Adsorption and intracellular uptake of mercuric mercury and methylmercury by methanotrophs and methylating bacteria.

The cell surface adsorption and intracellular uptake of mercuric mercury Hg(II) and methylmercury (MeHg) are important in determining the fate and transformation of Hg in the environment. However, current information is limited about their interactions with two important groups of microorganisms, i.e., methanotrophs and Hg(II)-methylating bacteria, in aquatic systems. This study investigated the adsorption and uptake dynamics of Hg(II) and MeHg by three strains of methanotrophs, Methylomonas sp. strain EFPC3, Methylosinus trichosporium OB3b, and Methylococcus capsulatus Bath, and two Hg(II)-me

Diverse Methylmercury (MeHg) Producers and Degraders Inhabit Acid Mine Drainage Sediments, but Few Taxa Correlate with MeHg Accumulation.

Methylmercury (MeHg) is a notorious neurotoxin, and its production and degradation in the environment are mainly driven by microorganisms. A variety of microbial MeHg producers carrying the gene pair hgcAB and degraders carrying the merB gene have been separately reported in recent studies. However, surprisingly little attention has been paid to the simultaneous investigation of the diversities of microbial MeHg producers and degraders in a given habitat, and no studies have been performed to explore to what extent these two contrasting microbial groups correlate with MeHg accumulation in the

Microbial mercury transformations: Molecules, functions and organisms.

Mercury (Hg) methylation, methylmercury (MeHg) demethylation, and inorganic redox transformations of Hg are microbe-mediating processes that determine the fate and cycling of Hg and MeHg in many environments, and by doing so influence the health of humans and wild life. The discovery of the Hg methylation genes, hgcAB, in the last decade together with advances in high throughput and genome sequencing methods, have resulted in an expanded appreciation of the diversity of Hg methylating microbes. This review aims to describe experimentally confirmed and recently discovered hgcAB gene-carrying Hg

Mercury transformation processes in nature: Critical knowledge gaps and perspectives for moving forward.

The transformation of mercury (Hg) in the environment plays a vital role in the cycling of Hg and its risk to the ecosystem and human health. Of particular importance are Hg oxidation/reduction and methylation/demethylation processes driven or mediated by the dynamics of light, microorganisms, and organic carbon, among others. Advances in understanding those Hg transformation processes determine our capacity of projecting and mitigating Hg risk. Here, we provide a critical analysis of major knowledge gaps in our understanding of Hg transformation in nature, with perspectives on approaches movi

Understanding the risks of mercury sulfide nanoparticles in the environment: Formation, presence, and environmental behaviors.

Mercury (Hg) could be microbially methylated to the bioaccumulative neurotoxin methylmercury (MeHg), raising health concerns. Understanding the methylation of various Hg species is thus critical in predicting the MeHg risk. Among the known Hg species, mercury sulfide (HgS) is the largest Hg reservoir in the lithosphere and has long been considered to be highly inert. However, with advances in the analytical methods of nanoparticles, HgS nanoparticles (HgS NPs) have recently been detected in various environmental matrices or organisms. Furthermore, pioneering laboratory studies have reported th

Development of Human Hair Reference Material Supporting the Biomonitoring of Methylmercury.

A certified reference material, NIMD-01, was developed for the analysis of mercury speciation in human hair. We collected the hair of Vietnamese males from a barbershop in Hanoi in 2016 and prepared 1200 bottles containing 3 g of sieved and blended hair powder. The certified value was given on a dry-mass basis, with the moisture content obtained by drying at 85°C for 4 h. Certified values with the expanded uncertainties (coverage factor, k = 2) were as follows: methylmercury, 0.634 ± 0.071 mg kg-1 as mercury; total mercury, 0.794 ± 0.050 mg kg-1; copper, 12.8 ± 1.4 mg kg-1; zinc, 234 ± 29 mg k

Development of an easy portable procedure for on-site determination of mercury and methylmercury.

A portable measurement and speciation procedure for inorganic mercury (HgIN) and methylmercury (CH3Hg) was developed. A portable sample pretreatment was optimized to determine total mercury content. A new home-made sorbent (CYXAD, CHYPOS 101 modified Amberlite XAD), was prepared to separate HgIN and CH3Hg. Mercury species were determined using square wave anodic stripping voltammetry (SW-ASV) with a solid gold electrode (SGE) and using a portable potentiostat. A certified reference material, five freeze-dried samples and three fresh samples were analysed with conventional voltammetric analyzer

Mercury accumulation in Yellowfin tuna (Thunnus albacares) with regards to muscle type, muscle position and fish size.

The concentrations and relationships between individual mercury species and total mercury were investigated in different muscle parts and sizes of Yellowfin tuna (Thunnus albacares). Fourteen Yellowfin tuna caught in the South Atlantic off the coast of South Africa had an average total Hg (tHg) concentration of 0.77 mg/kg wet weight. No differences were detected (p > 0.05) in tHg, MethylHg (MeHg) or inorganic Hg (iHg) accumulation among the four white muscle portions across the carcass, but both tHg and iHg were found in higher concentrations (p < 0.001) in dark muscle than white muscle. Modification of mercury toxicity by selenium: practical importance?

The interaction between mercury and selenium may involve a variety of toxicologically and biochemically distinct processes. In this paper, the interaction between inorganic mercury and sodium selenite, the interaction most extensively studied, as well as the interaction between methylmercury (MeHg) and selenium, the interaction perhaps most significant for non-occupational human populations, will be discussed. It has been shown that the former interaction can be understood as a modification of the kinetic behavior of inorganic mercury by selenite, but this interaction may occur only under very

Accumulation of methylmercury and inorganic mercury in the brain.

Differences in metabolism between different mercury species are well recognized. Conclusions that only a minor demethylation of methylmercury takes place in the brain are based primarily on results from short term studies. Results from a number of studies on humans exposed for many years to methylmercury have shown high concentrations of inorganic mercury in the brain in relation to total mercury. Similar evidence is available from studies on monkeys exposed for several years to methylmercury. The results indicate that a significant accumulation of inorganic mercury takes place with time despi

Evaluation of Methylmercury-Induced Motor Dysfunction and Proteomic Analysis of Cerebrospinal Fluid in Common Marmosets (Callithrix jacchus).

Methylmercury (MeHg) poisoning can lead to Minamata disease, often manifesting as motor dysfunction. We aimed to establish a quantitative evaluation method for MeHg exposure-associated motor dysfunction in the common marmoset (Callithrix jacchus). Three male marmosets were orally administered MeHg (1.5 mg Hg/kg/day) for 2 weeks, followed by a 2-week non-treatment period; three control marmosets received vehicle. Walking and spontaneous behavior were recorded weekly. After marking body segments, bounding gait videos were analyzed to assess locomotion speed, hip joint position, knee angle, dorsa

Crystalline phase regulates transformation and methylation of mercury sulfide nanoparticles in paddy systems.

Mercury sulfide nanoparticles (HgSNPs) represent an important source of bioavailable mercury (Hg) for microbial methylation in paddy systems, depending on their size and crystalline phases. However, little is known about the phase compositions of HgSNPs in Hg-contaminated paddy fields with dynamically changed redox conditions, their transformation, and methylation potential. Applying transmission electron microscopy (TEM) and synchrotron radiation X-ray absorption spectroscopy (SR-XAS), we found β-HgSNPs as the predominant Hg species in newly contaminated areas, whereas α-HgSNPs dominated in p

Disclosing the Preferential Mercury Chelation by SeCys Containing Peptides over Their Cys Analogues.

Methylmercury, mercury (II), and mercury (I) chlorides were found to react with vasopressin, a nonapeptide hormone cyclized by two cysteine residues, and its mono- and diselenium analogues to form several mercury-peptide adducts. The replacement of Cys by SeCys in vasopressin increased the reactivity toward methylmercury, with the predominant formation of -Se/S-Hg-Se-bridged structures and the consequent demethylation of methylmercury. In competitive experiments, CH3HgCl reacted preferentially with the diselenium analogue rather than with vasopressin. The diselenium peptide also showed the cap

Effects of methylmercury and food stress on migratory activity in song sparrows, Melospiza melodia.

Avian migration is a challenging life stage susceptible to the adverse effects of stressors, including contaminants like methylmercury (MeHg). Although birds often experience stressors and contaminants concurrently in the wild, no study to date has investigated how simultaneous exposure to MeHg and food stress affects migratory behavior. Our objectives were to determine if MeHg or food stress exposure during summer, alone or combined, has carry-over effects on autumn migratory activity, and if hormone levels (corticosterone, thyroxine) and body condition were related to these effects. We teste

Cellular Mercury Coordination Environment, and Not Cell Surface Ligands, Influence Bacterial Methylmercury Production.

The conversion of inorganic mercury (Hg(II)) to methylmercury (MeHg) is central to the understanding of Hg toxicity in the environment. Hg methylation occurs in the cytosol of certain obligate anaerobic bacteria and archaea possessing the hgcAB gene cluster. However, the processes involved in Hg(II) biouptake and methylation are not well understood. Here, we examined the role of cell surface thiols, cellular ligands with the highest affinity for Hg(II) that are located at the interface between the outer membrane and external medium, on the sorption and methylation of Hg(II) by Geobacter sulfur

Mercury dynamics in the pore water of peat columns during experimental freezing and thawing.

Biogeochemical processes in northern peatland ecosystems are influenced by seasonal temperature fluctuations that are changing with the climate. Methylmercury (MeHg), commonly produced in peatlands, affects downstream waters; therefore, it is important to understand how temperature transitions affect mercury (Hg) dynamics. We investigated how the freeze-thaw cycle influences belowground peat pore water total Hg (THg), MeHg, and dissolved organic carbon (DOC). Four large, intact peat columns were removed from an ombrotrophic peat bog and experimentally frozen and thawed. Pore water was sampled

Methyl mercury exposure in Niigata: case studies illustrating adult outcomes.

Methyl mercury (MeHg) is a potent neurotoxin. Methyl mercury exposure took place at Niigata in the late 1950s and early 1960s due to industrial pollution of the Agano River. We studied adults who were exposed in utero or as children during that epidemic to determine their long-term outcomes. We contacted individuals with known exposure. Those who consented completed a questionnaire and then were interviewed. Thirty-nine subjects agreed to participate. Twenty-six subjects had health problems during their childhood and difficulties with schoolwork. As adults, some had difficulty with employment,

Fate of mercury and methylmercury in full-scale sludge anaerobic digestion combined with thermal hydrolysis.

Methylmercury (MeHg) is one of the highly toxic and bio-accumulated forms of mercury. Its presence in wastewater treatment processes has been evidenced in recent studies. Considering its enrichment in sewage sludge and the ecological risk associated with its land application, this study investigated the fate of mercury and MeHg in full-scale anaerobic digestion combined with Cambi thermal hydrolysis based on one-year sampling. Results showed that the advanced anaerobic digestion could increase the total mercury (THg) content from 4.35 ± 0.43 mg/kg in raw sludge to 6.37 ± 1.05 mg/kg in digested

Biogeochemical changes and mercury methylation beneath an in-situ sediment cap.

In-situ capping has shown promise as a management strategy for contaminated aquatic sediments, however, little is known about how mercury methylation in underlying sediments will be affected. Changes to the location and extent of sulfate reduction and other biological processes were studied in estuarine sediment using laboratory microcosms. Observations in a model sediment showed increases of in situ total methylmercury concomitant with an upward extension of anaerobic bacterial activity beneath a sediment cap and under anoxic conditions. Increased methylmercury (up to 50%) was observed beneat
