBR (Complement Fixation reaction) tests. A total of 514 sera was examined gram 296 men and 218 women. We detected antibody to C. burnetti 156 (30,39%) of the sera and B. canis in 43 (8.3%) and to B. abortus in 32 (6.2%).

By adsorbing purified Tick-borne encephalitis (TBE) virus antigen onto beads of polystyrene, a solid-phase radioimmunoassay (RIA) for specific antibodies of the IgM- and IgG-class in untreated and nonfractionated human sera could be developed. The antibodies were measured by antihuman-gamma and antihuman-mu globulin labelled with 125 I. The RIA was about 20 times as sensitive as the hemagglutination-inhibition test. About 10% more cases of TBE could be diagnosed with the RIA than with the 2-mercaptoethanol test.

In mixed agglutination tests with patient's sera and sensitized sheep erythrocytes on a glass slide, IgG rheumatoid factor was dected in 15 of 30 rheumatoid arthritis, 3 of 4 progressive systemic sclerosis and 1 of 3 systemic lupus erythematosus sera as well as in 3 of 4 synovial fluids of rheumatoid patients but in none of 10 normal sera. The IgG nature of this factor was confirmed by demonstrating this factor in IgG fractions of positive sera eluted from DEAE-cellulose and its insensitivity to mercaptoethanol.

Thiol reagents dithiothreitol (DTT) and 2-mercaptoethanol (2-ME) are sulfhydryl reagents that can be used to disperse cold autoagglutinins coating red blood cells (RBCs). DTT and 2-ME are primarily used when warm washing of the coated RBCs fails to successfully disperse the cold autoantibody. Using a weak concentration of DTT or 2-ME, the cold IgM agglutinin can be removed from the coated RBCs without disrupting the IgG or complement coating the RBCs. The treated RBCs can be used for ABO typing, antigen typing, or the direct antiglobulin test.

Two hydrogen sulfide photoreleasing molecules (photo-HSRMs), 3-hydroxy-2-naphthalene-methanethiol and the corresponding disulfide, have been synthesized, and their properties and photochemical reactivity have been explored. Both compounds are stable in the neat form and in aqueous solutions in the dark but release H2S under exposure to UVA and UVB light in good quantum and chemical yields. In the presence of thiols, such as cysteine, glutathione, and mercaptoethanol, the yield of hydrogen sulfide release reaches 90%. The mechanism of H2S photorelease from these photo-HSRMs is discussed.

Snake venom cardiotoxin showed the ability to induce polymerization of G-actin from rabbit skeletal muscle in a low ionic strength buffer composed of 0.2 mM CaCl2/0.2 mM ATP/0.5 mM mercaptoethanol/2.0 mM Tris-HCl, pH 8.0. The activity was enhanced greatly when 0.4 mM MgCl2 was present in the buffer and could be inhibited if G-actin was preincubated with deoxyribonuclease I. Furthermore, the DNAase could also partially depolymerize actin polymer previously formed by the interaction of G-actin with the toxin.
