The transcription factor SbWRKY6 confers cadmium tolerance via activating SbPLAC8-17 expression in sorghum.

Cadmium (Cd) is a widespread environmental pollutant that severely threatens crop productivity and food safety. However, the regulatory mechanisms underlying Cd detoxification and tolerance in sorghum remain largely elusive. Herein, we functionally characterized SbWRKY6, a Cd‑induced WRKY transcription factor that localizes to the nucleus and functions as a transcriptional activator. Stable overexpression of SbWRKY6 significantly enhanced Cd tolerance in sorghum, as evidenced by improved growth performance, mitigated oxidative damage, and decreased Cd concentration in plant tissues, whereas si…

Hyaluronic acid-based hydrogel for cadmium removal and enhanced sorghum growth under cadmium stress.

Cadmium (Cd) is a highly toxic heavy metal that poses a threat to environmental and human health. The hydrophilic and modifiable properties … l absorbed Cd initially, thereby minimizing its biological accessibility and plant root absorption, sorghum seedlings treated with the HDT hydrogel exhibited healthier growth patterns and lower internal Cd concentrations compared to untreated controls in Cd-contaminated environments, ultimately contributing to an approximate 53.2 % total Cd removal. There …

Characterization of grain carotenoids in global sorghum germplasm to guide genomics-assisted breeding strategies.

BACKGROUND: Crop biofortification is a successful strategy to ameliorate Vitamin A deficiency. Sorghum is a good candidate for vitamin A biofortification, as it is a staple food in regions with high prevalence of vitamin A deficiency. β-carotene-the main provitamin A carotenoid-is below the target concentration in sorghum grain, therefore biofortification breeding is required. Previous studies found evidence that sorghum carotenoid variation is oligogenic, suggesting that marker-assisted selection can be an appropriate biofortification method. However, we hypothesize that sorghum carotenoids h…

Evaluation of A3 cytoplasmic male sterile forage sorghum lines for resistance to sugarcane aphid.

Three known sugarcane aphid-resistant pollinator parents were sterilized in A3 cytoplasmic male sterility and were confirmed in this study to be resistant to sugarcane aphid allowing for the development of sugarcane aphid-resistant forage hybrids. We utilized A3 cytoplasmic male sterility and converted known sugarcane aphid-resistant sorghum TX 2783, and newly released R. LBK1 (Reg. No. GP-865, PI 687244) and R. LBK2 (Reg. No. GP-866, PI 687245) into A3 sterility to determine if the sterile counterparts would also equally express tolerance and or antibiosis to sugarcane aphid. Free-choice flat…

In silico ligand binding studies of cyanogenic β-glucosidase, dhurrinase-2 from Sorghum bicolor.

Dhurrinase, a cyanogenic β-glucosidase from Sorghum bicolor is the key enzyme responsible for the hydrolysis of dhurrin to produce toxic hydrogen cyanide, as a part of plant defence mechanism. Dhurrinase 1 (SbDhr1) and dhurrinase 2 (SbDhr2), two isozymes have been isolated and characterized from S. bicolor. However, there is no information in the literature about the three dimensional (3D) structure of SbDhr2 and molecular interactions involved between the protein and ligand. In this study, the three dimensional structure of SbDhr2 was built based on homology modeling by using the X-ray crysta…

Pan-NLRome analysis uncovers genetic diversity and evolutionary dynamics among rice, maize and sorghum.

Nucleotide-binding leucine-rich repeat (NLR) genes constitute one of the largest families of plant immune receptors and are central to crop disease resistance. Rice (Oryza sativa L.), maize (Zea mays L.), and sorghum (Sorghum bicolor (L.) Moench) are major crops underpinning global food security, and enhancing their immunity is critical for yield stability. Here, we conducted a comprehensive analysis of NLR repertoires across 75 genomes from these three grass crops, identifying 24,944 genes that exhibit extensive intra- and interspecific variation. The crops share broadly comparable domain arc…

Disruption of Fatty Acid Desaturase Gene Alters Aphid Resistance and Volatile Emissions in Sorghum.

Jasmonic acid (JA)-deficient msd3 sorghum mutants exhibit contrasting resistance phenotypes against sugarcane aphids (SCA; Melanaphis sacchari), which promotes aphid settling at early time points while providing antibiosis-mediated resistance at later stages. Here we investigated the physiological, hormonal, and metabolic mechanisms underlying msd3-mediated resistance to SCA. Contrary to the widely reported growth-defence trade-off, msd3 plants maintained biomass comparable to uninfested controls, while wild-type (WT) plants showed significant reduction in biomass following SCA feeding. Gas ex…

A Grass-Specific Structural Myosin VIII Regulates Protoxylem Development and Hydraulic Conductance in Sorghum.

Sorghum (Sorghum bicolor), a C4 grass adapted to hot semi-arid environments, depends on reinforced xylem vessels to maintain hydraulic conductance under high evaporative demand. During xylem differentiation, coordinated microtubule and actin dynamics guide secondary cell wall (SCW) deposition; however, whether actin-based motors regulate vascular architecture and hydraulic performance has remained unknown. Here, we identify HEAT-SENSITIVE 1 (HS1), a previously uncharacterized myosin VIII, as a central regulator of protoxylem integrity and water transport in sorghum. The hs1 mutant exhibited se…

Colonic and systemic effects of extruded whole-grain sorghum consumption in growing Wistar rats.

Colonic effects of extruded whole-grain sorghum diets were evaluated using a model of growing rats. In all, twenty-four male Wistar rats were fed control (C), extruded white sorghum (EWS) or red sorghum (ERS). Consumption of sorghum diets showed satiety properties, with reduction of caecal pH, and lower activity of β-glucosidase and β-glucuronidase enzymes. Decreased copper zinc superoxide dismutase and manganese superoxide dismutase and increased catalase and glutathione peroxidase levels were observed in colonic mucosa. The induction of antioxidant enzymes occurred through the activation of …

Decreased egg production in commercial laying hens fed zearalenone- and deoxynivalenol-contaminated grain sorghum.

In an experiment to determine the effect of corn vs. grain sorghum on performance of laying hens, egg production decreased significantly in the grain sorghum-fed hens in each of two trials starting 24 weeks after the trials began. Necropsy of chickens fed both diets revealed buccal ulceration at the ventral aspect of the oral cavity and squamous metaplasia of the esophageal glands and submaxillary salivary glands. Lesions were much more severe in the sorghum-fed birds than in the corn-fed birds. Analysis of the grain sorghum and corn revealed the presence of mycotoxins. Zearalenone and deoxyni…

Advancing provitamin A biofortification in sorghum: from allele mining and marker development to prebreeding and breeding.

Allele mining for provitamin A biofortification in sorghum is advanced with new pre-bred germplasm and markers that are trait-predictive in diverse germplasm and breeding programs. Provitamin A biofortification of the staple cereal crop sorghum (Sorghum bicolor [L.] Moench) could reduce vitamin A deficiency in low-income countries of sub-Saharan Africa. To advance mining of alleles that confer high provitamin A from genebank accessions, we developed prebreeding germplasm and trait-predictive markers, and established the utility of these markers in a range of prebreeding and breeding materials.…

Impacts of combining aphid and headworm management on natural enemy conservation and grain sorghum production.

Grain sorghum (Sorghum bicolor L.) of the southern United States is attacked by numerous insect pests. These include lepidopteran larvae which infest sorghum heads (headworms) and have historically been managed by insecticide applications. The aphid pest Melanaphis sorghi (Theobald) has more recently emerged as an economic pest of sorghum leaves. Management for this aphid has included host plant resistance, insecticide applications, and suppression from insect natural enemies. It is currently unclear if the management practices for M. sorghi or headworms interferes with suppression of the othe…

Molecular insights into the mutualism that induces iron deficiency tolerance in sorghum inoculated with Trichoderma harzianum.

Iron (Fe) deficiency is a common mineral stress in plants, including sorghum. Although the soil fungus Trichoderma harzianum has been shown to mitigate Fe deficiency in some circumstances, neither the range nor mechanism(s) of this process are well understood. In this study, high pH-induced Fe deficiency in sorghum cultivated in pots with natural field soil exhibited a significant decrease in biomass, photosynthetic rate, transpiration rate, stomatal conductance, water use efficiency, and Fe-uptake in both the root and shoot. However, the establishment of T. harzianum colonization in roots of …

Revealing Differential Expression of Phytohormones in Sorghum in Response to Aphid Attack Using the Metabolomics Approach.

Sorghum (Sorghum bicolor) is an important multipurpose crop grown worldwide, but like many other crops, it is often threatened by insect pests. Sugarcane aphid (SCA, Melanaphis sacchari Zehntner), for example, is one of the most severe pests in sorghum, which causes plant damage and yield loss. The main objective of this study was to assess the effect of phytohormones on host plant resistance to aphid attack. Two sorghum genotypes, BTx623 (susceptible) and Tx2783 (resistant), were selected for a comparative analysis of differential expression of a group of phytohormones in response to aphid in…

Genome-wide identification and classification of Lipoxygenase gene family and their roles in sorghum-aphid interaction.

This report shows detailed characterization of LOX gene family in sorghum and provides new insight of sorghum LOX genes in genetic structure and their roles in plant response to infestation by sugarcane aphids. Lipoxygenases (LOXs) are monomeric, nonheme iron-containing dioxygenases that initiate the fatty acid oxidation pathway creating oxylipins and plant hormone jasmonate both have a key role in plant development and defense. To date, a comprehensive and systematic analysis of sorghum LOXs is still deficient. Thus, we performed a genome-wide analysis of the sorghum LOXs genome and identifie…
