Solitary polypoid laryngeal xanthoma.

We report the case of a 51-year-old male smoker with diabetes mellitus and hyperlipidaemia and a long history of human immunodeficiency virus (HIV)/hepatitis C virus (HCV) infection treated with various antiretroviral regimes, who was referred to the otolaryngology department with progressive dysphonia. Fibre-optic laryngoscopy showed a solitary, yellowish-white pedunculated polyp on the anterior third of the left cord, with no other abnormality. Pathological analysis revealed a polypoid laryngeal xanthoma that was immunoreactive against CD68, perilipin, and adipophilin. This unusual laryngeal

Metabolic stress remodels the Golgi-sphingolipid-lipid droplet axis in INS-1E β-cells: an integrated molecular signature of early β-cell vulnerability.

BACKGROUND: Glucotoxicity, lipotoxicity, and glucolipotoxicity contribute to pancreatic β-cell dysfunction in type 2 diabetes. Although endoplasmic reticulum stress has been widely studied, the response of the Golgi apparatus to metabolic stress and its relationship with sphingolipid metabolism, lipid droplets, and insulin granule-associated pathways remain poorly understood. We examined these processes together in INS-1E β-cells exposed to different forms of metabolic stress. METHODS AND RESULTS: INS-1E cells were exposed to high glucose (33 mM), palmitic acid (0.5 mM), or both for 24 h. Cel

Caveolin-2 knockout suppresses the formation and progression of oral leukoplakia via lipid metabolism disruption.

BACKGROUND: Caveolin-2 (CAV2) is a lipid droplet (LD)-associated protein. Its role in oral leukoplakia (OLK), an oral potentially malignant disorder, and the underlying lipid metabolism mechanisms remain unclear. METHODS: CAV2 conditional knockout (cKO; CAV2flox/flox; K14-Cre) and control (Flox) C57BL/6 mice were used to establish OLK models via 4-nitroquinoline-N-oxide (4NQO). A subset received a high-fat diet (HFD). Oil red O staining, RNA sequencing, multiplex immunofluorescence (Ki-67, E-cadherin, Perilipin-1, FABP5), ELISA (acetyl-CoA), serum biochemistry (ALT, TG), and targeted GC-MS/MS

Linoleic acid drives human hypertrophic scar derived fibroblasts toward a PLIN2 + /PPARγ + lipo-fibroblast phenotype.

BACKGROUND: Activated myofibroblasts are considered the primary drivers of hypertrophic scar (HS) formation. Although traditionally viewed as terminally differentiated, recent evidence in pulmonary fibrosis suggests that myofibroblasts can be transformed toward lipo-fibroblasts, representing a potential pathway for fibrosis resolution. Whether a similar phenotypic transition occurs in skin scarring remains unexplored. OBJECTIVES: Given the role of linoleic acid (LA) in adipogenesis, this study aimed to investigate whether LA can induce the transition of hypertrophic scar derived fibroblasts (

Validation of a murine proteome-wide phage display library for the identification of autoantibody specificities.

Autoimmunity is characterized by loss of tolerance to tissue-specific as well as systemic antigens, resulting in complex autoantibody landscapes. Here, we introduce and extensively validate the performance characteristics of a murine proteome-wide library for phage display immunoprecipitation and sequencing (PhIP-seq), to profile mouse autoantibodies. This system and library were validated using seven genetic mouse models across a spectrum of autoreactivity. Mice deficient in antibody production (Rag2-/- and μMT) were used to model non-specific peptide enrichments, while cross-reactivity was e

Endomyocardial proteomic signature corresponding to the response of patients with dilated cardiomyopathy to immunoadsorption therapy.

UNLABELLED: Dilated cardiomyopathy (DCM) is a disease of the myocardium with reduced left ventricular ejection fraction (LVEF). Cardiac autoantibodies (AAbs) play a causal role in the development and progression of DCM. Removal of AAbs using immunoadsorption (IA/IgG) has been shown as a therapeutic option to improve cardiac function. However, the response to therapy differs significantly among patients. The reasons for this variability are not completely understood. Hitherto, no potential biomarker is available to predict improvement of cardiac function after therapy accurately. This shotgun p

PLIN5 contributes to lipophagy of hepatic stellate cells induced by inorganic arsenic.

Arsenic exposure triggers the activation of hepatic stellate cells (HSCs), resulting in liver fibrosis (LF). A significant decrease in lipid droplets marks the activation of HSCs. However, the exact underlying molecular mechanism remains elusive. Lipophagy, a specialized form of selective autophagy, is crucial for the degradation of lipid droplets to maintain intracellular lipid metabolism homeostasis. In this study, arsenic treatment induced lipophagy, as evidenced by the co-localization of LC3 with lipid droplets. Remarkably, arsenic exposure increased the expression levels of Perilipin 5 (P

Unraveling the mechanism of tripterygium glycosides tablets-induced liver injury and the protective role of total glucosides of peony from immune-metabolic dysregulation to multi-cellular cascade.

Tripterygium glycosides tablets (TGT) are effective against autoimmune diseases but cause significant drug-induced liver injury (DILI) that limits clinical use. While TGT disrupts hepatic iron-lipid homeostasis and co-administration with Total Glucosides of Peony (TGP) mitigates its toxicity, the multi-cellular mechanisms remain unclear. In the current study, using integrative single-cell RNA sequencing and pathological validation in controlled mouse models (TGT vs.Con and TGT + TGP vs. TGT.) we elucidated a pathogenic iron-lipid axis driving hepatotoxicity via cellular cascades. TGT initiated

Sex-Specific Effects of Long-Term Antipsychotic Drug Treatment on Adipocyte Tissue and the Crosstalk to Liver and Brain in Rats.

Antipsychotic drug (APD) medication can lead to metabolic dysfunctions and weight gain, which together increase morbidity and mortality. Metabolically active visceral adipose tissue (VAT) in particular plays a crucial role in the etiopathology of these metabolic dysregulations. Here, we studied the effect of 12 weeks of drug medication by daily oral feeding of clozapine and haloperidol on the perirenal fat tissue as part of VAT of male and female Sprague Dawley rats in the context of complex former investigations on brain, liver, and blood. Adipocyte area values were determined, as well as tri

Exercise-inducible factors to activate lipolysis in adipocytes.

We examined the effects of exercise training on the levels of lipid droplet (LD)-associated and mitochondria-related proteins in diet-induced obese (DIO) rats. Furthermore, we assessed putative factors induced by exercise to activate lipolysis in differentiated 3T3-L1 adipocytes. DIO Wistar male rats (age 20 wk) were divided into sedentary control (SED, n = 7) and exercise training (EX, n = 7) groups. EX animals were subjected to treadmill running (25 m/min, 1 h/day, 5 days/wk) for 6 wk. Epididymal fat was dissected and used for protein analyses. 3T3-L1 adipocytes were incubated with media con

The origin and development of fat droplet and its role in health and illness.

Lipid droplets are found in most cells, where they are needed for lipid metabolism and energy homeostasis. Their biogenesis is still not entirely described process, in which an organelle with monomolecular layer on the surface originates from two-layer membrane of the smooth endoplasmic reticulum. Large lipid droplets can be formed either by growth or by fusion of smaller lipid droplets. Their basic functions are controlled lipid deposition and lipid metabolism and the protection of the intracellular environment from lipotoxicity. Currently, dysfunction of lipid droplet is associated with a nu

OPA1 in Lipid Metabolism: Function of OPA1 in Lipolysis and Thermogenesis of Adipocytes.

OPA1 (Optic Atrophy 1) is a mitochondrial GTPase known to regulate fission of mitochondria. It was recently also shown to locate on lipid droplets in adipocytes where it functions as an A-kinase anchoring protein (AKAP) that mediates adrenergic control of lipolysis by facilitating PKA phosphorylation of perilipin (Plin1). In brown adipocytes indirect evidence support the notion that OPA1 regulation of fission serves to increase thermogenesis, which thereby contributes to dissipation of energy. In white adipocytes, OPA1 located on lipid droplets serves as a gatekeeper to control lipolysis induc

Loss of cardiomyocyte AKT signaling causes deterioration of lipid metabolism and cellular atrophy.

The mammalian heart critically depends on oxidative metabolism of fatty acids, glucose, ketones, and amino acids to meet its extensive ATP d … ardiomyocytes lost the ability to store fatty acids in lipid droplets (LDs) due to an early loss of perilipins and other proteins involved in LD generation and function. In conclusion, our data show that general, isoform-independent AKT signaling in cardiac myocytes is indispensable for preservation of cardiac fatty acid metabolism and energy supply.

Lipases and lipid droplet-associated protein expression in subcutaneous white adipose tissue of cachectic patients with cancer.

BACKGROUND: Cancer cachexia is a multifactorial metabolic syndrome characterized by marked loss of adipose tissue and skeletal muscle. Fat loss from adipose tissue in cancer cachexia is partly the result of increased lipolysis. Despite the growing amount of studies focused on elucidating the mechanisms through which lipolysis-related proteins regulate the lipolytic process, there are scarce data concerning that profile in the adipose tissue of cancer cachectic patients. Considering its fundamental importance, it was our main purpose to characterize the expression of the lipolysis-related prote
