Protective effects of propafenone, propranolol, and amiodarone against isoproterenol-induced lethal arrhythmias in IGF1R deficiency mice.

The study investigates the role of the insulin-like growth factor 1 receptor (IGF1R) in cardiac function and its impact on stress-induced arrhythmias. IGF1R is a key receptor involved in cardiomyocyte growth and metabolism, with significant implications for cardiac development and stress response. However, its role in arrhythmias remains underexplored. The research utilized echocardiography to assess cardiac function in wild-type (WT) and IGF1R+/- mice, revealing no significant differences in cardiac contraction or relaxation indices under basal conditions. This suggests that IGF1R may not be

Insulin-like growth factor-I receptor in the cell biology of the ameloblast: an immunohistochemical study on the rat incisor.

The distribution of IGF-I receptor is reported in the odontogenic epithelium and mesenchyme of the continuously erupting mandibular incisor of the rat by immunohistochemistry using a polyclonal antibody specific to the IGF-I receptor. Odontogenic epithelium is a unique odontogenic sequence in that all stages of the complex life cycle of the ameloblast are represented along the length of the enamel-forming aspect of the tooth. Pre-ameloblasts become post-mitotic before secreting enamel matrix. When the full thickness of the enamel has been formed, a remarkable transition in phenotype takes plac

Linsitinib, an IGF-1R inhibitor, attenuates disease development and progression in a model of thyroid eye disease.

INTRODUCTION: Graves' disease (GD) is an autoimmune disorder caused by autoantibodies against the thyroid stimulating hormone receptor (TSHR) leading to overstimulation of the thyroid gland. Thyroid eye disease (TED) is the most common extra thyroidal manifestation of GD. Therapeutic options to treat TED are very limited and novel treatments need to be developed. In the present study we investigated the effect of linsitinib, a dual small-molecule kinase inhibitor of the insulin-like growth factor 1 receptor (IGF-1R) and the Insulin receptor (IR) on the disease outcome of GD and TED. METHODS:

Protegrin 1 Enhances Innate Cellular Defense via the Insulin-Like Growth Factor 1 Receptor Pathway.

Antimicrobial peptides (AMPs) represent a promising area of research to help combat the ever-growing problem of antibiotic resistance. Protegrin-1 is an AMP from the cathelicidin family. It is produced naturally in pigs and its mature form (mPG-1) has potent bactericidal properties and a unique β-hairpin structure that separates it from most AMPs found in mice and humans. While the antibacterial properties of protegrin-1 are well established, the role it plays in immune modulation has yet to be investigated, and our current study sought to explore this alternate role and potential mechanism be

The role of insulin-like growth factor I components in the regulation of vitamin D.

Several factors are known to be involved in the regulation of vitamin D and sunlight and diet are the two sources in humans, but the relative importance of each of them is not well defined. Vitamin D, parathyroid hormone and serum insulin-like growth factor-I (IGF-I) were found to be independent predictors of total bone density. Thus, the growth hormone (GH)/IGF-I is thought to play an important role in the regulation of bone mineral density and the skeleton is second only to the liver as a source of circulating levels of IGF-I. The mechanisms by which IGF-I may influence bone metabolism is no

Inhibition of HSF2 SUMOylation via MEL18 upregulates IGF-IIR and leads to hypertension-induced cardiac hypertrophy.

Cardiac hypertrophy is a major characteristic of early-stage hypertension-related heart failure. We have found that the insulin-like growth factor receptor II (IGF-IIR) signaling was critical for hypertensive angiotensin II-induced cardiomyocyte hypertrophy and apoptosis. Moreover, this IGF-IIR signaling was elegantly modulated by the heat shock transcription factors (HSFs) during heart failure. However, the detailed mechanism by which HSFs regulates IGF-IIR during hypertension-induced cardiac hypertrophy remains elusive. In this study, we found that heat shock transcription factor 2 (HSF2) ac

Challenges in Orphan Drug Development: Identification of Effective Therapy for Thyroid-Associated Ophthalmopathy.

Thyroid-associated ophthalmopathy (TAO), the ocular manifestation of Graves' disease, is a process in which orbital connective tissues and extraocular muscles undergo inflammation and remodeling. The condition seems to result from autoimmune responses to antigens shared by the thyroid and orbit. The thyrotropin receptor (TSHR), expressed at low levels in orbital tissues, is a leading candidate antigen. Recent evidence suggests that another protein, the insulin-like growth factor-I receptor (IGF-IR), is overexpressed in TAO, and antibodies against IGF-IR have been detected in patients with the

Expression patterns of T-type Cav3.2 channel and insulin-like growth factor-1 receptor in dorsal root ganglion neurons of mice after sciatic nerve axotomy.

Substantial evidence indicates that T-type Cav3.2 channel and insulin-like growth factor-1 (IGF-1) contribute to pain hypersensitivity within primary sensory nerves. A recent study suggested that activation of IGF-1 receptor (IGF-1R) could increase Cav3.2 channel currents and further contribute to inflammatory pain sensitivity. However, the expression patterns of Cav3.2 and IGF-1R and their colocalization in dorsal root ganglion (DRG) in chronic neuropathic pain condition remain unknown. In this study, we explored expression patterns of Cav3.2, IGF-1R and their colocalization, and whether phen

Effects of maternal undernutrition on lung growth and insulin-like growth factor system expression in rat offspring.

BACKGROUND: Maternal undernutrition may alter the development of the lung structure in rat offspring. METHODS: We investigated the effects of maternal undernutrition (50% rations of the control food intake) during the last week of gestation on the expression of the rat lung insulin-like growth factor (IGF) system in the postnatal period. RESULTS: Body weights of undernourished pregnant rats were significantly lower than those of control rats from gestational days 16 to 21. Rats subjected to intrauterine growth restriction (IUGR) exhibited significantly lower body weights and lower lung weigh

Computational framework to quantify synergistic ligand activity in insulin secretion and resistance pathways in type 2 diabetes.

Type 2 diabetes mellitus (T2DM) involves dysregulation of both insulin secretion and insulin resistance pathways. However, current therapies often target only one pathway, leading to limited effectiveness. We aimed to develop a computational framework to quantify synergistic ligand activity across both pathways, providing a foundation for multi-target therapeutic strategies. We built a synergy assessment framework targeting six key proteins: glucagon-like peptide-1 receptor (GLP1R) and kinesin family member 11 (KIF11) in the insulin secretion pathway; and insulin-like growth factor 1 receptor

Insulin-like growth factor-I (IGF-I) and IGF-I receptor (IGF-IR) immunoreactivity in normal and osteopetrotic (toothless, tl/tl) rat tibia.

Insulin-like growth factor-I (IGF-I) plays a major role in regulating cell growth. This study examined the immunohistochemical distribution of IGF-I and IGF-I receptor (IGF-IR) in tibias from normal and osteopetrotic (toothless, tl/tl) rats, following treatment with colony stimulating factor-1 (CSF-1). In normal rats, immunoreactivity for IGF-I and IGF-IR was detected in cells of the articular and epiphyseal cartilage, secondary ossification centres, zones of resting and proliferating chondrocytes and bone marrow. Bone marrow cells immunoreactive for IGF-I and IGF-IR were significantly reduced

Insulin-like and fibroblast growth factors and their receptors are differentially expressed in the oviducts of the common marmoset monkey (Callithrix jacchus) during the ovulatory cycle.

It is suggested that growth factors support the process of maturation and differentiation in the mammalian oviduct. Fibroblast growth factors (FGF) and insulin-like growth factors (IGF) are possible influences on these processes. The present study describes for the first time the expression of FGF-1 and -2 and their receptors as well as IGF-1 and -2 and the corresponding IGF receptor type 1 in the oviduct of the New World monkey Callithrix jacchus. Because of the limited RNA yields from oviducts, reverse transcription-polymerase chain reaction (RT-PCR) was performed to estimate expression leve

Regulation of human trophoblast migration and invasiveness.

The human placenta is an invasive structure in which highly proliferative, migratory, and invasive extravillous trophoblast (EVT) cells migrate and invade the uterus and its vasculature. Using in vitro propagated normal first-trimester EVT cells and immortalized EVT cells, which share all of the phenotypic and functional characteristics of the normal EVT cells, it has been shown that migration/invasion of human EVT cells is stringently regulated by many growth factors, their binding proteins, extracellular matrix (ECM) components, and some adhesion molecules in an autocrine/paracrine manner at

MicroRNAs link chronic inflammation in childhood to growth impairment and insulin-resistance.

MicroRNAs are involved in multiple pathophysiological networks and in the pathogenesis of a broad spectrum of human disorders, including cancer and inflammatory diseases. Impaired linear growth is encountered in children with chronic inflammatory conditions such as cystic fibrosis, inflammatory bowel diseases, juvenile idiopathic arthritis, celiac disease and in subjects born intrauterine growth restricted/small for gestational age. Children with inflammatory conditions may also be at risk of developing insulin resistance as a result of the inflammatory process and concurrent therapy. Chronic

Expression of messenger ribonucleic acids for fibroblast growth factors 7 and 10, hepatocyte growth factor, and insulin-like growth factors and their receptors in the neonatal ovine uterus.

In sheep, uterine development begins during fetal life but is only completed postnatally with proliferation and branching morphogenetic differentiation of the endometrial glandular epithelium (G) from the luminal epithelium (L) between birth or Postnatal Day (PND) 0 and PND 56. In other epithelial-mesenchymal organs, fibroblast growth factor (FGF)-7 and FGF-10, hepatocyte growth factor (HGF), and insulin-like growth factor (IGF)-I and IGF-II play essential roles in ductal branching morphogenesis. The interaction of IGF-1/IGF-1R and hydrogen sulfide on the proliferation of mouse primary vascular smooth muscle cells.

Hydrogen sulfide (H2S) is mostly produced by cystathionine-gamma-lyase (CSE) in vascular system and it inhibits the proliferation of vascular smooth muscle cells (SMCs). Insulin-like growth factor-1 (IGF-1), via its receptor (IGF-1R), exerts multiple physiological and pathophysiological effects on the vasculature, including stimulating SMC proliferation and migration, and inhibiting SMC apoptosis. Since H2S and IGF-1/IGF-1R have opposite effects on SMC proliferation, it becomes imperative to better understand the interaction of these two signaling mechanisms on SMC proliferation. SMCs isolated

An inhibitory function for JNK in the regulation of IGF-I signaling in breast cancer.

Insulin-like growth factor-I receptor (IGF-IR) is frequently overexpressed in a variety of cancer types. Since many breast tumors and cancer cell lines overexpress IGF-IR, we tested IGF-I effects on chemotherapy-treated breast cancer cells. IGF-I protects from chemotherapy-induced apoptosis, suggesting that overlapping signaling pathways modulate IGF-I and chemotherapy treatment outcomes. Taxol and other chemotherapy drugs induce c-Jun N-terminal kinase (JNK), a kinase that conveys cellular stress and death signals. Notably, in this paper we show that IGF-I alone induces a potent JNK response
