Methods to Estimate Carbon Export from Leaves.

In most plant species, the majority of carbon assimilation takes place in leaves. The export of carbon from leaves to the rest of the plant's organs is thus a fundamental process in plant biology. At the same time, only a few studies have estimated the rate of this process and its dynamics. In the current chapter, we will describe two simple methods for evaluating the rate of carbon export from leaves. Both methods calculate the export as the difference between carbon assimilation and carbon storage. The first method evaluates the carbon storage based on the leaf mass per area (LMA) difference

Manganese deficiency of silver fir trees (Abies alba) at a reforested site in the Jura mountains, Switzerland: aspects of cause and effect.

We examined manganese uptake and translocation in 30-year-old silver fir trees (Abies alba Mill.) showing severe symptoms of needle chlorosis by analyzing both xylem and phloem sap of shoots and an extract of root sap originating from both xylem and phloem elements. Manganese concentrations in shoot xylem and phloem sap were significantly lower in chlorotic trees than in neighboring healthy trees. The Mn concentration of root sap was also lower in these Mn-deficient trees than in healthy trees, indicating reduced Mn uptake by Mn-deficient trees. Although Mn deficiency had no effect on the Mn c…

Based on the collective data, a tentative model describing the relationship between long-distance phloem transport and cadmium distribution in sink organs is proposed. The implications for fo …

Immune activation drives citrus Huanglongbing.

Citrus Huanglongbing (HLB), caused by the phloem-limited bacterium Candidatus Liberibacter asiaticus (CLas) and considered the most destructive citrus disease worldwide, has long been regarded as a typical bacterial infection. However, recent compelling evidence shows that CLas drives HLB through chronic effector-triggered immunity, not via direct pathogenicity, thereby redefining HLB as an immune-mediated disease (Huang et al.).

Inducible biosynthesis and immune function of the systemic acquired resistance inducer N-hydroxypipecolic acid in monocotyledonous and dicotyledonous plants.

Recent work has provided evidence for the occurrence of N-hydroxypipecolic acid (NHP) in Arabidopsis thaliana, characterized its pathogen-in … stinct monocotyledonous and dicotyledonous species. Pronounced systemic accumulation of NHP in leaf phloem sap of locally inoculated cucumber supports a function for NHP as a phloem-mobile immune signal. Our study thus generalizes the existence and function of an NHP resistance pathway in plant systemic acquired resistance.

Elevated CO2 alters the feeding behaviour of the pea aphid by modifying the physical and chemical resistance of Medicago truncatula.

Elevated CO(2) compromises the resistance of leguminous plants against chewing insects, but little is known about whether elevated CO(2) modifies the resistance against phloem-sucking insects or whether it has contrasting effects on the resistance of legumes that differ in biological nitrogen fixation. We tested the hypothesis that the physical and chemical resistance against aphids would be increased in Jemalong (a wild type of Medicago truncatula) but would be decreased in dnf1 (a mutant without biological nitrogen fixation) by elevated CO(2). The non-glandular and glandular trichome density…

Combined transcriptome and metabolome analysis of Nerium indicum L. elaborates the key pathways that are activated in response to witches' broom disease.

BACKGROUND: Nerium indicum Mill. is an ornamental plant that is found in parks, riversides, lakesides, and scenic areas in China and other parts of the world. Our recent survey indicated the prevalence of witches' broom disease (WBD) in Guangdong, China. To find out the possible defense strategies against WBD, we performed a MiSeq based ITS sequencing to identify the possible casual organism, then did a de novo transcriptome sequencing and metabolome profiling in the phloem and stem tip of N. indicum plants suffering from WBD compared to healthy ones. RESULTS: The survey showed that Wengyuen …

Engineering principles in plant metabolism: integrating control, mechanics, and transport under dynamic environments.

Plant responses to dynamic environmental conditions remain challenging to describe within a unified mechanistic framework. This is largely b … pendent scaling relationships in plant tissues; and (iii) fluid dynamics, which describes xylem and phloem transport as flow through networks under variable demand. For each domain, we present plant-specific case studies demonstrating how engineering analysis can reveal constraints and dynamic relationships that are difficult to extract from descriptive …

Differential Costs of Two Distinct Resistance Mechanisms Induced by Different Herbivore Species in Arabidopsis.

Plants respond to herbivory with the induction of resistance, mediated by distinct phytohormonal signaling pathways and their interactions. Phloem feeders are known to induce plant resistance via the salicylic acid pathway, whereas biting-chewing herbivores induce plant resistance mainly via the jasmonate pathway. Here, we show that a specialist caterpillar (biting-chewing herbivore) and a specialist aphid (phloem feeder) differentially induce resistance against Pieris brassicae caterpillars in Arabidopsis (Arabidopsis thaliana) plants. Caterpillar feeding induces resistance through the jasmon…

A transcriptomic network underlies microstructural and physiological responses to cadmium in Populus x canescens.

Bark tissue of Populus × canescens can hyperaccumulate cadmium, but microstructural, transcriptomic, and physiological response mechanisms are poorly understood. Histochemical assays, transmission electron microscopic observations, energy-dispersive x-ray microanalysis, and transcriptomic and physiological analyses have been performed to enhance our understanding of cadmium accumulation and detoxification in P. × canescens. Cadmium was allocated to the phloem of the bark, and subcellular cadmium compartmentalization occurred mainly in vacuoles of phloem cells. Transcripts involved in microstru…

Transgenic tomato strategies targeting whitefly eggs from apoplastic or ovary-directed proteins.

BACKGROUND: Transgenic plants expressing proteins that target the eggs of the ubiquitous plant pest Bemisia tabaci (whitefly) could be an effective insecticide strategy. Two approaches for protein delivery are assessed using the mCherry reporter gene in transgenic tomato plants, while accommodating autofluorescence in both the plant, phloem-feeding whitefly and pedicle-attached eggs. RESULTS: Both transgenic strategies were segregated to homozygous genotype using digital PCR. The first strategy uses a glycotransferase secretion signal peptide. Despite bright apoplastic accumulation, mCherry i…

Orally Delivered Scorpion Antimicrobial Peptides Exhibit Activity against Pea Aphid (Acyrthosiphon pisum) and Its Bacterial Symbionts.

Aphids are severe agricultural pests that damage crops by feeding on phloem sap and vectoring plant pathogens. Chemical insecticides provide an important aphid control strategy, but alternative and sustainable control measures are required to avoid rapidly emerging resistance, environmental contamination, and the risk to humans and beneficial organisms. Aphids are dependent on bacterial symbionts, which enable them to survive on phloem sap lacking essential nutrients, as well as conferring environmental stress tolerance and resistance to parasites. The evolution of aphids has been accompanied …

The distribution of monolignol glucosides coincides with lignification during the formation of compression wood in Pinus thunbergii.

The distributions of monolignol glucosides (MLGs) in compression and opposite woods of Pinus thunbergii were assessed using cryo-time-of-flight secondary ion mass spectrometry to investigate their involvement in lignification. p-Glucocoumaryl alcohol (PG) was identified in the region of the differentiating xylem adjacent to the cambial zone only in compression wood, whereas coniferin (CF) was similarly localized in both compression and opposite woods. Their distribution from the phloem to the xylem was evaluated by high-performance liquid chromatography (HPLC) using serial tangential sections.…

Unveiling Foliar-Applied Calcium Translocation in Tomato: Evidence from Leaf-to-Fruit Movement Using Strontium Tracer.

Calcium (Ca) deficiency can impair fruit development even under optimal soil Ca levels due to its transpiration-dependent transport. Because fruits exhibit significantly lower transpiration rates than leaves, any further reduction in fruit transpiration during development can limit Ca delivery, leading to lower fruit Ca content and diminished quality. Foliar application of Ca offers a potential strategy to mitigate these effects; however, its low mobility in the phloem often limits treatment efficacy. To better understand this phenomenon, we employed X-ray fluorescence spectroscopy (XRF) to in…

Sulla carnosa modulates root invertase activity in response to the inhibition of long-distance sucrose transport under magnesium deficiency.

Being the principal product of photosynthesis, sucrose is involved in many metabolic processes in plants. As magnesium (Mg) is phloem mobile, an inverse relationship between Mg shortage and sugar accumulation in leaves is often observed. Mg deficiency effects on carbohydrate contents and invertase activities were determined in Sulla carnosa Desf. Plants were grown hydroponically at different Mg concentrations (0.00, 0.01, 0.05 and 1.50 mM Mg) for one month. Mineral analysis showed that Mg contents were drastically diminished in shoots and roots mainly at 0.01 and 0.00 mM Mg. This decline was a…

Constitutive overexpression of the sucrose transporter SoSUT1 in potato plants increases arbuscular mycorrhiza fungal root colonization under high, but not under low, soil phosphorus availability.

The sucrose transporter SUT1 functions in phloem loading of photoassimilates in solanaceous plant species. In the present study, wildtype and transgenic potato plants with either constitutive overexpression or antisense inhibition of SUT1 were grown under high or low phosphorus (P) fertilization levels in the presence or absence of the arbuscular mycorrhizal (AM) fungus Glomus intraradices. At a low soil P fertilization level, the extent of AM fungal root colonization was not different among the genotypes. In all plants, the AM symbiosis contributed significantly to P uptake under these condit…
