Normalization of LC-MS mycotoxin determination using the N-alkylpyridinium-3-sulfonates (NAPS) retention index system.

A major challenge for LC-MS analysis is the ability to compare data between laboratories and across instrument platforms. Currently, mycotoxin determination relies on dereplication strategies based on physicochemical properties such as the m/z of the precursor and product ions. Unlike these intrinsic properties, retention time (tR) is an extrinsic property impacted by LC conditions, including mobile phases and column chemistry, making exchange of data between groups difficult. To address this, we are introducing the concept of incorporating an electrospray compatible, retention index (RI) syst

Coupled Hydrogen-Bond-Electrostatic Recognition of Phosphatidylglycerol Drives the Design of Resistance-Suppressing Miniature Peptidomimetics.

The emergence of multidrug-resistant (MDR) pathogens has urged us to find new antimicrobial strategies. Phosphatidylglycerol (PG) is an attractive bacterial-specific lipid target but is targeted by only one clinical agent, daptomycin. Yet daptomycin, like most reported PG binders, binds PG through an imprecise hydrophobic-electrostatic mode, necessitating a relatively large molecular size. This requirement, together with its strict Ca2+ dependence, significantly limits its efficacy. Here, we report bis-pyridinium amides (BisPAs), a rationally designed class of small molecules capable of precis

The Pof1 nicotinamide mononucleotide adenylyl transferase has a non-canonical role in NAD+ metabolism in the budding yeast Saccharomyces cerevisiae.

The regulation of NAD+ metabolism and its interaction with other cellular pathways are not yet completely understood. Here we study why cells lacking the Pof1 NMNAT have high levels of nicotinamide riboside (NR), produced from NAD+ turnover and in response to nutritional stress. To understand the mechanism, we identify novel NR-producing factors using genetic screens and further characterize Phm8 as an NMN nucleotidase. In addition, loss of Pof1 moderately delays the turnover of Sdt1, another nucleotidase contributing to NR production. Interestingly, the ATPase domain of Pof1 appears to play a

A randomized placebo-controlled trial of nicotinamide riboside in older adults with mild cognitive impairment.

Nicotinamide riboside (NR) increases blood levels of NAD+, a cofactor central to energy metabolism, and improves brain function in some rodent models of neurodegeneration. We conducted a placebo-controlled randomized pilot study with the primary objective of determining safety of NR in older adults with mild cognitive impairment (MCI). Twenty subjects with MCI were randomized to receive placebo or NR using dose escalation to achieve, and maintain, a final dose of 1 g/day over a 10-week study duration. The primary outcome was post-treatment change from baseline measures of cognition (Montreal C

Oral nicotinamide riboside raises NAD+ and lowers biomarkers of neurodegenerative pathology in plasma extracellular vesicles enriched for neuronal origin.

Declining nicotinamide adenine dinucleotide (NAD+ ) concentration in the brain during aging contributes to metabolic and cellular dysfunction and is implicated in the pathogenesis of aging-associated neurological disorders. Experimental therapies aimed at boosting brain NAD+ levels normalize several neurodegenerative phenotypes in animal models, motivating their clinical translation. Dietary intake of NAD+ precursors, such as nicotinamide riboside (NR), is a safe and effective avenue for augmenting NAD+ levels in peripheral tissues in humans, yet evidence supporting their ability to raise NAD+

Unexpected cholinergic crisis caused by distigmine bromide: A case report.

RATIONALE: Distigmine bromide is a cholinesterase (ChE) inhibitor used to treat dysuria due to a hypotonic bladder. We encountered a case of cholinergic crisis caused by distigmine bromide, which resulted in a rapid decrease in serum ChE levels, hypothermia, respiratory failure, and circulatory failure within a short period of time. PATIENT CONCERNS: A 51-year-old man was admitted to a psychiatric hospital to treat behavioral disorders due to irritability and violent behavior. The patient was referred to our hospital for septic shock secondary to urinary tract infection and respiratory failur

Effect of distigmine on the contractile response of guinea pig urinary bladder to electrical field stimulation.

Distigmine bromide (distigmine) is a reversible carbamate group cholinesterase (ChE) inhibitor. Although mainly used clinically for the treatment of myasthenia gravis, distigmine is also indicated for detrusor underactivity in Japan. According to the pharmacological classification of distigmine, its therapeutic effect against detrusor underactivity appears to be produced by enhanced urinary bladder smooth muscle (UBSM) contractility due to an increased concentration of acetylcholine between parasympathetic nerve endings and UBSM cells. However, ATP as well as acetylcholine is also released fro

Formation of cyanide after i.v. administration of the oxime HI 6 to dogs.

HI 6(pyridinium, 1-[[[4-(aminocarbonyl)pyridinio]methoxy]-2- [(hydroxyimino)methyl]-dichloride belongs to a series of bisquaternary pyridinium oximes that are effective against poisoning with extremely toxic organophosphates. Since HI 6 has been shown to be unstable at pH 7.4 and to release significant amounts of cyanide, a study was undertaken to determine the degree of cyanide formation from HI 6 in vivo. When HI 6 (100 mumol/kg) was administered i.v. to dogs, the animals showed no signs of cyanide toxicity but exhibited some cholinomimetic symptoms, including retching, hypersalivation and e

[2-Chloro-1-methylpyridinium iodide. A suitable reagent for peptide synthesis].

2-Chloro-1-methylpyridinium iodide is a coupling reagent for peptide synthesis. By its use different peptides were synthesized with protected di- and trifunctional amino acids. While urethan-protected amino acids react free of racemization, the fragment condensation needs N-Hydroxysuccinimide, as shown by the Young test.

[Effect of pyrithiamine on rat sciatic nerve. (I) Morphological changes during the early stage of thiamine deficiency (author's transl)].

We observed under light and electron microscopes morphological changes in the rat sciatic nerve during the early stages of a thiamine deficient state as induced by a pyrithiamine (PT: 50 microgram/100g X 6 days) and thiamine deficient diet (TDD). We simultaneously determined thiamine levels in the whole sciatic nerve of rats. Experiments were undertaken with normal control, TDD (rats fed a TDD), PT (PT treated rats) and PTD (PT treated rats fed a TDD) groups. Microscopically, there were numerous shrunken myelinated axons with myelin ovoids of folds in the PT group and many swollen ones in the

Ammonium Catecholaldehydes as Multifunctional Bioactive Agents: Evaluating Antimicrobial, Antioxidant, and Antiplatelet Activity.

A wide range of water-soluble quaternary ammonium acylhydrazones based on catecholaldehyde were synthesized and characterized using NMR, IR spectroscopy, and elemental analysis. The total antioxidant capacity of the acylhydrazones discussed herein was estimated via coulometric titration with electrogenerated bromine. Pyridinium derivatives 11a-e exhibited the highest antioxidant capacity. Quaternary ammonium acylhydrazones demonstrated high antimicrobial activity against Gram-positive bacteria, including methicillin-resistant Staphylococcus aureus strains. Furthermore, low hemo- and cytotoxici

Expanding the Variety of Pyridinium-Based Bis-QACs with Antimicrobial Properties: Investigation into Linker Structure-Activity Correlation.

For decades quaternary ammonium compounds (QACs) have served as main component of a top antiseptic and disinfectant compositions. Among them, bis-QACs are the most prominent and effective class of biocides. Although mono-QACs still dominate the antiseptic market, their activity against Gram-negative bacteria is largely inferior to bis-QACs. Moreover, the new wave of bacterial resistance during the COVID-19 pandemic is threatening the efficiency of popular antiseptics. Therefore, the requirement for novel biocides is urgent. Reported here is a unified and simple two-step synthesis to achieve no

Источник пометил публикацию как отозванную

The H1N1 influenza pandemic of 2009 has claimed over 18,000 lives. During this pandemic, development of drug resistance further complicated efforts to control and treat the widespread illness. This research utilizes traditional Chinese medicine Database@Taiwan (TCM Database@Taiwan) to screen for compounds that simultaneously target H1 and N1 to overcome current difficulties with virus mutations. The top three candidates were de novo derivatives of xylopine and rosmaricine. Bioactivity of the de novo derivatives against N1 were validated by multiple machine learning prediction models.
