A two-generation female reproduction and teratology study was undertaking using 0, 1.5 and 3.0 ppm of T-2 toxin in a semisynthetic diet. Conception rate was lowest at the first breeding and lowest in the control group at this time, but in later breedings the conception rates of each group were very similar. No significant differences were noted in litter size, death of fetuses, sex of fetuses and gestational length. Feed consumption was similar in all groups, as were the non-pregnant or pregnant female weight gains. Although the second-generation offspring grew faster than the first-generation

The mechanism of environmental factors in Kashin-Beck disease (KBD) remains unknown. We aimed to identify single nucleotide polymorphisms (SNPs) and protein alterations of selenium- and T-2 toxin-responsive genes to provide new evidence of chondrocytic damage in KBD. This study sampled the cubital venous blood of 258 subjects including 129 sex-matched KBD patients and 129 healthy controls for SNP detection. We applied an additive model, a dominant model, and a recessive model to identify significant SNPs. We then used the Comparative Toxicogenomics Database (CTD) to select selenium- and T-2 to

Both T-2 toxin and selenium (Se) cause immune impairment in the spleen, but the mechanism of their occurrence is not clear. In this experiment, the Se content of the spleen was tested by atomic fluorescence spectrometry after a 12-week intervention in Sprague-Dawley (SD) rats, divided into normal, normal + T-2 toxin (10 ng/g), normal + T-2 toxin (100 ng/g), low Se, low Se + T-2 toxin (10 ng/g), and low Se + T-2 toxin (100 ng/g) groups. The pathological changes and fibrosis of spleen tissue were observed using hematoxylin-eosin (HE) staining and Masson staining, respectively. Mitogen-activated

This study was conducted to clarify and reevaluate the cardiac and autonomic nervous effects of T-2 toxin, which had been previously examined by several acute experiments, in unrestrained and conscious rats implanted with telemetric transmitters. Two groups of rats were given two subcutaneous injections of 0.1 and 0.5 mg/kg of T-2 toxin with an interval of 3 days. Two other groups of rat were pre-implanted with osmotic minipumps by which atropine (20 mg/kg/day) or propranolol (100 mg/kg/day) was continuously administered preceding subcutaneous injection of T-2 toxin (0.5 mg/kg). The present st

T-2 toxin, a type-A trichothecene mycotoxin, exists ubiquitously in mildewed foods and feeds. Betulinic acid (BA), a pentacyclic triterpenoid derived from plants, has the effect of relieving inflammation and oxidative stress. The purpose of this study was to investigate whether BA mitigates lung impairment caused by T-2 toxin and elucidate the underlying mechanism. The results indicated that T-2 toxin triggered the inflammatory cell infiltration, morphological alterations and cell apoptosis in the lungs. It is gratifying that BA ameliorated T-2 toxin-caused lung injury. The protein expression

T-2 toxin, a highly toxic trichothecene mycotoxin widely found in food and feed, poses a significant threat to human health as well as livestock and poultry industry. Liver, being a crucial metabolic organ, is particularly susceptible to T-2 toxin induced damage characterized by inflammation and oxidative stress. Despite the role of Sirtuin 5 (SIRT5) in mitigating liver injury has been confirmed, its specific impact on T-2 toxin induced liver injury remains to be elucidated. The objective of this study was to investigate the protective role of SIRT5 against T-2 toxin induced liver injury in mi

BACKGROUND: The gastrointestinal tract is the first target for the potentially harmful effects of mycotoxins after intake of mycotoxin contaminated food or feed. With deoxynivalenol (DON), T-2 toxin (T-2), fumonisin B1 (FB1) and zearalenone (ZEA) being important Fusarium toxins in the northern hemisphere, this study aimed to investigate in vitro the toxic effect of these mycotoxins on intestinal porcine epithelial cells derived from the jejunum (IPEC-J2 cells). Viability of IPEC-J2 cells as well as the proportion of apoptotic and necrotic IPEC-J2 cells was determined by flow cytometry after 72

T-2 toxin is a persistent, bioaccumulative environmental contaminant that poses major health threats to humans and animals. Endoplasmic reticulum (ER) stress and autophagy are two interconnected stress responses critical for maintaining cellular homeostasis. Berbamine (BBM) is an important member of bis-benzy lisoquinoline alkaloid with diverse biological activities. This study aimed to identify the molecular target of BBM against T-2 toxin-induced hepatotoxicity, focusing on autophagy-ER stress crosstalk. We systematically evaluated autophagy and ER stress in human HepaRG cells using immunobl

Kashin-Beck disease (KBD) is a debilitating endemic osteochondropathy. Environmental toxins, particularly T-2 toxin, and selenium deficiency are established etiological factors. This study aimed to investigate whether T-2 toxin induces ferroptosis in articular chondrocytes and explore the therapeutic potential and mechanism of the iron chelator, Deferoxamine (DFO). A total of 32 three-week-old male Sprague-Dawley rats were randomly allocated into four groups: Control, T-2 toxin (100 ng/g·BW/d), T-2 toxin + low DFO (10 mg/kg), and T-2 toxin + high DFO (100 mg/kg). Conventional multicolor fluorescent labeling has inconsistent signal and low quantum yield, restricting sensitivity for low-abundance mycotoxin detection. This study developed a dual-channel immunochromatographic assay (ICA) strip based on color-coding properties of persistent luminescent nanoparticles (PLNPs). By precisely controlling the type of doping ions in Zn2GeO₄ (ZGO) and optimizing the reaction pH, PLNPs with intense green and blue emission were successfully synthesized. The as-prepared PLNPs exhibit high quantum yield and robust optical stability, which effectively reduce background

Thymic epithelial cells (TECs) are critical for thymic structure and function, yet the impact of T-2 toxin (T-2) on TECs and related molecular pathways remains unclear. This study sheds light on the mechanisms of T-2-induced TEC damage, focusing on the ROS-NF-κB-NLRP3 signaling axis. The in vivo and in vitro analyses suggest that T-2 induces TEC injury through ROS-driven NLRP3 inflammasome activation, NF-κB signaling, inflammation, and apoptosis. Molecular docking analysis verified the binding of T-2 to critical components involved in oxidative stress, inflammatory signaling pathways, and apop

T-2 toxin, the most toxic type A trichothecene, is widely present in grain and animal feed, causing growth retardation and tissue damage in poultry. Geese are more sensitive to T-2 toxin than chickens and ducks. Although T-2 toxin has been reported to cause tibial growth plate (TGP) chondrodysplasia in chickens, tibial damage caused by T-2 toxin in geese has not been fully demonstrated. This study aims to investigate the adverse effects of T-2 toxin on tibial bone development, bone quality, chondrocyte differentiation, and bone metabolism. T-2 toxin, a mycotoxin found in foods and feeds, poses a threat to female reproductive health in both humans and animals. LncRNA CUFF.253988.1 (CUFF.253988.1), highly expressed in pigs, has an undisclosed regulatory role. This study aimed to establish a model of T-2 toxin-induced ovarian injury in sows, both in vivo and in vitro, and to explore the regulatory role and potential mechanisms of CUFF.253988.1. The results showed that feeding T-2 toxin-contaminated feed (1 mg/kg) induced ovarian follicle atresia and mitochondrial structural damage, accompanied by a significant upregulation of CUFF.

As common pathogenic agents in the world and widely distributed globally, T-2 toxin and selenium deficiency might exacerbate toxic effects by combined exposure, posing a dramatic health hazard to humans and animals. In this study, we aim to elucidate the underlying mechanisms of renal fibrosis triggered by T-2 toxin and selenium deficiency exposure. A total of thirty-two rats are randomly divided into the normal control, T-2 toxin, selenium deficiency, and combined intervention groups. T-2 toxin (100 ng/g) is intragastric gavaged to the rats in compliance with the body weight. Both the standar

The T-2 toxin, a major secondary metabolite of Fusarium Gramineae, is considered a great risk to humans and animals due to its toxicity, such as inducing emesis. The mechanism of emesis is a complex signal involving an imbalance of hormones and neurotransmitters, as well as activity of visceral afferent neurons. The T-2 toxin has been proven to induce emesis and possess the capacity to elevate expressions of intestinal hormones glucagon-like peptide-17-36 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP), both of which are important emetic factors. In addition, the activation of c

LncRNA myocardial infarction associated transcript (MIAT) has been shown to be involved in osteoarthritis (OA), but its role in Kashin-Beck Disease (KBD) has rarely been reported. In this study, rats were administered with low selenium and/or T-2 toxin for 4 weeks to establish a KBD animal model. The serum selenium level, TNF-α and IL-1β contents, phosphorylated p65 (p-p65) and MIAT expression were increased in each intervention group. Next, we isolated the primary epiphyseal chondrocytes, and found that selenium treatment reversed the effects of T-2 toxin on chondrocyte injury, p-p65 and MIAT

BACKGROUND: T-2 toxin, a highly toxic trichothecene mycotoxin contaminating cereal grains, poses serious health threats. However, its specific effects on hepatic drug-metabolizing enzymes (DMEs) remain poorly characterized. This study aimed to investigate the impact of T-2 toxin on key cytochrome P450 (CYP) and carboxylesterase (CES) enzymes in mouse liver.
METHODS: Male C57BL/6 mice were randomly divided into control and T-2 toxin-treated groups, receiving either normal saline or T-2 toxin (0.2 mg/kg) by oral gavage daily for four weeks. Histopathological and ultrastructural alterations were

Although Kashin-Beck disease (KBD) in China has been effectively controlled through comprehensive measures such as selenium supplementation and grain replacement, the underlying environmental risk factors may still persist. There remains a potential risk of disease resurgence if prevention and control efforts are relaxed. To dynamically assess the selenium exposure levels in humans (using children’s hair selenium as a biomarker) and the selenium status in the external environment (soil selenium and grain selenium) in historical severe endemic areas and adjacent non-endemic areas in China, whil

It has been strongly suggested that selenium deficiency and T-2 contamination in cereals are responsible for the development of Kashin-Beck disease (KBD). In order to assess these risk factors of KBD in the internal and external environments, our team undertook a two-stage survey in some areas of Heilongjiang and Gansu Provinces, China. The selenium content in children's hair (293), cereal (192), and soil (46) samples were determined using the 2, 3-diamino-naphthalene fluorometric assay technique. The T-2 toxin contamination level in the cereal samples (704) was assayed using an ELISA kit. The
