Where metabolism and virulence meet-sulfoxide reductases as determinants of bacterial survival.

Bacterial metabolism is increasingly recognised as a driver of virulence in pathogens, and this includes the reduction of sulfoxides, which has traditionally been linked to anaerobic energy generation and maintenance of cellular proteins. Sulfoxide reduction is catalysed by two major enzyme groups, the Msr peptide methionine sulfoxide reductases that repair damaged proteins, and several types of mononuclear molybdenum enzymes that either convert small molecular S- and N-oxides or repair damaged proteins. Where these enzymes support virulence, they appear to be regulated by exposure to oxidisin…

Central Carbon Metabolism in Candida albicans Biofilms Is Altered by Dimethyl Sulfoxide.

The effect of dimethyl sulfoxide (DMSO) on fungal metabolism has not been well studied. This study aimed to evaluate, by metabolomics, the impact of DMSO on the central carbon metabolism of Candida albicans. Biofilms of C. albicans SC5314 were grown on paper discs, using minimum mineral (MM) medium, in a dynamic continuous flow system. The two experimental conditions were control and 0.03% DMSO (v/v). After 72 h of incubation (37 °C), the biofilms were collected and the metabolites were extracted. The extracted metabolites were subjected to gas chromatography-mass spectrometry (GC/MS). The exp

Range of therapeutic prothipendyl and prothipendyl sulfoxide concentrations in clinical blood samples.

Due to a lack of reference blood concentrations in the literature, the forensic evaluation of prothipendyl findings in blood samples is difficult. Interpretations with regard to the assessment of blood concentrations as well as an estimation of the ingested prothipendyl amounts were often vague. To describe a concentration range in clinical samples, prothipendyl and prothipendyl sulfoxide concentrations were determined in serum samples of 50 psychiatric patients receiving 40 mg, 80 mg, or 160 mg doses of prothipendyl. The analyses of prothipendyl and prothipendyl sulfoxide were carried out usi

Detection and localization of methionine sulfoxide residues of specific proteins in brain tissue.

Methionine sulfoxide is a common posttranslational oxidative modification that can alter protein function. Vulnerability of specific proteins to methionine oxidation varies and depends on their structure. In the current study, detection of methionine sulfoxide in intact proteins is mediated by novel anti-methionine sulfoxide antibody that resulted in the identification of three major methionine sulfoxide-proteins in brain: bisphosphate aldolase A and C, α and β subunits of hemoglobin, and serum albumin. The locations of the methionine sulfoxide residues were determined by massspectrometry anal

Dexrazoxane: new indication. Anthracycline extravasation: continue using dimethyl sulfoxide.

1) Anthracycline extravasation can provoke extensive tissue necrosis, sometimes with serious consequences. Topical dimethylsulfoxide (DMSO) is the main antidote known to prevent this necrosis. It is used off-licence in France, based on the results of non-comparative trials. Among nearly 150 patients treated with dimethylsulfoxide, only one required surgery and about 10% of patients had sequelae; 2) A product based on dexrazoxane, an iron chelator, also approved to prevent anthracycline cardiotoxicity, has now been authorized for intravenous treatment of anthracycline extravasation; 3) Clinical

Inhibition of the sulfoxidation of omeprazole by ketoconazole in poor and extensive metabolizers of S-mephenytoin.

BACKGROUND: The metabolism of omeprazole includes hydroxylation catalyzed by CYP2C19 and, to a minor extent, sulfoxidation, presumably by CYP3A4. Sulfoxidation may be the predominant pathway in individuals devoid of the genetically determined CYP2C19 activity. Ketoconazole is a known CYP3A4 inhibitor in daily doses from 200 to 400 mg. In this study ketoconazole was used as a probe to investigate the extent to which CYP3A4 is involved in omeprazole metabolism in vivo. METHODS: A single oral 20 mg dose of omeprazole before and after four daily doses of 200, 100, or 50 mg ketoconazole was given

Vasomotor effects of dimethyl sulfoxide on cat cerebral arteries in vitro and in vivo.

We studied the direct vascular effects of dimethyl sulfoxide (DMSO) in isolated middle cerebral arteries and on pial arteriolar caliber after subarachnoid perivascular microinjection in chloralose-anesthetized cats, and on brain retraction in cats given DMSO intravenously. DMSO did not constrict isolated cerebral arteries at any of the concentrations studied (10(-10) to 4 X 10(-1) M). In middle cerebral arteries precontracted with potassium, 5-hydroxytryptamine, prostaglandin F2 alpha, or with mechanically raised tone, DMSO at concentrations of 10(-10) to 10(-2) M had no significant effects; a

Radioimmunoassay for the sulfoxide metabolite of trifluoperazine and its application to a kinetic study in humans.

Antibodies were produced in rabbits immunized with 10-[[3-[4-(2-carboxyethyl)-1-piperazinyl]-propyl]]-2 -trifluoromethyl-10H-phenothiazine sulfoxide-bovine serum albumin conjugate. The subsequently developed radioimmunoassay (RIA) procedure enables, for the first time, the quantitation of the sulfoxide metabolite of trifluoperazine in the plasma of humans after administration of therapeutic doses of trifluoperazine, in which 60 pg of the sulfoxide metabolite in 200 microL of plasma can be measured with a CV of less than 3%. Similar results were obtained by this assay with or without a benzene

Reduction of protein excretion by dimethyl sulfoxide in rats with passive Heymann nephritis.

Passive Heymann nephritis, a model of immune complex nephritis, was produced in rats by injection of rabbit antibrush border membrane vesicle antibodies to examine the effect of treatment of epimembranous glomerulonephritis with dimethyl sulfoxide. Administration of DMSO twice a day, 5 days a week for 4 weeks significantly reduced protein excretion in the autologous phase of the model. This beneficial effect occurred in animals in which treatment was started a day after administration of the antibody and persisted for 4 weeks after treatment was discontinued. Serum triglyceride concentrations

Reconsidering the Use of Dimethyl Sulfoxide for Xenobiotic-Gut Microbiota Interaction Studies.

INTRODUCTION: Gut microbiota are vulnerable to foreign chemicals (xenobiotics) including pharmaceuticals, environmental pollutants, and dietary contaminants such as aflatoxin B1 (AFB1) and fumonisin B1 (FB1). Assessing the effect of these xenobiotics in the laboratory requires their dissolution in a solvent vehicle, such as dimethyl sulfoxide (DMSO). While DMSO is typically used at low concentrations under the assumption of neutrality, its independent impact on microbial dynamics is a potential experimental confounder that has not been fully explored. METHODS: Human fecal microbiota were cult

Investigation of the effect of dimethyl sulfoxide on growth and biofilm formation of Pseudomonas aeruginosa.

BACKGROUND AND OBJECTIVES: The antimicrobial resistance of Pseudomonas aeruginosa bacteria limits the spectrum of effective antibiotics. Considerable focus has been placed on the identification of more contemporary and cost-effective antimicrobial drugs. In this study, the antibacterial properties of a commonly used solvent, dimethyl sulfoxide (DMSO), against P. aeruginosa were investigated. MATERIALS AND METHODS: The microtiter broth dilution technique was employed to establish the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of DMSO. The solvent's impa

Intraventricular dimethyl sulfoxide (DMSO) induces hydrocephalus in a dose-dependent pattern.

INTRODUCTION: Dimethyl sulfoxide (DMSO), a widely utilized solvent in the medical industry, has been associated with various adverse effects, even at low concentrations, including damage to mitochondrial integrity, altered membrane potentials, caspase activation, and apoptosis. Notably, therapeutic molecules for central nervous system treatments, such as embolic agents or some chemotherapy drugs that are dissolved in DMSO, have been associated with hydrocephalus as a secondary complication. Our study investigated the potential adverse effects of DMSO on the brain, specifically focusing on the

Synthesis of homochiral sulfanyl- and sulfoxide-substituted naphthyltriazoles and study of the conformational stability.

The preparation of a series of novel homochiral atropisomeric sulfanyl- and sulfoxide-substituted naphthyltriazoles is described. The triazolization methodology used presents a new way towards novel and highly stable 1,2,3-triazole-based atropisomers, and introduces a new and complementary synthetic pathway towards 4-sulfanyl substituted 1,2,3-triazoles. Starting from sulfanyl-substituted naphthyl ketones, enantiopure amines, and 4-nitrophenyl azide, a collection of 16 sulfanyl-substituted naphthyltriazoles were obtained via the triazolization reaction in which the homochiral diastereomers are

In Vitro Bioavailability of the Hydrocarbon Fractions of Dimethyl Sulfoxide Extracts of Petroleum Substances.

Determining the in vitro bioavailable concentration is a critical, yet unmet need to refine in vitro-to-in vivo extrapolation for unknown or variable composition, complex reaction product or biological material (UVCB) substances. UVCBs such as petroleum substances are commonly subjected to dimethyl sulfoxide (DMSO) extraction in order to retrieve the bioactive polycyclic aromatic compound (PAC) portion for in vitro testing. In addition to DMSO extraction, protein binding in cell culture media and dilution can all influence in vitro bioavailable concentrations of aliphatic and aromatic compound

Neurotoxicity Associated With Dimethyl Sulfoxide Used in Allogeneic Stem Cell Transplantation.

Dimethyl sulfoxide (DMSO) is a cryoprotective agent used in storage of frozen stem cells in stem cell transplantation. Central nervous system side effects of DMSO such as epileptic seizures, stroke, transient global amnesia, and temporary leucoencephalopathy are rarely seen. Here, we report a pediatric patient who developed seizures after DMSO-cryopreserved stem cell infusion and whose magnetic resonance imaging of the brain demonstrated parietal and occipital focal cortical T2-signal intensity increase. DMSO toxicity should be kept in mind in patients who received cryopreserved stem cell infu
