Zygomycosis of the Scalp Caused by Rhizopus oryzae Presenting as Kerion in an Immunocompetent Child.

Deep mycosis of the scalp caused by Rhizopus oryzae mimicking kerion is rare. Herein, we report a case of such infection in a 5-year-old immunocompetent boy who presented with multiple painful boggy swellings with discharging sinuses on the scalp of 4 months' duration. Purulent discharge from the swelling cultured on Sabouraud's dextrose agar yielded R. oryzae species which was confirmed by molecular analysis by polymerase chain reaction. The child was managed with parenteral liposomal amphotericin-B which helped in clearance of infection.

Lack of evidence of endosymbiotic toxin-producing bacteria in clinical Rhizopus isolates.

Infections by Rhizopus spp. account for about 90% of zygomycoses, many of which are lethal in immunocompromised patients. We recently noted that several strains of Rhizopus microsporus harbour rare bacterial endosymbionts (Burkholderia sp.) for the production of 'mycotoxins', which might play a role as virulence factors in human Rhizopus infections. In this study eight clinical Rhizopus spp. isolates have been investigated for the presence of toxin-producing bacterial endosymbionts. By metabolomic data, PCR targeting bacterial 16S rDNA and microscopic investigations with fluorescence dyes we p…

Peritonitis due to Rhizopus in a patient undergoing continuous ambulatory peritoneal dialysis.

A 61-year-old man who had end-stage renal disease secondary to diabetes mellitus and hypertension developed peritonitis due to infection with Rhizopus as a complication of receiving continuous ambulatory peritoneal dialysis (CAPD). At the onset of infection, the patient was neither acidemic nor hyperglycemic; in addition, deferoxamine had not been administered and Elastoplast dressings had not been applied. The infection occurred after the technique for disinfection of the catheter used for CAPD had been changed. The catheter was removed, and therapy with amphotericin B was initiated. Although…

Fungus Rhizopus arrhizus promotes MASLD-associated hepatocellular carcinoma through generating kynurenic acid.

Fungi are crucial components of the human gut microbiota. Here, we investigated the role of fungi in metabolic dysfunction-associated steatotic liver disease-associated hepatocellular carcinoma (MASLD-HCC). Rhizopus arrhizus was the top enriched fungus across the progression from MASLD to MASLD-HCC, accelerating tumorigenesis in multiple mouse models. R. arrhizus-conditioned medium increased cell proliferation while reducing cell cycle arrest and apoptosis in human MASLD-HCC cells. Kynurenic acid (KYNA) was identified as the crucial metabolite generated from R. arrhizus, and its upregulation w…

Pseudomonas aeruginosa inhibits Rhizopus microsporus germination through sequestration of free environmental iron.

Rhizopus spp are the most common etiological agents of mucormycosis, causing over 90% mortality in disseminated infection. Key to pathogenesis is the ability of fungal spores to swell, germinate, and penetrate surrounding tissues. Antibiotic treatment in at-risk patients increases the probability of the patient developing mucormycosis, suggesting that bacteria have the potential to control the growth of the fungus. However, research into polymicrobial relationships involving Rhizopus spp has not been extensively explored. Here we show that co-culturing Rhizopus microsporus and Pseudomonas aeru…

Primary Oral Mucormycosis Due to Rhizopus microsporus after Allogeneic Stem Cell Transplantation.

We herein report a rare case of oral mucormycosis following allogeneic hematopoietic stem cell transplantation. Oral mucormycosis due to Rhizopus microsporus manifested as localized left buccal mucositis with a 1-cm black focus before neutrophil recovery. Combination therapy with liposomal amphotericin B was initiated and surgical debridement was performed; however, the patient died due to progressive generalized mucormycosis. Considerable attention needs to be paid to the diagnosis and management of oral mucormycosis in post-transplant patients, thereby suggesting the importance of fully unde…

Basilar artery occlusion after multifactor coagulopathy including Rhizopus oryzae infection in burns.

The authors report a case of a 29-year-old male patient with severe burn injuries (54% TBSA) and inhalation injury. He developed a candidemia and a cutaneous zygomycotic superinfection with Rhizopus oryzae while he received burn intensive care. Despite aggressive surgery, sepsis persisted, and therapy was limited by uncontrollable coagulopathy and catecholamine refractory shock after 15 days. Autopsy revealed a thromboembolic occlusion of the basilar artery that resulted in liquefactive necrosis of the basal brain tissue and the brain stem. Because cerebral vessel occlusions after burn injurie…

Cutaneous infection with Rhizopus oryzae and Aspergillus niger following bone marrow transplantation.

We describe a case of mixed Rhizopus oryzae and Aspergillus niger infection in a 38 year-old woman who had undergone allogeneic bone marrow transplantation for multiple myeloma. The fungi were isolated from a necrotic ulcer under a plaster cast stabilizing a pathological fracture of the left humerus. We emphasize the necessity for frequent inspection of any covered area of skin and the use of sterile dressings as far as possible in neutropenic patients.

Assessment of Rhizopus stolonifer tolerance to methotrexate: morphological responses and biodegradation limitations.

This study evaluates the tolerance of Rhizopus stolonifer to methotrexate (MTX) and its capacity to remove and transform this antifolate compound, highlighting both morphological responses and biodegradation limitations. The xenobiotic affected fungal differentiation more strongly than vegetative growth: although radial expansion remained unchanged up to 500 ppm, sporulation, hyphal integrity, and cellular organization were progressively impaired. Spore germination decreased sharply at low concentrations, yielding an average half maximal inhibitory concentration (IC₅₀) of 138 ppm. In pellet sy…

Identification and Validation of Rhizopus-Specific Glucoamylase as a Novel Biomarker for Diagnosing Mucormycosis.

BACKGROUND: Mucormycosis is a life-threatening invasive fungal infection. Current diagnosis relies on insensitive techniques with prolonged turnaround times, underscoring the need for rapid noninvasive diagnostic tools. The current study aimed to identify the antigens of Rhizopus arrhizus that can facilitate serodiagnosis of mucormycosis and assess their diagnostic potential in patients. METHODS: This was a 2-stage systematic study combining antigen discovery and clinical validation. Using immunoproteomics, we identified immunoreactive protein spots in R. arrhizus secretory extract, using poo…
