Osteopetrosis is a rare inherited bone disorder characterized by increased bone density owing to failure in bone resorption by the osteoclasts. Osteopetrosis is a rare hereditary disorder of osteoclast dysfunction leading to abnormally dense and sclerotic bones that are fragile and break easily. The clinical radiologic manifestations of 8 patients with osteopetrosis (4 males and 4 females) are analyzed. Six of the cases are benign. The pathologic basis for the radiologic manifestations is discussed, and a classification is suggested. "Broom-like" metaphyses, metaphyses containing three or four dense layers of bone, and ossification of soft tissues around joints were found and differential diagnosis was made.

The histologic features of bone callus after a traumatic fracture in a patient with osteopetrosis are presented. The fracture callus develops in stages that are apparently normal. The tissue is initially rich in bone-forming cells and vessels. One year later, however, unlike mature osteopetrotic bone, the tissue shows no Haversian organization.

Since decades, retroviruses are known to induce the formation of dense bones (osteopetrosis) in animals, either by increasing osteoblastic proliferation (ALVs viruses) or by decreasing osteoclastic bone resorption (FeLV virus).
