Immunohistochemical localization of the matrix glycoprotein tenascin in the skull of the growing rat.

Tenascin is an extracellular matrix glycoprotein which interacts with other matrix molecules and with cells, and which appears to play important roles in growth and differentiation. The immunohistochemistry of sections of newborn, 5-day and 20-day-old rats showed that accumulation of tenascin was largely restricted to bones, cartilages and teeth. It was also present in the periosteal and endosteal surfaces of membrane bones, in perichondrium, and ion the dental pulp, but was absent from mature bone, cartilage and dentine. In nasal cartilage, tenascin was present only in the perichondrium, wher…

Modeling Tenascin-C-rich metastatic tumor niches in engineered hydrogel biomaterials.

The extracellular matrix (ECM) plays a pivotal role in shaping tumor behavior by providing biochemical and biophysical cues to cancer cells. … ollagen I and fibronectin. To model metastatic ECM and further tune the hydrogel, we incorporated a tenascin-C (TNC)-derived peptide. To study the impact of these tunable ECM parameters on cancer cell behavior, we encapsulated Ewing sarcoma (EwS) cells within the hydrogels. EwS is an aggressive bone and soft tissue tumor that commonly metastasizes to lung. …

Tenascin-C levels in polycystic ovary syndrome and polycystic ovarian morphology.

OBJECTIVE: It is considered that the underlying reason for metabolic anomalies and ovarian dysfunction in polycystic ovary syndrome (PCOS) is low-grade chronic inflammation. Therefore, we aimed to investigate Tenascin-C (TN-C) levels in PCOS. This is the first study to document the impact of inflammation with TN-C in this specific population. METHODS: A total of 90 participants, consisting of 30 PCOS patients, 30 polycystic ovarian morphology (PCOM) patients, and 30 matched healthy controls between September 2018 and April 2019 enrolled. All participants were randomized by age and body mass i…

Chemokine Binding to Tenascin-C Influences Chemokine-Induced Immune Cell Migration.

Tenascin-C (TNC) is a complex glycoprotein of the extracellular matrix (ECM) involved in a plethora of (patho-)physiological processes, such as oncogenesis and inflammation. Since chemokines play an essential role in both disease processes, we have investigated here the binding of TNC to some of the key chemokines, namely CCL2, CCL26, CXCL8, CXCL10, and CXCL12. Thereby, a differential chemokine-TNC binding pattern was observed, with CCL26 exhibiting the highest and CCL2 the lowest affinity for TNC. Heparan sulfate (HS), another member of the ECM, proved to be a similarly high-affinity ligand o…

Bone Marrow-Derived Tenascin-C Attenuates Cardiac Hypertrophy by Controlling Inflammation.

BACKGROUND: Tenascin-C (TNC) is a highly conserved matricellular protein with a distinct expression pattern during development and disease. Remodeling of the left ventricle (LV) in response to pressure overload leads to the re-expression of the fetal gene program. OBJECTIVES: The aim of this study was to investigate the function of TNC in cardiac hypertrophy in response to pressure overload. METHODS: Pressure overload was induced in TNC knockout and wild-type mice by constricting their abdominal aorta or by infusion of angiotensin II. Echocardiography, immunostaining, flow cytometry, quantit…

Reduced quantitative muscle function in tenascin-X deficient Ehlers-Danlos patients.

The Ehlers-Danlos Syndrome (EDS) is a heterogeneous group of heritable connective tissue disorders. Skeletal muscle features belong to the clinical criteria of EDS and are generally interpreted to result from increased tendon distensibility or exercise avoidance. However, muscle function in EDS has hardly been investigated as such. We performed a pilot study consisting of clinical investigations, electromyography, muscle ultrasound, muscle biopsy, and quantitative muscle function tests on two EDS patients with deficiency of tenascin-X. Quantitative muscle function proved severely reduced despi…

TGF-beta2, tenascin, and integrin beta1 expression in superior limbic keratoconjunctivitis.

PURPOSE: To determine the pathophysiological etiology of superior limbic keratoconjunctivitis (SLK), we compared the superior limbic conjunctivae of SLK patients and normal controls. METHODS: Frozen sections of conjunctival specimens from five SLK patients and two controls were examined by immunohistochemical methods. Transforming growth factor (TGF)-beta2, integrin beta1, and tenascin (TN) were chosen for analysis because their expression is known to be affected by mechanical stress or injury. The staining pattern was observed by confocal laser scanning microscopy. RESULTS: Prominent positi…

Tenascin-C drives cardiovascular dysfunction in a mouse model of diabetic cardiomyopathy.

BACKGROUND: Diabetic cardiomyopathy (DCM) is a complex condition linked to diabetes, characterized by cardiac and vascular dysfunction, frequently concomitant with heart failure with preserved ejection fraction. The extracellular matrix glycoprotein Tenascin-C (TNC) has been found to be upregulated under diabetic conditions. However, the potential contributory role of TNC in the progression of DCM remains largely unclear. This study was designed to elucidate the role of TNC in the pathogenesis of DCM. METHODS: Diabetes was induced in adult male wild-type (WT) and TNC knockout (TNC-KO) mice, t…

Tenascin-X as a causal gene for classical-like Ehlers-Danlos syndrome.

Tenascin-X (TNX) is an extracellular matrix glycoprotein for which a deficiency results in a recessive form of classical-like Ehlers-Danlos syndrome (clEDS), a heritable connective tissue disorder with hyperextensible skin without atrophic scarring, joint hypermobility, and easy bruising. Notably, patients with clEDS also suffer from not only chronic joint pain and chronic myalgia but also neurological abnormalities such as peripheral paresthesia and axonal polyneuropathy with high frequency. By using TNX-deficient (Tnxb -/-) mice, well-known as a model animal of clEDS, we recently showed that…

Increased expression of tenascin-x in thoracic and abdominal aortic aneurysm tissues.

Tenascin-X (TNX), which has a molecular mass of roughly 450 kDa, is the largest member of the tenascin family. Complete deficiency of TNX in humans leads to a recessive form of Ehlers-Danlos syndrome (EDS). TNX is expressed abundantly in a variety of tissues, especially in cardiac muscle and in perivascular stroma. Human TNX is also present in serum with an apparent molecular size of 140 kDa. In the present study, we investigated the expression levels of TNX protein in thoracic and abdominal aortic aneurysm tissues. The level of TNX was significantly increased in both aortic aneurysm tissues c…

Novel localization of tenascin-X in adult mouse leptomeninges and choroid plexus.

Tenascin-X (Tn-X) belongs to the tenascin family of glycoproteins and is clearly associated with the human connective tissue disorder Ehlers-Danlos syndrome. Recently, human single nucleotide polymorphism analyses showed that Tn-X is associated with schizophrenia. Tn-X-related central nervous system (CNS) disorder has been reported in recent years. However, details of Tn-X localization are not clear in the adult cerebral cortex and its meninges. Using immunohistochemical techniques, we found novel localizations of Tn-X in the leptomeningeal trabecula (TB) of adult mice and in the connective ti…

Progression of human aortic valve stenosis is associated with tenascin-C expression.

OBJECTIVES: We sought to assess tenascin-C (TN-C) expression and its possible pathobiological impact in human aortic valve stenosis. BACKGROUND: Tenascin-C, a large extracellular matrix glycoprotein, has lately been increasingly connected to cardiovascular pathologies. As TN-C is a multifunctional protein implicated in cell proliferation, migration and differentiation, we investigated the pattern of its expression in diseased human aortic valves. METHODS: Fifty-five tricuspid, non-rheumatic stenotic aortic valves were collected from patients undergoing aortic valve replacement, and the contr…

Tenascin expression in mucocutaneous diseases and related lesions of human oral mucosa.

The expression of tenascin was assessed immunohistochemically. In normal oral mucosa, immunoreactivity for tenascin was seen either as a delicate line underlining the epithelium or in the stromal papillae. In oral lichen planus, a marked enhancement of tenascin immunoreactivity in the lamina propria was associated with focal infiltrates of inflammatory cells and seemed to reflect the intensity of inflammation. In lichenoid reactions in which only a sparse inflammatory infiltrate was present a band-like tenascin reactivity was seen. Oral psoriform reactions and chronic hyperplastic candidosis s…

Dermo-epidermal separation is associated with induced tenascin expression in human skin.

Tenascin, a large glycoprotein of the extracellular matrix, shows a site-restricted distribution during embryogenesis, and can be found in adults in a variety of pathological conditions. In normal skin, tenascin is expressed at low levels, but it is upregulated in skin tumours, in a number of skin diseases with epidermal hyperproliferation and during wound healing. Several tenascin variants have been described, and these arise by alternative splicing. Using a monoclonal antibody recognizing all tenascin variants, and polyclonal antibodies specific for the large tenascin variants, we have inves…

Nerve-dependent and -independent tenascin expression in the developing chick limb bud.

The extracellular matrix protein, tenascin, appears in a restricted pattern during organ morphogenesis. Tenascin accumulates along developing peripheral nerves as they leave the spinal cord and enter the limb mesenchyme (Wehrle and Chiquet, Development 110, 401-415, 1990). Here we found that most but not all tenascin deposited along growing nerves is of glial origin. By in situ hybridization with a tenascin cDNA probe, we determined the site of tenascin mRNA accumulation both in normal and nerve-free limbs. In normal wing buds, tenascin mRNA was first detected within the developing limb nerves…

Tenascin-XB plays a role in the infiltration of immune cells in tumor microenvironment.

We previously showed that tenascin-XB (TNXB) contributes to tumor suppressor function. The present study aimed to assess the tumor-suppressive mechanism of TNXB by focusing on immune cell infiltration into the tumor microenvironment (TME). We revealed that B16-OVA melanoma cells (MO5)-bearing TNXB-deficient (Tnxb-/-) mice exhibited significant tumor progression and a poor survival rate. Allogeneic mixed lymphocyte reaction showed reduced numbers and increased activation of both CD4+ and CD8+ T cells from Tnxb-/- spleens. Moreover, T cell activation assay further proved that CD4+ and CD8+ T cel…

Tenascin-C in cardiac disease: a sophisticated controller of inflammation, repair, and fibrosis.

Tenascin-C (TNC) is a large extracellular matrix glycoprotein classified as a matricellular protein that is generally upregulated at high levels during physiological and pathological tissue remodeling and is involved in important biological signaling pathways. In the heart, TNC is transiently expressed at several important steps during embryonic development and is sparsely detected in normal adult heart but is re-expressed in a spatiotemporally restricted manner under pathological conditions associated with inflammation, such as myocardial infarction, hypertensive cardiac fibrosis, myocarditis…

TNIIIA2, The Peptide of Tenascin-C, as a Candidate for Preventing Articular Cartilage Degeneration.

OBJECTIVE: TNIIIA2 is a peptide of the extracellular matrix glycoprotein tenascin-C. We evaluated whether intra-articular injection of TNIIIA2 could prevent articular cartilage degeneration without inducing synovitis in an osteoarthritis mice model. DESIGN: Ten micrograms per milliliter of TNIIIA2 were injected into the knee joint of mice (group II) to evaluate the induction of synovitis. The control group received an injection of phosphate buffered saline (group I). Synovitis was evaluated using synovitis score 2 and 4 weeks after injection. The ligaments of knee joints of mice were transect…
