Amaryllidaceae Alkaloids and Isoquinoline Alkaloids: A Perspective on Historical Approaches to Pathway Elucidation.

Alkaloid biosynthesis is a central topic in plant specialized metabolism because many alkaloids have ecological, pharmacological, and biotechnological relevance. Isoquinoline alkaloids (IAs) and Amaryllidaceae alkaloids (AAs) are both connected to aromatic amino acid metabolism, but they differ in taxonomic distribution, scaffold-forming chemistry, pathway resolution, and biotechnological development. This review compares the historical and methodological trajectories that have shaped IA and AA pathway elucidation, from compound isolation, radiotracer experiments, and biochemical inference to

Effects of the cannabinoid CB1-receptor neutral antagonist AM4113 and antagonist/inverse agonist rimonabant on fentanyl discrimination in male rats.

Evidence suggests the existence of a functional interaction between endogenous cannabinoid (CB) and opioid systems. Thus, targeting CB1 receptors might be a viable approach to develop new medications for opioid use disorders (OUD). The present studies were undertaken to evaluate the effects of the neutral CB1 antagonist AM4113 and the CB1 antagonist/inverse agonist rimonabant in male rats trained to discriminate 0.032 mg/kg fentanyl from saline under a 10-response fixed-ratio (FR-10) schedule of food reinforcement. Results show that the µ-opioid agonists (fentanyl, oxycodone, and morphine) sub

Functional Characterization of 4'OMT and 7OMT Genes in BIA Biosynthesis.

Alkaloids are diverse group of secondary metabolites generally found in plants. Opium poppy (Papaver somniferum L.), the only commercial source of morphinan alkaloids, has been used as a medicinal plant since ancient times. It produces benzylisoquinoline alkaloids (BIA) including the narcotic analgesic morphine, the muscle relaxant papaverine, and the anti-cancer agent noscapine. Though BIAs play crucial roles in many biological mechanisms their steps in biosynthesis and the responsible genes remain to be revealed. In this study, expressions of 3-hydroxy-N-methylcoclaurine 4'-methyltransferase

Epidemiologic and molecular pathophysiology of chronic opioid dependence and the place of naltrexone extended-release formulations in its clinical management.

Naltrexone implants and depot injections (NI) are a novel form of treatment for opiate dependence (OD). Major questions relate to their absolute and relative efficacy and safety. Opportunely, six recent clinical trial data from several continents have uniformly provided dramatic evidence of the potent, dose-related and highly significant efficacy of NI, with minimal or manageable accompanying toxicity and safety concerns. The opiate-free lifestyle is attained significantly more often with NI adjusted O.R. = 6.00 (95% C.I. 3.86-9.50), P < 10(-10). Other drug use and drug craving are also rapidl

Sinomenine hydrochloride injection for knee osteoarthritis: A protocol for systematic review and meta-analysis.

BACKGROUND: Knee osteoarthritis (KOA) is a degenerative disease in the knee joint, with chronic joint pain, swelling, stiffness, and dysfunction as the primary manifestations. Sinomenine hydrochloride injection is a proprietary Chinese medicine injection of sinomenine, the main active component of traditional Chinese medicine (TCM). Clinical studies show that Sinomenine hydrochloride injection has a good effect on the treatment of KOA. At present, there is still a lack of systematic reviews and meta-analyses to evaluate the efficacy and safety of sinomenine hydrochloride injection in the treat

The nonnarcotic antitussive drug dimemorfan: a review.

The antitussive dimemorfan phosphate was discovered through extensive screening of morphinic derivatives and was introduced in Japan in 1975. The majority of studies on dimemorfan have been published in Japanese, and this review aims to make these data more generally available. The antitussive action of dimemorfan appears to be directly on the cough center in the medulla. Dimemorfan does not induce any significant physical or psychologic dependence, and its antitussive action is not affected by the opioid-receptor blocker levallorphan. Dimemorfan is therefore considered a nonnarcotic antitussi

Influence of age on the pharmacokinetics of butorphanol.

Butorphanol kinetics were studied in 8 young (23-34 years of age) and 9 elderly (65-79 years of age) healthy male subjects following a single, intravenous 2-mg dose of butorphanol tartrate. Plasma and blood concentrations collected over 24 h were analyzed by a second antibody radioimmunoassay for unchanged drug. Systemic blood clearance (21.29 +/- 7.18 ml/min/kg), apparent steady-state volume of distribution (9.02 +/- 2.40 liters/kg), distribution half-life (7.67 +/- 4.66 min), and the blood/plasma concentration ratio (1.21 +/- 0.36) in the elderly did not differ from those in the younger subj

Cardiopulmonary effects of butorphanol tartrate in horses.

The cardiopulmonary and behavioral effects of butorphanol were evaluated in pain-free adult horses. Butorphanol tartrate was administered IV in doses of 0.1, 0.2, and 0.4 mg/kg to the same horses on 3 separate occasions. There were no significant (P less than 0.05) changes in heart rate, mean and diastolic arterial pressure, mean and diastolic pulmonary arterial blood pressure, or cardiac output recorded in the horses given these doses. Systolic arterial blood pressure was significantly (P less than 0.05) increased in only the horses given the 0.2 mg/kg dose. Significant (P greater than 0.05)

Priapism after sedation, neuroleptanalgesia and anaesthesia in the horse.

The paper reports 6 cases of priapism with subsequent protrusion and paresis after neuroleptanalgesia and anaesthesia in horses. Five were ultimately treated by amputation of the penis but the sixth horse is responding satisfactorily to conservative treatment at the time of writing. Causative factors, preventive measures and possible methods of treatment are discussed.

New Sinomenine Derivative as a Potential Drug for Cervical Cancer via Inducing Ferroptosis.

Cervical cancer remains a serious threat to women's health due to chemotherapy resistance, poor prognosis, and survival rates. Novel anti-cervical cancer agents are urgently needed. Sinomenine derivatives, which were prepared from sinomenine (a natural morphinane alkaloid), have been previously reported by us to be cytotoxic toward various cancer cell lines. Also, an increase in ROS production, which is associated with ferroptosis, was observed in cancer cells. In this study, we synthesized 18 sinomenine derivatives and evaluated their antitumor activity in vitro against four human tumor cell

Synovial Targeting and Redox-Triggered Release: A Dual Strategy in Peptide-Drug Conjugates for Rheumatoid Arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by synovial inflammation and joint destruction. To overcome the rapid clearance and systemic side effects of sinomenine (SIN), three critical components of peptide-drug conjugates (PDCs), including the targeting peptide, linker, and payload, were optimized, and two synovial homing peptide-sinomenine conjugates, SIN-SS-DRL and dSIN-SS-DRL, linked via reduction-sensitive disulfide bonds, were developed. Both new conjugates exhibited markedly improved plasma stability and demonstrated selective, glutathione-triggered release

A Novel Compound 3a-M1, from Metabolites of Sinomenine Derivative 3a, Exerts Potent Anti-Aplastic Anemia Activity via IP3R/ORAI-Mediated CTL Ferroptosis.

Aplastic anemia (AA) is a refractory hematological disease with limited therapeutic effectiveness and serious treatment-related side effects. Cytotoxic T lymphocytes (CTLs) play a key role in AA pathogenesis. In our previous study, sinomenine derivative 3a was obtained, which demonstrated potential anti-AA activity by targeting CTLs with low toxicity. In this study, a novel metabolite, 3a-M1, was identified with optimized bioavailability from 3a metabolism, which exhibited a more notable effect in alleviating anemia symptoms, suppressing bone marrow CTLs activation, and improving hematopoietic

[Clinical Research Progress on Using κ-Opioid Receptor Agonists to Treat Uremic Pruritus].

Uremic pruritus, a severe complication in patients with chronic kidney disease, is associated with a high prevalence. It can cause depression and sleep disorders, and seriously affect the quality of life and the social relations of patients. Recently, there is growing evidence showing that κ-opioid receptor agonists, including nalfurafine, difelikefalin, and nalbuphine, can effectively and safely reduce itching symptoms in patients with refractory uremic pruritus. Herein, we reviewed the epidemiology, pathogenesis, clinical symptoms, and treatment strategies of uremic pruritus, and summarized

Immunosuppressive and anti-inflammatory activities of sinomenine.

Sinomenine (SN), a pure compound extracted from the Sinomenium acutum plant, has been found to inhibit T- and B-lymphocyte activation, proliferation and function and to interfere with the differentiation, recruitment and function of several other cell types, such as dendritic cells (DC). SN has demonstrated its potential anti-inflammatory role for treating immune-related disorders in experimental animal models and in some clinical applications. This review will summarize its potential effects, mechanisms and applications.

Role for μ-opioid receptor in antidepressant effects of δ-opioid receptor agonist KNT-127.

Previous pharmacological data have shown the possible existence of functional interactions between μ- (MOP), κ- (KOP), and δ-opioid receptors (DOP) in pain and mood disorders. We previously reported that MOP knockout (KO) mice exhibit a lower stress response compared with wildtype (WT) mice. Moreover, DOP agonists have been shown to exert antidepressant-like effects in numerous animal models. In the present study, the tail suspension test (TST) and forced swim test (FST) were used to examine the roles of MOP and DOP in behavioral despair. MOP-KO mice and WT mice were treated with KNT-127 (10 m
