Subcutaneous mucormycosis caused by Mucor hiemalis Wehmer f. luteus (Linnemann) Schipper 1973.

The authors report a case of subcutaneous mucormycosis in a diabetic patient with verrucous lesions in the fourth finger of the right hand. Initially diagnosed as a fixed cutaneous sporotrichosis case, success was obtained with the use of potassium iodide with healing of the lesion in a period of nearly 5 months. At present the patient remains under clinical observation with no treatment or relapse.

Backbone resonance assignment of mucoricin: a step towards structural insights into mucor pathogenesis.

Mucoricin is a key virulence factor in mucormycosis, as it targets host endothelial cells and inhibits protein translation through its N-glycosylase activity, leading to vascular leak and increased vascular permeability, thereby contributing to further disease progression. Its structure is quite unique, being structurally homologous to the Ricin B chain as confirmed by cross-reactivity with monoclonal anti-Ricin B chain antibodies, while exhibiting enzymatic activity similar to the Ricin A chain. Here, we report near-complete backbone (Soliman in Nat Microbiol 6(3):313–326, 2021) 1H, 15N, and …

Necrotizing fasciitis caused by Mucor indicus in a pediatric bone marrow transplant recipient.

Necrotizing fasciitis is a life-threatening, rapidly progressing infection of fascia and subcutaneous cellular tissue typically caused by mixed aerobic and anaerobic bacteria. We present a case report of an immunocompromised 4-year-old female with necrotizing fasciitis from a rare fungal organism, Mucor indicus. The patient underwent multiple debridements and was treated for 10 months, first on liposomal amphotericin B (2 months) then posaconazole (8 months). Mucor indicus is a rarely described pathogen with only nine other cases described. Identification of this organism remains a challenge, …

Localized oxalosis associated with simultaneous Aspergillus and Mucor infection in diabetic foot gangrene.

A case of simultaneous aspergillosis and mucormycosis complicating diabetic foot gangrene is reported. The recognition of this combined opportunistic fungal infection was brought about by an initial finding of birefringent calcium oxalate crystals associated with a background of acute inflammatory exudate, which is considered a strong clue to the diagnosis of infection caused by Aspergillus niger.

Biosynthesis of Active Ginsenoside Aglycone Analogues Using Two Oxidases Mined from Mucor spinosus.

The pharmacological activity of protopanaxadiol (PPD) can be enhanced by oxidative modification. Compared with chemical synthesis and biotransformation, synthetic biology offers a more efficient and eco-friendly approach for modifying PPD. In this study, we identified two oxidases from Mucor spinosus. MsSDR3 oxidizes C3-OH of both dammarenediol-II and PPD to a ketone, while MsCYP3 oxidizes C12-OH of PPD to a ketone and hydroxylates PPD at C7β, C15α, and C11β positions. Combining MsSDR3/MsCYP3 with the enzymes involved in ginsenoside biosynthesis, we achieved de novo biosynthesis of seven ginse…

First report of Mucor circinelloides occurring on yellow catfish (Pelteobagrus fulvidraco) from China.

Infected yellow catfish (Pelteobagrus fulvidraco) were sent from Niushan Lake Fishery, Hubei Province, China, to our laboratory for diagnosis. Macroscopic daffodil yellow mold was observed on the heads and fins of the fish and one Mucor species was isolated. Based on the morphological and molecular analysis, the species was identified as Mucor circinelloides. Its optimum growth temperature was 30 degrees C and it could not grow at 40 degrees C. The infectivity results showed wound infection could cause 100% cumulative mortalities at all experimental CFU (10(6), 10(7) and 10(8)). The cumulative…

Cryptic Mucor infection leading to massive cerebral infarction at initiation of antileukemic chemotherapy.

A 74-year-old man with newly diagnosed acute myelogenous leukemia unexpectedly suffered a massive cerebral infarct on day 2 of induction chemotherapy. Clinically, the hemorrhagic infarct was thought to be due to leukostasis and thrombocytopenia. Necropsy, however, revealed that Zygomycetes-type hyphae had infiltrated cerebral vessels in and near the infarct. The fungal infection was clinically silent otherwise, although fungal elements were also identified in the lung at autopsy. This case illustrates how closely fungal infection may resemble a leukemia-associated cerebrovascular accident.

Role of the putative sit1 gene in normal germination of spores and virulence of the Mucor lusitanicus.

Mucormycosis is a life-threatening infection caused by certain members of the fungal order Mucorales, with increased incidence in recent years. Individuals with untreated diabetes mellitus, and patients treated with deferoxamine are particularly susceptible to this infection. Elevated free iron concentrations in serum contribute to the development of mucormycosis. Pathogenic fungi have evolved multiple mechanisms to acquire and utilize free iron or extract it from the various iron-binding molecules within the host. The utilization of hydroxamate siderophores as xenosiderophores may contribute …

A Novel Resistance Pathway for Calcineurin Inhibitors in the Human-Pathogenic Mucorales Mucor circinelloides.

Mucormycosis is an emerging lethal fungal infection in immunocompromised patients. Mucor circinelloides is a causal agent of mucormycosis and serves as a model system to understand genetics in Mucorales. Calcineurin is a conserved virulence factor in many pathogenic fungi, and calcineurin inhibition or deletion of the calcineurin regulatory subunit (CnbR) in Mucor results in a shift from hyphal to yeast growth. We analyzed 36 calcineurin inhibitor-resistant or bypass mutants that exhibited hyphal growth in the presence of calcineurin inhibitors or in the yeast-locked cnbRΔ mutant background wi…

Detection of the Dimorphic Phases of Mucor circinelloides in Blood Cultures from an Immunosuppressed Female.

Mucormycosis fungemia is rarely documented since blood cultures are nearly always negative. We describe a case of Mucor circinelloides fungemia in a patient with a history of a sinus infection, sarcoidosis, and IgG deficiency. The identity of the isolate was supported by its microscopic morphology and its ability to convert into yeast forms under anaerobic conditions. The early detection, initiation of liposomal amphotericin B treatment, and reversal of underlying predisposing risk factors resulted in a good outcome.

Gastrointestinal zygomycosis caused by Mucor indicus in a patient with acute traumatic brain injury.

We report a gastrointestinal infection caused by Mucor indicus in a patient with severe head injuries. Monotherapy with high-dose liposomal amphotericin B successfully eradicated the mucormycosis. Nevertheless, subsequently a hemicolectomy was necessary due to recurrent bleeding from a deep ulcer. Mucor indicus is an uncommon fungal pathogen, typically found in starters used for food fermentation. Reviewing other reports on Mucor indicus infections, primary gastrointestinal manifestations seem to be typical and indicate an oral route of infection.

Fracture-Related Infection of the Lower Limb Caused by Mucor velutinosus: Amputation or Salvation?

Fracture-related infections caused by mucormycosis are rare and potentially fatal. Evidence-based experience with its treatment is limited, and surgical management ranges from limb salvage to amputation, with indications not always clear. A 56-year-old woman was admitted after an aircraft accident, sustaining major trauma injuries, including a Gustilo-Anderson type III open ankle joint fracture. Initial damage control surgery with external fixation ensued, followed by secondary, definitive internal fixation with plate and screws. The patient developed a fracture-related infection in the ankle …

Clinical Significance of Mucor in Airway Culture of Immunocompetent Patients With Chronic Lung Disease.

OBJECTIVES: Mucor within the airways of immunocompromised patients often signifies an invasive life-threatening infection. However, its significance in immunocompetent patients with chronic lung diseases is less clear. We aimed to assess the clinical implication of mucor in airway-secretion cultures of these patients. METHODS: A single-center retrospective cohort study was performed. Patients with cystic fibrosis (CF), primary ciliary dyskinesia (PCD) or non-CF/non-PCD bronchiectasis followed in our Pediatric Pulmonary Institute, with sputum or bronchoalveolar lavage cultures growing Mucorale…

Response of Psychrotolerant Fungus Mucor flavus to Cell Wall Stress, Induced by Azo Dyes.

The cell wall (CW) of Mucoromycota has a unique chitin/chitosan complex, unlike chitin/glucan complex in Ascomycota. Under cell wall stress (CWS), induced by azo dyes, ascomycetes increase the amount of CW chitin. This study analyzes the response of Mucor flavus to CWS, induced by Congo red and Calcofluor white. It was found that azo dyes significantly reduced the biomass yield and inhibited apical growth and branching but did not lead to an increase in the amount of CW chitin/chitosan, neutral polysacchrides and cytosol osmolytes. Non-bilayer phosphatidic acids and phosphatidylethanolamines d…

Mucor melanoleucus (Mucorales, Mucoromycota), A New Species Isolated from Ailuropoda melanoleuca (Giant Panda) Feces.

Mucor is a species-rich genus in Mucoraceae, predominantly saprotrophic in soil and dung. Some Mucor members can cause mucormycosis but also have valuable practical applications in kinds of industries such as the food and pharmaceutical industry. Mucor melanoleucus sp. nov. isolated from giant panda feces is described herein based on morphological characteristics, physiological experiments and phylogenetic analyses. Phylogenetic analysis of the internal transcribed spacer (ITS) - large subunit of ribosomal DNA (LSU rDNA) sequences demonstrates that this new species is closely allied to M. satu…
