Considerable interest has been made in developing nonculture approaches to diagnosing fungal infections, as early diagnosis may guide appropriate treatment and prevent mortality. These approaches include detection of specific host immune responses to fungal antigens, and detection of specific macromolecular antigens using immunologic reagents. The detection of a specific host antibody response is attractive because such tests can be performed rapidly and do not require invasive sampling procedures. However, presence of host antibodies does not always correlate with presence of invasive disease

An indirect immunoreaction is proposed to demonstrate anti-Borrelia antibodies in the serum of patients with Lyme disease (LD). The binding of specific IgG antibodies to dried smears of Borrelia antigen (strain Ip21) without fixation is visualized by subsequent incubation with peroxidase-conjugated antibodies to human IgG, followed by incubation with H2O2 and 3,3-diaminobenzidine tetrahydrochloride as a chromogenic substrate. This test is easy-to-perform and readable by a simple routine light microscope. It requires no special devices (a fluorescence microscope, a photometer, etc.) and may rea

This article addresses current issues regarding serologic testing for selected STDs, particularly those for which serologic testing has been useful for diagnosis, monitoring therapy, or seroepidemiology. For each selected pathogen, the authors discuss the strengths and weaknesses of the serologic tests as diagnostic assays, as measures of disease incidence and relevance, and as prospective markers identifying risk alterations in a given population. Also, for some conditions such as syphilis, the usefulness of serology as a measure of therapy or success of disease intervention is briefly outlin…

Sera from 44 patients with clinically suspected pulmonary aspergillosis (mainly of an allergic type) were examined for antibodies to Aspergillus fumigatus using an enzyme-linked immunosorbent assay (ELISA) and counter-immunoelectrophoresis (CIE). Of the sera, 15 were considered to contain Aspergillus antibodies using ELISA; 11 of these also contained precipitins by CIE. In no instance was a CIE-positive serum negative by ELISA. The serum of 1 patient with autopsy-verified invasive pulmonary aspergillosis was negative in both tests. Protein-enriched antigens derived by ammonium sulphate precipi

Circulating anticoagulant (CA) and particularly lupus anticoagulant are commonly associated with biological false-positive tests for syphilis (BFP-STS) in patients with collagen diseases. CA was presently found in the sera of 8 out of 30 subjects with chronic BFP-STS without collagen or autoimmune diseases. It was not found in any of the 21 patients with various stages of syphilis. In 5 out of 21 elderly subjects (age greater than 70 years) in whom a positive BFP-STS was detected, there was no CA. It is concluded that the association between CA and BFP-STS is found only in patients with collag

In an enzyme-linked immunosorbent assay test for malaria antibodies, antibodies to Plasmodium vivax and P. falciparum in man are detected using a crude antigen prepared from the simian malaria parasite P. knowlesi. The test may be suitable for epidemiological studies.

BACKGROUND: Previous studies have shown low frequencies of seroreactivity to HIV diagnostic assays for infected infants treated with antiretroviral therapy (ART) early in infection.
METHODS: Fifty-eight HIV-infected infants treated with ART at a median age of 1.9 months (range: 0.2-5.4) for up to 4 years of life were assessed for seroreactivity to 4 routinely used HIV clinical immunoassays (IA): Second-generation (2ndG) IA and 2 rapid diagnostic tests (RDT), based on third-generation principles, measuring antibody only and a fourth-generation (4thG) antigen/antibody IA. HIV Western blot assay

The indirect hemagglutination (IHA) and immunoelectrophoresis (IEP) tests were used for diagnosis and follow-up evaluation of 17 patients with alveolar hydatid disease caused by Echinococcus multilocularis. Follow-up periods ranged from 2 to 22 years. At the time of diagnosis 16 (94%) patients' sera gave IHA titers greater than or equal to 1:128. Serum specimens from 13 patients were examined by IEP; nine (69%) revealed the arc 5, and three of the four arc 5-negative sera revealed one or more non-characterized bands. Titers declined markedly during the first year following radical surgical res

The case report presents a male patient in his mid-60s with a history of hypertension, benign prostatic hyperplasia and chronic kidney disease (CKD). He presented with gradually increasing serum creatinine levels and hyperglobulinemia, leading to suspicion of multiple myeloma. However, subsequent testing revealed features consistent with systemic lupus erythematosus (SLE) and IgG4-related kidney disease (IgG4-RKD). The patient's laboratory results included anaemia, positive ANA and anti-dsDNA and elevated serum IgG4 levels. A kidney biopsy showed extensive interstitial fibrosis, plasma cell in

The objective of the current study was to determine whether detection of Canine distemper virus (CDV)-specific serum antibodies correlates with resistance to challenge with virulent virus. Virus neutralization (VN) assay results were compared with resistance to viral challenge in 2 unvaccinated Beagle puppies, 9 unvaccinated Beagle dogs (4.4-7.2 years of age), and 9 vaccinated Beagle dogs (3.7-4.7 years of age). Eight of 9 (89%) unvaccinated adult dogs exhibited clinical signs after virus challenge, and 1 (13%) dog died. As compared to adult dogs, the 2 unvaccinated puppies developed more seve

Serologic tests have been widely used for the diagnosis of asymptomatic visceral leishmaniasis. This study evaluated five serologic tests used for the diagnosis of asymptomatic infection: enzyme-linked immunosorbent assay (ELISA) using promastigote antigen (ELISAp), ELISA using recombinant K39 (ELISA rK39), and K26 (ELISA rK26) antigens, an indirect immunofluorescence test using Leishmania (Leishmania) amazonensis promastigote antigen (IIFT), and an immunochromatographic test using rK39 antigen (TRALd). As a reference regarding the performance of the tests, patients with classic visceral leish…

OBJECTIVE: To estimate sensitivity and specificity of 4 commonly used brucellosis screening tests in cattle and domestic water buffalo of Trinidad, and to compare test parameter estimates between cattle and water buffalo.
ANIMALS: 391 cattle and 381 water buffalo.
PROCEDURE: 4 Brucella-infected herds (2 cattle and 2 water buffalo) and 4 herds (2 of each species) considered to be brucellosis-free were selected. A minimum of 100 animals, or all animals > 1 year of age, were tested from each herd. Serum samples were evaluated for Brucella-specific antibodies by use of standard plate agglutinati
