Response of heterocyst differentiation of Dolichospermum to different forms of nitrogen deficiency.

In recent years, initiatives aimed at mitigating eutrophication have successfully reduced nitrogen and phosphorus concentrations in numerous lakes across China. Notably, the management of total nitrogen levels has prompted a shift in the dominant genera responsible for harmful algal blooms. Among these, Dolichospermum, a typical diazotrophic cyanobacterium, exhibits the ability to differentiate heterocysts for atmospheric N2 fixation under nitrogen-limited conditions. However, the underlying mechanisms driving heterocyst differentiation in response to the absence of specific nitrogen compounds

Production of nitrogen oxides by lightning and coronae discharges in simulated early Earth, Venus and Mars environments.

We present measurements for the production of nitrogen oxides (NO and N2O) in CO2-N2 mixtures that simulate different stages of the evolution of the atmospheres of the Earth, Venus and Mars. The nitrogen fixation rates by two different types of electrical discharges, namely lightning and coronae, were studied over a wide range in CO2 and N2 mixing ratios. Nitric oxide (NO) is formed with a maximum energy yield estimated to be ~1.3 x 10(16) molecule J-1 at 80% CO2 and ~1.3 x 10(14) molecule J-1 at 50% CO2 for lightning and coronae discharges, respectively. Nitrous oxide (N2O) is only formed by

Interaction Mechanisms Among Soil Environmental Factors, Microbial Communities, and Nitrogen-Cycling Functional Genes in Cool-Climate Maize Fields.

Cool-climate maize fields are characterized by low soil temperatures, strong seasonal hydrothermal fluctuations, and peat-influenced soil profiles, which may lead to patterns of nitrogen (N) cycling distinct from those in conventional agricultural soils. During maize growth, soils from three depths were characterized using physicochemical measurements, N-transformation and enzyme-activity assays, metagenomic sequencing, Mantel tests, variation partitioning analysis, and partial least squares path modeling (PLS-PM). Soil environmental factors varied significantly over time and with depth; soil

PII protein is essential for transcriptional regulation of anf gene cluster for iron-only nitrogenase in Rhodopseudomonas palustris.

In addition to catalyzing the biological nitrogen fixation, iron-only (Fe-only) nitrogenase is also able to reduce carbon dioxide (CO2) to formate (HCOO-) and methane (CH4). AnfA is responsible for the transcriptional activation of the anf gene cluster for Fe-only nitrogenase, whose expression is repressed by fixed nitrogen. However, it remains unclear how AnfA is regulated to control the expression of Fe-only nitrogenase. Here, we found that in Rhodopseudomonas palustris, PII proteins play a critical role in regulating the expression of Fe-only nitrogenase genes via AnfA. We hypothesize that

Liquid nitrogen-treated recycled autograft for reconstruction of a large tibial defect following wide resection of adamantinoma: a case report.

INTRODUCTION AND IMPORTANCE: Adamantinoma is a rare, low-grade malignant bone tumor with a predilection for the tibial diaphysis. Wide resection is the standard treatment, but reconstruction of large segmental tibial defects remains technically challenging. Liquid nitrogen-treated recycled autografts offer biological reconstruction, though experience with adamantinoma is extremely limited. CASE PRESENTATION: A 36-year-old male presented with a 6-month history of progressive left tibial pain and swelling. Imaging revealed a 20-cm expansile osteolytic mid-diaphyseal lesion, confirmed histologic

Elevated CO2 alters the feeding behaviour of the pea aphid by modifying the physical and chemical resistance of Medicago truncatula.

Elevated CO(2) compromises the resistance of leguminous plants against chewing insects, but little is known about whether elevated CO(2) modifies the resistance against phloem-sucking insects or whether it has contrasting effects on the resistance of legumes that differ in biological nitrogen fixation. We tested the hypothesis that the physical and chemical resistance against aphids would be increased in Jemalong (a wild type of Medicago truncatula) but would be decreased in dnf1 (a mutant without biological nitrogen fixation) by elevated CO(2). The non-glandular and glandular trichome density

Networks in plant nitrate foraging: From model plants to crop improvement.

Land plants must cope with the spatiotemporal heterogeneity of soil nutrients. During evolution, plants have developed sophisticated systems to perceive nutrient distribution and adaptively remodel their root system architecture (RSA) to maximize nutrient acquisition while minimizing energy expenditure. This process, referred to as nutrient foraging, involves local nutrient perception at the root, signal integration in the shoot, and subsequent RSA adjustment. Over the past decade, significant progress has been made in understanding how plants sense spatially heterogeneous nutrient availabilit

The Power of Symbiosis in Life and Science.

This article traces more than four decades of Eva Kondorosi's personal life and scientific journey in symbiotic nitrogen fixation, from early insights into nitrogenase structure to the molecular mechanisms governing root nodule development and symbiotic cell differentiation in the Medicago-Sinorhizobium meliloti symbiosis. Effective symbiosis depends on precise molecular communication between the partners, beginning in the soil and continuing through a highly coordinated, progressive differentiation program. A defining feature of symbiotic cell development is endoreduplication in both host pla

[Inhibitory analysis of the respiration of bacteroids from the nodules of yellow lupine].

Oxygen uptake and reduction of C2H2 by bacteroids was found to be inhibited by low concentrations of cyanide and azide. However, oxygen uptake was not completely suppressed even by 10(-3) M KCN. Cyanide-resistant respiration was not inhibited by salicyl-hydroxamic acid, and seemed to be accomplished at the account of autoxidable flavo-proteins. A small light-reversible inhibition of respiration by carbon monoxide was found only in the bacteroids with a high rate of nitrogen fixation. Rotenone, antimycin A, and tenoyltrifluoroacetone inhibited oxygen uptake and methylene reduction. Nitrogen fix

The enigma of progress in denitrification research.

Humans have dramatically increased the amount of reactive nitrogen (primarily ammonium, nitrogen oxides, and organically bound N) circulating in the biosphere and atmosphere, creating a wide array of desirable products (e.g., food production) and undesirable consequences (e.g., eutrophication of aquatic ecosystems and air pollution). Only when this reactive N is converted back to the chemically unreactive dinitrogen (N2) form, do these cascading effects of elevated reactive N cease to be of concern. Among the quantitatively most important processes for converting reactive N to N2 gas is the bi

Transcriptomic responses to shifts in light and nitrogen in two congeneric diatom species.

Light and nitrogen availability are basic requirements for photosynthesis. Changing in light intensity and nitrogen concentration may require adaptive physiological and life process changes in phytoplankton cells. Our previous study demonstrated that two Thalassiosira species exhibited, respectively, distinctive physiological responses to light and nitrogen stresses. Transcriptomic analyses were employed to investigate the mechanisms behind the different physiological responses observed in two diatom species of the genus Thalassiosira. The results indicate that the congeneric species are diffe

Natural 15N15N abundances constrain fixed nitrogen loss.

Nitrogen regulates primary productivity across much of the biosphere, yet fixed nitrogen loss remains poorly constrained because existing methods rely on indirect proxies or ex situ experiments. In this study, we show that natural abundances of the rare 15N15N isotopologue of dinitrogen (N2) provide a direct tracer of biological N2 production across diverse aquatic environments. N2 produced by denitrification and anammox has a near-stochastic 15N15N distribution [0 per mil (‰)], whereas atmospheric N2 carries a distinct 15N15N excess (19‰), allowing the two sources to be quantitatively disting

Functional Nitrogenase Cofactor Maturase NifB in Mitochondria and Chloroplasts of Nicotiana benthamiana.

Engineering plants to synthesize nitrogenase and assimilate atmospheric N2 will reduce crop dependency on industrial N fertilizers. This technology can be achieved by expressing prokaryotic nitrogen fixation gene products for the assembly of a functional nitrogenase in plants. NifB is a critical nitrogenase component since it catalyzes the first committed step in the biosynthesis of all types of nitrogenase active-site cofactors. Here, we used a library of 30 distinct nifB sequences originating from different phyla and ecological niches to restore diazotrophic growth of an Azotobacter vineland

[Effects of the nitrogen-fixing and phosphate-solubilizing bacterial strain T4 on the growth and rhizosphere environment of Astragalus membranaceus var. mongholicus].

Plant growth promoting rhizobacteria (PGPR) play an important role in promoting plant growth and improving soil environment. This is particularly relevant in the context of the current high-quality development of the traditional Chinese medicine industry, where the application of PGPR in the cultivation of medicinal plants contributes to ensuring the quality and safety of medicinal materials and promoting sustainable and high-quality industrial development. In this study, Astragalus membranaceus var. mongholicus was used as the object to explore the effects of nitrogen-fixing and phosphate-sol

Role of Environmental Factors in Legume-Rhizobium Symbiosis: A Review.

Legumes play a pivotal role in addressing global challenges of food and nutrition security by offering a sustainable source of protein and bioactive compounds. The capacity of legumes to establish symbiotic relationships with rhizobia bacteria enables biological nitrogen fixation (BNF), reducing the dependence on chemical fertilizers while enhancing soil health. However, the efficiency of this symbiosis is significantly influenced by environmental factors, such as soil acidity, salinity, temperature, moisture content, light intensity, and nutrient availability. These factors affect key process

Cross-Feeding of Carbon and Nitrogen Between Aquificales and Thermus in Hot Springs.

Acquisition and cycling of carbon and nitrogen among members of hot spring communities are not well understood. Metagenomic analyses of 105 communities inhabiting high temperature hot springs across Yellowstone and Iceland showed a co-distribution pattern of putatively autotrophic and/or diazotrophic (nitrogen-fixing) Aquificales and Thermus populations. Targeted enrichment of autotrophic and diazotrophic populations in an Icelandic hot spring produced a co-culture of Pampinifervens (Aquificales) that encoded carbon dioxide and nitrogen fixation pathways and Thermus (Thermales). Growth experim

Phenotypic and molecular characterisation of efficient nitrogen-fixing Azotobacter strains from rice fields for crop improvement.

Biological nitrogen fixation (BNF) is highly effective in the field and potentially useful to reduce adverse effects chemical fertilisers. Here, Azotobacter species were selected via phenotypic, biochemical and molecular characterisations from different rice fields. Acetylene reduction assay of Azotobacter spp. showed that Azotobacter vinelandii (Az3) fixed higher amount of nitrogen (121.09 nmol C2H4 mg(-1) bacteria h(-1)). Likewise, its plant growth functions, viz. siderophore, hydrogen cyanide, salicylic acid, IAA, GA3, zeatin, NH3, phosphorus solubilisation, ACC deaminase and iron tolerance

Spatial Variation in Nodule Flavonoids of Soybean by Exogenous Nitrogen.

Exogenous nitrogen (N) strongly affects soybean nodulation and N fixation; the regulatory mechanisms by which it modulates the flavonoid metabolic network remain largely unclear. In this study, we used a unilateral nodulation system in dual-rooted soybean plants and combined physiological measurements, quantitative real-time PCR (qPCR), and matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) to examine how exogenous N affects flavonoid metabolism, nodule development and N fixation activity. Exogenous N significantly decreased nodule number, dry weight and nitrogen

The Medicago genome provides insight into the evolution of rhizobial symbioses.

Legumes (Fabaceae or Leguminosae) are unique among cultivated plants for their ability to carry out endosymbiotic nitrogen fixation with rhizobial bacteria, a process that takes place in a specialized structure known as the nodule. Legumes belong to one of the two main groups of eurosids, the Fabidae, which includes most species capable of endosymbiotic nitrogen fixation. Legumes comprise several evolutionary lineages derived from a common ancestor 60 million years ago (Myr ago). Papilionoids are the largest clade, dating nearly to the origin of legumes and containing most cultivated species.

Mechanism of coordinated synthesis of the antagonistic regulatory proteins NifL and NifA of Klebsiella pneumoniae.

The nifLA operon of Klebsiella pneumoniae codes for the two antagonistic regulatory proteins which control expression of all other nitrogen fixation genes. NifA is a transcriptional activator, and NifL inhibits NifA. The importance of a correct NifL-NifA stoichiometry for efficient regulation of nitrogen fixation genes has been investigated by constructing a strain with an altered nifL-nifA gene dosage ratio, resulting from the integration of an extra copy of nifA. Results showed that a balanced synthesis of both gene products is essential for correct regulation. Effects of mutations provoking
