Group B streptococci (GBS) are a major cause of sepsis and meningitis in infants. While antibodies directed to the type-specific GBS capsule have been shown to be protective, it is less clear whether antibodies to the group B polysaccharide, a noncapsular, cell wall-associated antigen, may play a role in immunity. To investigate the functional activity of group B polysaccharide-specific antibodies, we tested sera from rabbits vaccinated with group B polysaccharide coupled to tetanus toxoid (B-TT). Anti-B-TT was weakly opsonic in vitro for a highly encapsulated type III strain, while antiserum

Bacterial capsular polysaccharides have been used as effective vaccines for adults but infants and seniors respond poorly to these immunogens because pure polysaccharides are unable to activate T-cells resulting in antibodies of low affinity and poor immunological memory. These deficiencies are addressed by conjugate vaccines composed of bacterial polysaccharide covalently attached to protein carriers such as tetanus or diphtheria toxoids. These vaccines activate T-cells and have been hugely effective in reducing the incidence of dangerous infectious diseases such as bacterial meningitis in in

Inherited deficiencies of complement components are associated with an increased risk of infection by encapsulated, high grade bacterial pathogens such as Streptococcus pneumoniae, Haemophilus influenzae type b and Neisseria meningitidis. Hence, the levels of antibodies to bacterial capsular polysaccharide antigens were measured using ELISA in 65 patients with inherited deficiencies covering the classical, alternative and terminal components of the complement cascade. Three of the four C3-deficient individuals studied were found to be almost totally deficient in specific anti-pneumococcal caps

Hypervirulent Klebsiella pneumoniae is known for its increased extracellular polysaccharide production. Biofilm matrices of hypervirulent K. pneumoniae have increased polysaccharide abundance and are uniquely susceptible to disruption by peptide bactenecin 7 (bac7 (1-35)). Here, using confocal microscopy, we show that polysaccharides within the biofilm matrix collapse following bac7 (1-35) treatment. This collapse led to the release of cells from the biofilm, which were then killed by the peptide. Characterization of truncated peptide analogs revealed that their interactions with polysaccharid

Hepatocellular carcinoma (HCC) is a major cause of cancer mortality worldwide, and its limited response to conventional chemotherapeutics underscores the need for safer, multifunctional approaches. Natural polysaccharides, complex carbohydrates derived from plants, fungi, algae, animals, and bacteria, represent a structurally diverse class of bioactive macromolecules that combine therapeutic and material properties. Their monosaccharide composition, linkage types, branching patterns, molecular weights, and charge densities determine receptor recognition, intracellular trafficking, and signalin

OBJECTIVE: Systemic lupus erythematosus (SLE) is characterized by the production of multiple autoantibodies. Anti-DNA antibodies are associated with glomerulonephritis in SLE. It has been shown that anti-DNA antibodies cross-react with bacterial polysaccharide and, thus, might be elicited by microbial exposure. Non-DNA-binding antibodies also contribute significantly to the pathogenesis of lupus nephritis. The goal of this study was to characterize non-DNA-binding, kidney-binding antibodies.
METHODS: We generated a combinatorial library derived from spleen cells of a patient with SLE who had

Helicobacter pylori infects over half of the global population and is a major cause of gastritis, peptic ulcer disease, and gastric cancer. Although antibiotic-based regimens remain the clinical standard, their efficacy is being increasingly compromised by rising antimicrobial resistance, poor gastric retention, and adverse side effects. This review explores polysaccharide-based strategies as adjunctive or alternative approaches. Sulphated polysaccharides inhibit bacterial adhesion and biofilm formation while modulating inflammatory pathways, whereas chitosan provides mucoadhesive properties.

Studies exploring the potential of outer membrane protein C (OmpC) as a vaccine candidate and diagnostic antigen for Vibrio parahaemolyticus remain limited. In this study, native OmpC and a thioredoxin-fused form (TrxA-OmpC) were successfully expressed in a prokaryotic system and evaluated for their surface exposure and immunogenicity. Surface accessibility of OmpC in the presence of bacterial surface polysaccharides was investigated using antisera raised against polysaccharide-deficient mutant strains. Immune responses induced by both recombinant proteins were assessed through immunoassays, W

Fluorescent flow cytometry has become the method of choice for interrogation of bacterial populations at the single-cell level. However, limitations of this technique include issues of dynamic range, spectral overlap, photobleaching, and overall low signal intensity due to the small size of bacteria. The recent development of mass cytometry allows single-cell analysis with the resolution of inductively coupled plasma mass spectrometry, facilitating multiparametric analysis. Using a combination of a metal-based membrane stain and lectins conjugated to lanthanide-chelating polymers, we demonstra

Our current understanding of enzymatic polysaccharide degradation has come from a huge number of in vitro studies with purified enzymes. While this vast body of work has been invaluable in identifying and characterizing novel mechanisms of action and engineering desirable traits into these enzymes, a comprehensive picture of how these enzymes work as part of a native in vivo system is less clear. Recently, several model bacteria have emerged with genetic systems that allow for a more nuanced study of carbohydrate active enzymes (CAZymes) and how their activity affects bacterial carbon metaboli

BACKGROUND: Biofilm formation on voice prostheses disrupts the function and limits the lifespan of voice prostheses. There is still no effective clinical strategy for inhibiting or removing these biofilms. Silver sulfadiazine (SSD), as an exogenous antibacterial agent, has been widely used in the prevention and treatment of infection, however, its effect on voice prosthesis biofilms is unknown. The purpose of this study was to explore the effect of SSD on the mature mixed bacterial biofilms present on voice prostheses.
METHODS: Quantitative and qualitative methods, including the plate countin

The polysaccharide capsule is recognized as a major virulence factor of many bacterial pathogens, playing a significant role in evading host immunity. However, it has not yet been characterized for the bacteria within the emerging multidrug-resistant opportunistic pathogens of the Stenotrophomonas maltophilia complex. In this study, we aimed to evaluate whether bacteria of the S. maltophilia complex form a bona fide polysaccharide capsule and determine its virulence-related functions. We demonstrated that a part of clinical and environmental isolates of the S. maltophilia complex forms a polys

Pathogenic bacteria employ many strategies to overcome the host immune system for extended survival and propagation in their hosts. Components of the bacterial outer-membrane play an important role in this process. When invading the host, Gram-negative bacteria often use a strategy, known as phase variation, that involves a reversible change in antigenic determinants, frequently polysaccharides. This means that the genes encoding the outer-membrane antigens undergo reversible changes within repeated simple DNA sequence motifs. The antigenic structure of the bacterial outer-membrane is influenc

Bacteroides fragilis is the anaerobe most commonly isolated from clinical cases of intra-abdominal sepsis. In a rodent model of this disease process, intraperitoneal injection of the capsular polysaccharide complex (CPC) from B. fragilis provokes abscess formation, while subcutaneous administration of this complex confers protection against B. fragilis-induced intra-abdominal abscesses. The CPC consists of two discrete polysaccharides, polysaccharides A and B (PS A and PS B), each possessing oppositely charged structural groups critical to the ability of these carbohydrates to induce the forma

Polysaccharides are ubiquitous components of the Gram-positive bacterial cell wall. In Lactococcus lactis, a polysaccharide pellicle (PSP) forms a layer at the cell surface. The PSP structure varies among lactococcal strains; in L. lactis MG1363, the PSP is composed of repeating hexasaccharide phosphate units. Here, we report the presence of an additional neutral polysaccharide in L. lactis MG1363 that is a rhamnan composed of α-l-Rha trisaccharide repeating units. This rhamnan is still present in mutants devoid of the PSP, indicating that its synthesis can occur independently of PSP synthesis

BACKGROUND: Gallic acid (GA) is recognized for its significant antioxidative and anti-inflammatory properties. The processing and consumption of GA-rich foods influence the interaction with cell wall polysaccharides, thereby impacting the bioavailability and bioaccessibility of GA. This study aimed to develop a biomimetic dietary fiber (BDF) model composed of bacterial cellulose, pectin, and GA to investigate the interactions, digestive behavior, and antioxidant synergism between polysaccharides and GA within the BDF.
RESULTS: The microstructural, compositional, and crystalline characteristic

There is evidence that the treatment of bacterial meningitis with antibiotics liberates harmful bacterial products in the subarachnoid space (SAS). This enhances meningeal inflammation and in particular the recruitment of leukocytes into the cerebrospinal fluid (CSF), which has been shown to be more harmful than beneficial in this disease. In this study, we used a rabbit meningitis model based on intracisternal injection of live Streptococcus pneumoniae. Ampicillin (40 mg/kg of body weight given intravenously [i.v.] 16 h after induction of meningitis) caused a fivefold increase in CSF leukocyt

Chronic inflammation of the colon has been described as an inflammatory condition of the colon that is characterized by recurring flare-ups of the disease (ulcerative colitis). Ulcerative colitis is a complex inflammatory disorder of the colon that is determined by the combination of several mechanisms: abnormal bacterial flora, inappropriate immune response, and the breakdown of the protective lining of the colon. The last 10 years have seen the emergence of plant derived polysaccharides as new biological active compounds with the potential to intervene at the different levels of the disease.
