Characterization of hepatitis C virus pseudoparticles by cryo-transmission electron microscopy using functionalized magnetic nanobeads.

Cell entry and membrane fusion of the hepatitis C virus (HCV) depend on its envelope glycoproteins E1 and E2. HCV pseudotyped particles (HCVpps) are relevant and popular models to study the early steps of the HCV life cycle. However, no structural characterization of HCVpp has been available so far. Using cryo-transmission electron microscopy (cryo-TEM), providing structural information at nanometric resolution, the molecular details of HCVpps and their fusion with liposomes were studied. Cryo-TEM revealed HCVpps as regular 100 nm spherical structures containing the dense retroviral nucleocaps

Partial liquid ventilation with perfluorocarbon in acute lung injury: light and transmission electron microscopy studies.

Liquid ventilation using perfluorocarbon has been shown to improve gas exchange in animal models of acute lung injury as well as in children with acute respiratory distress syndrome. This study was designed to define structural features of lung injury following partial liquid ventilation (PLV) using light and transmission electron microscopy in a rabbit model of acute respiratory distress. Animals were treated with either conventional mechanical ventilation (CMV-gas) (n = 6) or PLV (n = 5) for 4 h after the induction of acute lung injury with saline lavage. Control animals were killed after th

Transmission electron microscopy of chromosomes by longitudinal section preparation: application to fragile X chromosome analysis.

We developed a method for the preparation of ultrathin longitudinal sections of chromosomes enabling TEM studies of whole chromosomes. By using a novel "repeat chill" method of exposing the glass slide and plastic block interface to liquid nitrogen, it was possible to separate consistently hardened epoxy resin-embedded chromosome spreads from glass slides for ultrathin longitudinal sectioning of entire spreads and of individual chromosomes. The method was applied to analyze the fragile X chromosome. The ultrastructure of centromeres, telomeres, fragile sites and other chromosomal areas can now

Transmission electron microscopy of early microbial colonization of human enamel and root surfaces in vivo.

This study describes the early microbial colonization of teeth by the use of light- and transmission electron microscopy. Six dental students carried a total of 60 test pieces of unerupted enamel and root surface in intraoral acrylic appliances for 4, 8, 12, 24 and 48 h, during which periods oral hygiene was abandoned. Pronounced variations were recorded in structure and thickness of the pellicle across the individual surfaces of both dental tissues. Bacterial single-cell colonization increased the electron density of the adjoining pellicle. Micro-colonies of bacteria were observed in relation

WAFMGAN: A frequency-aware CycleGAN for structure-consistent virtual staining of transmission electron microscopy images.

Conventional transmission electron microscopy (TEM) staining improves image contrast but requires additional sample preparation, consumes reagents and tissue, and may introduce deformation, local mismatch, or tissue loss. When strictly paired stained and unstained sections are difficult to obtain, unpaired virtual staining provides a practical route for improving the readability of unstained TEM images while retaining correspondence with source-image structural cues. This paper presents WAFMGAN, a frequency-aware CycleGAN framework that combines Wavelet Asymmetric Frequency Mamba (WAFM) blocks

Automated detection of overlapping and well-separated nanoparticles in transmission electron microscopy via deep learning.

Accurate detection of nanoparticles in transmission electron microscopy (TEM) images is essential for high-throughput characterization in chemistry and materials science. TEM micrographs of advanced photocatalysts often contain densely packed and overlapping nanoparticles, creating electron-dense regions where individual particle boundaries cannot be resolved. This challenges accurate particle counting, sizing, and morphological analysis, and motivates the development of reliable automated analysis workflows. Here, we present a deep learning workflow for robust nanoparticle detection that comb

Dengue virus identification by transmission electron microscopy and molecular methods in fatal dengue hemorrhagic fever.

Dengue virus is the most significant virus transmitted by arthropods worldwide and may cause a potentially fatal systemic disease named dengue hemorrhagic fever. In this work, dengue virus serotype 4 was detected in the tissues of one fatal dengue hemorrhagic fever case using electron immunomicroscopy and molecular methods. This is the first report of dengue virus polypeptides findings by electron immunomicroscopy in human samples. In addition, not-previously-documented virus-like particles visualized in spleen, hepatic, brain, and pulmonary tissues from a dengue case are discussed.

Human islet amyloid polypeptide fibril binding to catalase: a transmission electron microscopy and microplate study.

The diabetes-associated human islet amyloid polypeptide (IAPP) is a 37-amino-acid peptide that forms fibrils in vitro and in vivo. Human IAPP fibrils are toxic in a similar manner to Alzheimer's amyloid-beta (A-beta) and prion protein (PrP) fibrils. Previous studies have shown that catalase binds to A-beta fibrils and appears to recognize a region containing the Gly-Ala-Ile-Ile sequence that is similar to the Gly-Ala-Ile-Leu sequence found in human IAPP residues 24-27. This study presents a transmission electron microscopy (TEM)-based analysis of fibril formation and the binding of human eryth

Scanning and transmission electron microscopy of mercuric chloride-induced acute tubular necrosis in rat kidney.

Scanning (SEM) and transmission (TEM) electron microscopy were used to elucidate morphological changes associated with acute tubular necrosis induced by high doses of mercuric chloride. Marked morphological changes were demonstrated in proximal tubules with TEM at one hour and with both SEM and TEM at six hours. These changes appeared earlier than reported in previous studies using any dose. The scanning microscopic provided a three-dimensional view of proximal cells showing changes in early injury with subsequent separation of the injured cells from the remaining cells. Certain of these resid

Effects of Microtubule and Actin Inhibitors on Cryptococcus neoformans Examined by Scanning and Transmission Electron Microscopy.

BACKGROUND: Cryptococcus neoformans is one of the most important human fungal pathogens. Its cells contain rich microtubules required for nuclear division and rich F-actin cytoskeletons for cell division. Disruption of microtubules by a microtubule inhibitor should block nuclear division, and disruption of F-actin by an actin inhibitor should block cell division. We investigated the effects of microtubule and actin inhibitors to find out whether the cytoskeletons of C. neoformans can become a new anti-fungal target for the inhibition of cell division, when examined at the ultrastructural level

The ultrastructure of the lung after exposure to ionizing radiation as seen by transmission and scanning electron microscopy.

Male mice of the BALB/c strain were exposed at an age of 12-14 weeks, to different doses of X-rays, either to the entire thorax or to the right hemithorax. At various times after exposure mice were sacrificed and the lung was examined by transmission or scanning electron microscopy. Radiation symptoms following exposure to 15-20 Gy can be divided into three phases: Early, from a few hours to a few weeks after exposure, changes in capillary permeability and morphological alterations in all types of lung cells are prominent. At intermediate times, from a few weeks to about 7 months after exposur

The combination of scanning and transmission electron microscopy techniques in pathology of the organ of Corti in guinea pigs.

The pathology of the organ of Corti in guinea pigs is studied by means of SEM and TEM. It was the purpose of this study to examine the surface of an experimentally altered organ of Corti by means of SEM screening method). As it was our main interest to find out the localization of acoustic stimulation regarding the tonotopicity on the surface of the organ of Corti, we always examined the whole cochlear by means of SEM first. In acoustic experiments we used long-time pure-tone stimulation beyond the directly damaging sound pressure level, as well as white noise or shots for experimental damage

Scanning and transmission electron microscopy of the rugose portion of the human endolymphatic sac--a preliminary report.

The rugose portion of two human endolymphatic sacs--one from a Meniere's disease patient, the other from a patient who underwent removal of an eighth nerve tumor and had no other evidence of inner ear disease--has been examined by scanning and transmission electron microscopy. The only morphological finding that distinguished the normal from the Meniere's disease specimen was more folds in the epithelium of the normal sac. Two types of cells have been distinguished in the two sacs through both scanning and transmission electron microscopy: cuboidal-to-columnar cells with a convex luminal surfa

Operando identification of anion effect on lithium nucleation and growth via in situ transmission electron microscopy.

Lithium metal batteries are considered promising candidates for next-generation energy storage due to the high capacity and low redox potential of lithium negative electrodes. However, dendritic Li growth and unstable solid-electrolyte interphase formation remain critical bottlenecks for practical implementation. While electrolyte anion chemistry critically governs solid-electrolyte interphase formation, nanoscale observations of anion-regulated Li nucleation and growth mechanisms remain limited in tracking dynamic interfacial processes. Here, we employ in situ liquid-phase transmission electr

Artificial Intelligence-Assisted Workflow for Transmission Electron Microscopy: From Data Analysis Automation to Materials Knowledge Unveiling.

(Scanning) transmission electron microscopy ((S)TEM) has significantly advanced materials science but faces challenges in correlating precise atomic structure information with the functional properties of devices due to its time-intensive nature. To address this, an analytical workflow is introduced for the holistic characterization, modelling, and simulation of device heterostructures. This workflow automates the experimental (S)TEM data analysis, providing an in-depth characterization of crystallographic information, 3D orientation, elemental composition, and strain distribution. It reduces
