[A useful marker for differential diagnosis of Parkinson's disease--MIBG myocardial scintigraphy].

We performed [123I]-meta-iodobenzylguanidine (MIBG) myocardial scintigraphy in 180 patients with PD, 24 patients with DLB and 73 patients with the other related diseases. The heart-to-mediastinum (H/M) ratio in PD and DLB was significantly low compared to that in MSA, PSP, CBD and AD. The H/M ratio tended to decrease with the disease progression in PD. These findings suggest that MIBG myocardial scintigraphy could be a marker for differential diagnosis of PD and DLB. We immunohistochemically examined heart tissues from patients with pathologically confirmed PD, DLB and the other related diseas

Excitatory and inhibitory responses of the ongoing sympathetic discharge in single renal neurons to liminal stimulation of aortic C-fibres in the rabbit.

Excitatory and inhibitory responses of sympathetic discharge were recorded in single renal postganglionic neurons of rabbits anaesthetized with urethane and chloralose. The animals were vagotomized and had transected aortic nerves. Responses were elicited by single volleys in the aortic C-fibres. Excitatory responses consisted in short-lasting increase in the rate of ongoing sympathetic discharge and were followed by inhibitory responses. Excitatory effects together with inhibitory responses were seen in 68% of units (19/28). Only excitatory effects appeared in 2 neurons (7.1%) and only inhibi

Diagnosing Horner's syndrome.

The diagnosis of Horner's syndrome is divided into two stages: (1) the recognition of the sympathetic deficit, and (2) the localization of the lesion. The second step is of vital importance in the management of the patient with Horner's syndrome. If the lesion is preganglionic, the risk of malignancy is high; the patient should have cervical spine and chest roentgenograms (PA, lateral and apical lordotic) and perhaps a referral to a surgeon. If the lesion is postganglionic, it is most likely a benign vascular headache syndrome and the patient should go to a neurologist.

Electrically induced quantitative sudomotor axon reflex test in human volunteers.

Chemically-induced quantitative sudomotor axon reflex test (QSART) and quantitative sensory testing (QST) are established clinical tools to assess thin fiber function in humans. We investigated stimulus-response functions to transcutaneous electrical stimuli of different current intensity (3.75 to 10mA) and pulse frequency (5 to 100Hz) comparing sweat output (ml/h/m(2)) and pain intensity (numeric rating scale [NRS], 0-10). Efferent sudomotor and afferent nociceptive responses were recorded after a 30s electrical stimulation period of distal (hand and foot) and proximal (forearm and thorax) bo

Distribution of sympathetic nerve endings within the rabbit carotid body: a histochemical and ultrastructural study.

The distribution of sympathetic (noradrenergic) nerves within the rabbit carotid body was investigated by fluorescence microscopy and electron microscopy. With the Falck-Hillarp method, fluorescent nerve fibres were found associated with blood vessels and type I/type II cell groups. After the injection of 6-hydroxydopamine, degenerating nerve profiles were found near blood vessels, near type I/type II cell groups and in contact with type II cells. Following the administration of [3H]noradrenaline, labelled nerve profiles were observed in close association with blood vessels and around cell gro

Action of beta-adrenoceptor blocking agents on hemodynamic variables and on baroreceptor and postganglionic sympathetic activities in rabbits.

Fifty-six experiments were performed in rabbits anesthetized by urethane or sodium pentobarbital. Baroreceptor (aortic nerve) and post-ganglionic sympathetic (renal nerve) activities were recorded, and peripheral resistances were determined after infusion of beta-adrenoceptor blocking agents (propranolol, pindolol, timolol). A progressive vasodilation was observed after the onset of infusion of the 3 drugs; at the same time arterial pressure decreased, but aortic flow did not vary significantly. Baroreceptor activity was enhanced, whereas efferent renal nerve activity was decreased.

Plasticity in Thoracic Paravertebral Sympathetic Postganglionic Neurons after High Spinal Cord Transection.

Various presympathetic descending brain circuits recruit spinal cord preganglionic neurons to encode central sympathetic drive via their synaptic actions onto sympathetic postganglionic neurons (SPNs)-the final sympathetic output neurons. Thoracic paravertebral ganglia SPNs (tSPNs) provide distributed control over body tissue systems via functional subpopulations. High thoracic spinal cord injuries (SCIs) compromise supraspinal control of SPNs, causing dysautonomias including hypotension. In adult mice of either sex, we tested whether SCI-induced chronic loss of supraspinal control of tSPN act

Adrenergic and cholinergic innervation of the chicken pancreas.

The distribution, as well as the morphological characteristics of adrenergic and cholinergic nerve fibres was studied in the pancreas of the hen and the cock. The presence of numerous adrenergic and moderately numerous cholinergic structures was revealed in the organ. They were seen as nerve fibre bundles or single nerve fibres located in the vicinity of blood vessels and exocrine ducts, as well as the cells of the exocrine and endocrine pancreas. Single TH- and ChAT-positive nerve cell bodies were also found in the organ under study.

Causalgia, pathological pain, and adrenergic receptors.

Control of expression of molecular receptors for chemical messengers and modulation of these receptors' activity are now established as ways to alter cellular reaction. This paper extends these mechanisms to the arena of pathological pain by presenting the hypothesis that increased expression of alpha-adrenergic receptors in primary afferent neurons is part of the etiology of pain in classical causalgia. It is argued that partial denervation by lesion of peripheral nerve or by tissue destruction induces a change in peripheral nociceptors, making them excitable by sympathetic activity and adren

Physiological and metabolic functions of melatonin.

Melatonin is a lipophilic hormone, mainly produced and secreted at night by the pineal gland. Melatonin synthesis is under the control of postganglionic sympathetic fibers that innervates the pineal gland. Melatonin acts via high affinity G protein-coupled membrane receptors. To date, three different receptor subtypes have been identified in mammals: MT1 (Mel 1a) and MT2 (Mel 1b) and a putative binding site called MT3. The chronobiotic properties of the hormone for resynchronization of sleep and circadian rhythms disturbances has been demonstrated both in animal models or in clinical trials. S

Properties of Na+ currents in putative submandibular and cardiac sympathetic postganglionic neurones.

This study was performed to compare the kinetic properties of Na+ currents in putative salivary and cardiac postganglionic sympathetic neurones isolated from the superior cervical and stellate ganglia, respectively. Neurones were labelled with a fluorescent tracer-Fast Blue, injected into the submandibular gland (in the case of salivary neurones) and into the pericardial cavity or left ventricular wall (in the case of cardiac neurones). Voltage-dependent Na+ current was then isolated and recorded from labelled cells. The major findings of this study were: (1) Peak Na+ current was larger in sal

Delayed sympathetic dependence in the spared nerve injury (SNI) model of neuropathic pain.

BACKGROUND: Clinical and experimental studies of neuropathic pain support the hypothesis that a functional coupling between postganglionic sympathetic efferent and sensory afferent fibers contributes to the pain. We investigated whether neuropathic pain-related behavior in the spared nerve injury (SNI) rat model is dependent on the sympathetic nervous system. RESULTS: Permanent chemical sympathectomy was achieved by daily injection of guanethidine (50 mg/kg s.c.) from age P8 to P21. SNI was performed at adulthood followed by 11 weeks of mechanical and thermal hypersensitivity testing. A signi

Paradoxical muscle sympathetic reflex activation in human heart failure.

BACKGROUND: Muscle sympathetic activation in heart failure with reduced ejection fraction (HFrEF) has been attributed, on the basis of multiunit recordings, to attenuated inhibitory feedback from stretch-sensitive cardiopulmonary mechanoreceptors. However, such preparations integrate 2 populations of single units exhibiting directionally opposite firing when atrial pressure is perturbed. We tested the hypothesis that the proportion of single units firing paradoxically when filling pressure increases is augmented in HFrEF. METHODS AND RESULTS: Muscle sympathetic nerve activity and estimated ce

Modulation of parasympathetic neuron phenotype and function by sympathetic innervation.

Selective sympathetic nerve dysfunction occurs during aging and in certain disease states. Here, we review findings concerning the effects of chronic sympathetic denervation on parasympathetic innervation to orbital target tissues in the adult rat. Long-term sympathetic denervation was induced by excising the ipsilateral superior cervical ganglion for 5-6 weeks prior to analyses. Following sympathectomy, pterygopalatine ganglion parasympathetic neurons show reduced nitric oxide synthase protein in their somata and projections to vascular targets. Laser Doppler measurements of ocular blood flow

Ultrastructure of the nerves in veins from human omentum.

To study the ultrastructural characteristics of the sympathetic nerve terminals of human omental veins, and of their relationship to the innervated smooth muscle cells, biopsy specimens were taken during abdominal surgery, rapidly fixed in glutaraldehyde/osmium and stained with uranylacetate. The results indicate that the veins have an extensive noradrenergic innervation, penetrating into the tunica media. The distance between nerve terminals partly or wholly free from enveloping Schwann cells, to the surface of smooth muscle cells ranged from 30 to 500 nm. Large dense core vesicles were promi

Role of NaV1.7 in postganglionic sympathetic nerve function in human and guinea-pig arteries.

NaV1.7 plays a crucial role in inducing and conducting action potentials in pain-transducing sensory nociceptor fibres, suggesting that NaV1.7 blockers could be effective non-opioid analgesics. While SCN9A is expressed in both sensory and autonomic neurons, its functional role in the autonomic system remains less established. Our single neuron rt-PCR analysis revealed that 82% of sympathetic neurons isolated from guinea-pig stellate ganglia expressed NaV1.7 mRNA, with NaV1.3 being the only other tetrodotoxin-sensitive channel expressed in approximately 50% of neurons. We investigated the role

Inhibition of glial glutamate transporter GLT1 in the nucleus of the solitary tract attenuates baroreflex control of sympathetic nerve activity and heart rate.

The astrocytic glutamate transporter (GLT1) plays an important role in the maintenance of extracellular glutamate concentration below neurotoxic levels in brain. However, the functional role of GLT1 within the nucleus of the solitary tract (NTS) in the regulation of cardiovascular function remains unclear. We examined the effect of inhibiting GLT1 in the subpostremal NTS on mean arterial pressure (MAP), renal sympathetic nerve activity (RSNA) and heart rate (HR) in anesthetized, artificially ventilated rats. It was found that dihydrokainate (DHK; inhibitor of GLT1, 5 mmol/L, 100 nL) injections
