High cell density cultivation by anaerobic respiration.

BACKGROUND: Oxygen provision is a bottleneck in conventional aerobic high cell density culturing (HCDC) of bacteria due to the low O2 solubility in water. An alternative could be denitrification: anaerobic respiration using nitrogen oxides as terminal electron acceptors. Denitrification is attractive because NO3- is soluble in water, the end-product (N2) is harmless, and denitrification is widespread among bacteria, hence suitable organisms for most purposes can be found. The pH must be controlled by injection of an inorganic acid to compensate for the pH increase by NO3--consumption, resultin…

Camera-Based Respiration Monitoring of Unconstrained Rodents.

Animal research has always been crucial for various medical and scientific breakthroughs, providing information on disease mechanisms, genetic predisposition to diseases, and pharmacological treatment. However, the use of animals in medical research is a source of great controversy and ongoing debate in modern science. To ensure a high level of bioethics, new guidelines have been adopted by the EU, implementing the 3R principles to replace animal testing wherever possible, reduce the number of animals per experiment, and refine procedures to minimize stress and pain. Supporting these guideline

Regulation of Mitochondrial Respiration by Hydrogen Sulfide.

Hydrogen sulfide (H2S), the third gasotransmitter, has positive roles in animals and plants. Mitochondria are the source and the target of H … d cycle (TCA) and the mitochondrial respiratory chain. A study of the roles of H2S in mitochondrial respiration in plants to elucidate the botanical function of H2S in plants would be highly desirable.

[Elective caesarean: impact of evolution neonatal respiration].

The number of Caesarean births has increased worldwide. Respiratory distress syndrome is associated with caesarean delivery, especially in the absence of labor. During the latter stages of pregnancy physiologic changes occur that are accelerated with the onset of labor, which is accompanied by changes in hormones and mediators in the mother and fetus. An acceleration in the evacuation of lung liquid is held in this period, largely dependent sodium channels sensitive to amiloride than are found in the alveolar epithelium. The failure of these mechanisms can lead to newborn severe respiratory di

Central sleep apnea and Cheyne-Stokes respiration.

Cheyne-Stokes respiration with central sleep apnea (CSR-CSA) is a form of periodic breathing, commonly observed in patients with heart failure (HF), in which central apneas alternate with hyperpneas that have a waxing-waning pattern of tidal volume. Uniform criteria by which to diagnose a clinically significant degree of CSR-CSA have yet to be established. CSR-CSA is caused by respiratory control system instability characterized by a tendency to hyperventilate. Central apnea occurs when Pa(CO(2)) falls below the threshold for apnea during sleep due to ventilatory overshoot. Patients with CSR-C…

[Positive pressure ventilation in Cheyne-Stokes respiration].

Cheyne-Stokes respiration (CSR) is found in patients with chronic left ventricular failure and associated with a reduced prognosis. Continuous positive airway pressure (CPAP) improves the survival rate. In this retrospective study we report on the effect of different positive pressure ventilation modes in CSR. The observation period lasted from 1995 - 1999. Inclusion criteria was CSR with a respiratory disturbance index > 10/h whereas > 50 % of the events had to be central and/or mixed. In each patient a 4 week lasting intervention with each of CPAP, Bilevel CPAP in spontan mode (i.e. BiPAP S …

Upper airway function and dysfunction in respiration.

1. The upper airway not only provides a passage for air to be breathed in and out of the lungs, but it also heats, humidifies and filters the air and is involved in cough, swallowing and speech. 2. The complex muscle structure of the upper airway that produces speech and swallowing in humans also modulates respiratory airflow throughout the respiratory cycle, but is vulnerable to functional problems that may compromise respiration. 3. Even in normals, there is some collapse of the upper airway and increased upper airway resistance during sleep. 4. A substantial proportion of people suffer from…

Respiration patterns during feeding in Rett syndrome.

Feeding problems are common in Rett syndrome in which there are characteristic oropharyngeal abnormalities. This study investigated the ways in which individuals regulated their respiration accordingly, and how this affected their overall feeding ability. Respiration during feeding was studied in 28 individuals, recording nasal airflow, chest and abdominal movements, and swallow sounds. Time to first swallow was defined as that between introduction of liquid/solid on a spoon and the first swallow. Six individuals also had videofluoroscopy with simultaneous respiration monitoring. Results indic…

Rest and respiration during obstructive sleep apnoea.

Obstructive sleep apnoea (OSA) is a condition in which the airway is obstructed during sleep. Although often harmless, OSA can produce significant morbidity. This paper describes the condition and how it is assessed in a respiratory diseases unit. A case study is given that highlights the far-reaching physical, social and psychological sequelae to this condition.

Periodic respiration in patients with heart failure.

Patients with heart failure are known to demonstrate periodic respiration (PR) during sleep. The factors causing PR are not well known. We therefore studied 20 patients (aged 18-66 years) with idiopathic dilated cardiomyopathy. Full-night polysomnography and evaluation of respiration and transcutaneous oxygen saturation were performed. Hypercapnic ventilatory response (HCVR) was evaluated during daytime. The patients showed PR for 25 +/- 26% (mean +/- standard deviation) of total sleep time. During PR, oxygen desaturated 7.1 +/- 4.6%. Sleep was impaired. HCVR was normal. Oxygen desaturation du…

Sleep and respiration under acute hypobaric hypoxia.

The purpose of this study was to elucidate the effects of acute hypobaric hypoxia on nocturnal sleep architecture and respiratory responses in a hypobaric simulator. Five healthy young males (19-23 years old) were recruited to sleep for 8 h at sea level and at simulated altitudes of 1,500, 3,000, and 4,000 m in the simulator (61.2 m3, 20 degrees C, and 60% RH). Each experimental run was separated by at least 3 d. Standard polysomnograph, respiration, and arterial oxygen saturation (SaO2) during sleep were observed. 1) SaO2 decreased significantly with increasing altitude. At 4,000 m, SaO2 show…

Effect of chlorpromazine on bovine sperm respiration.

Chlorpromazine (CPZ), an antipsychosis drug, causes the in vitro inhibition of sperm motility in various species, but the effect of this drug on sperm respiration is not well studied. This study was undertaken to evaluate the effect of this drug on ejaculated bovine sperm endogenous respiration. CPZ acted as a stimulator and an inhibitor of respiration at 0.1 and 0.75 mM CPZ, in the respiration medium, respectively. CPZ caused the release of sperm protein in the supernatant in a dose-dependent manner and the lactate dehydrogenase activity has also been demonstrated in the supernatant. Succinat…

Buspirone, an anxiolytic drug that stimulates respiration.

The recently released drug buspirone is an anxiolytic agent that appears not to have the sedating effects of barbiturates and benzodiazepine … a change of slope or shape of the CO2 response curve. In contrast, diazepam led to a depression of respiration and a shift of the apneic threshold to a higher PCO2. The findings indicate that buspirone does not have the typical neural respiratory depressant actions of diazepam but instead stimulates respiration. Although the findings will need to be shown to …

Cheyne-Stokes respiration revisited: controversies and implications.

Investigation of the periodic crescendo-decrescendo alterations in tidal volume in Cheyne-Stokes respiration (CSR) has provided remarkable insight into the physiology of respiratory control. Many patients with periodic breathing have both cardiac and neurologic disease. Considerable controversy has surrounded determination of the relative importance of cardiac and neurologic mechanisms in the genesis of this breathing abnormality. Several investigators have considered the respiratory center as a chemostat model with three basic components: the controller system (chemoreceptors), the controlled…

Carotid sinus baroreceptor reflex control of respiration.

We have studied the effect of the carotid sinus baroreceptor reflex on respiration in 10 vagotomized, spontaneously breathing, pentobarbital anesthetized dogs. The carotid body chemoreceptor reflex response was eliminated by surgically excluding the carotid bodies from the carotid sinus baroreceptor area. Steady state frequency, tidal volume, and minute ventilation were measured after 25 mm Hg step changes in intrasinus pressure between 50 and 200 mm Hg. Over this range, the step decreases in intrasinus pressure caused concomitant increases in mean arterial pressure from 86 to 182 mm Hg. All o…

Restricted nasal respiration, influence on facial growth.

Used as models on restricted nasal respiration, 11 individuals with a unilateral inborn occlusion of the nose were examined with regard to growth of the mid-facial skeleton. The preference to oral breathing, also a fairly constant feature in patients with chronic or allergic rhinitis with hyperplasia of the pharynx tonsil, appeared to result in a significant underdevelopment of the maxilla in the saggittal plane.
