Stat3 in resident macrophages as a repressor protein of inflammatory response.

Inflammation is counterbalanced by anti-inflammatory cytokines such as IL-10, in which Stat3 mediates the signaling pathway. In this study, we demonstrate that resident macrophages, but not other cell types, are important targets of IL-10 in a murine model of acute peritonitis. Injection of thioglycollate i.p. induced a considerable number of neutrophils and macrophages in the peritoneum, which was significantly augmented in mice with a cell-type specific disruption of the Stat3 gene in macrophages and neutrophils (LysMcre/Stat3flox/- mice). The augmented leukocyte infiltration was accompanied

Apparent successful mesothelial cell transplantation hampered by peritoneal activation.

BACKGROUND: Mesothelial cell transplantation has been suggested to improve mesothelial repair after surgery, recurrent peritonitis and peritoneal dialysis. METHODS: In this study we evaluated mesothelial cell transplantation during the resolution phase of experimentally thioglycollate-induced peritonitis in rats. To this end 4 x 10(6) DiO-labeled autologous mesothelial cells were transplanted 1 week after peritonitis induction. Peritoneal inflammation and permeability characteristics were evaluated after another week. RESULTS: Mesothelial cell transplantation after peritonitis resulted in in

Differential expression of IL-1 and TNF receptors in murine macrophages infected with virulent vs. avirulent Legionella pneumophila.

Infection of macrophages from genetically susceptible A/J mice with Legionella pneumophila induces high levels of various cytokines in serum as well as in cultures of spleen or peritoneal cells from the mice. However, modulation of receptor expression for these cytokines during infection has not been studied in detail, even though these receptors on macrophages have a critical role in inflammatory responses during the infection. In the present study, the differential expression of mRNA for TNF and IL-1 receptors as well as receptor antigens during infection of macrophages with virulent vs. avi

Group B streptococcus-induced nitric oxide production in murine macrophages is CR3 (CD11b/CD18) dependent.

Nitric oxide (NO) is produced by murine macrophages in response to cytokines and/or gram-negative bacterial lipopolysaccharide. NO induction by gram-positive bacteria such as group B streptococci (GBS), the major etiologic agents of neonatal pneumonia and meningitis, has received little study. GBS as well as two other gram-positive bacterial species, Staphylococcus aureus and Staphylococcus epidermidis, were found to stimulate NO production in thioglycolate-elicited murine macrophages and in the mouse macrophage cell line J774A.1 in the presence of gamma interferon. Serotype Ia and III GBS wer

[A case of chronic actinomycosis in a carcinomatous lesion].

A gram positive, catalase-positive Actinomyces viscosus strain has been isolated from cervicofascial carcinomatous lesion. Identification of this human strain of Actinomyces viscosus was carried out with biochemical tests. Tests for catalase, indole, nitrate reduction, methyl red reaction acetoin (VP) and gelatin hydrolysis were done by the procedure recommended by the technique described by Grencser and Slack. Carbohydrate fermentation tests were done with three basal media: Actinomyces fermentation broth, a thioglycolate fermentation base and a brain-heart infusion broth.

Activation of K+ current in macrophages by platelet activating factor.

Puff application of platelet activating factor (10(-8) M) onto peritoneal macrophages from thioglycollate-stimulated mice induced an outward current at a holding potential of -63 mV. The current was suppressed by an antagonist Y-24180 but not by CV-3988. Charybdotoxin (10(-6) M) suppressed the current. Reversal potentials were dependent on external K+ concentrations. The current was not suppressed in Ca(2+)-free EGTA-containing solution but was completely abolished in BAPTA-AM containing solution. These results suggest that platelet activating factor activates a Ca(2+)-dependent K+ channel.

Synthesis of some N-substituted indole derivatives and their biological activities.

Acylation of 2,3-diphenyl-5-methoxy-indole using ethyl chloroformate or chloroacetyl chloride in dimethylformamide and sodium hydride yielded the N-substituted derivatives 1 and 2, respectively. While Friedel-Crafts acylation using chloroacetyl chloride afforded di-4,6-chloroacetyl derivative 3, the reaction of the N-chloroacetyl derivative 2 with amines, hydrazines, urea, semicarbazide hydrochloride, thiophenol, benzimidazole-2-thiol, thiosemicarbazide, 2-mercaptoethanol and thioglycolic acid was studied. Several of the compounds were tested for their effect on arterial blood pressure, antiin

Permanent waving and straightening of hair.

A significant number of consumers use products to curl or straighten their hair. These products function by chemically breaking and reforming keratin disulfide bonds. The most popular waving products contain salts of thioglycolic acid to break the bonds and hydrogen peroxide to reform them. Straighteners generally rely on high alkalinity to break the bonds and reform them with acid. Problems with permanent waving are usually due to overtreatment or undertreatment of the hair rather than allergy or irritation. Straighteners can be irritating because of their high alkalinity and can seriously da

[Peculiarities of alkyl tin effects on respiration and oxidative phosphorylation of rat liver mitochondria].

Electron transfer and oxidative phosphorylation were studied as affected by tinalkyls. It is shown that all of them inhibit effectively respiration of the rat liver mitochondria. The action of bis(tributyl tin)oxide and dibutyl diisooctyl thioglycolate tin on the mitochondria in a state of dissociation is localized in the terminal step of the respiration chain. The other alkyls act in the site located before cytocrome c. The effect of bis (tributyl tin) oxide on both the ADP-activated respiration and DNP-activated ATPase of intact mitochondria is similar to that of oligomycin. These effects mi

The effect of thiols on immunologic release of slow reacting substance of anaphylaxis. I. Human lung.

Preincubation of human lung fragments with cysteine for 2.5 to 5.0 min resulted in dose-dependent, selective enhancement of the antigen-induced or anti-IgE-induced formation and release of slow reacting substance of anaphylaxis (SRS-A). Comparable effects were observed with sodium sulfide and thioglycolate but not with other more potent reducing agents or metabolites of cysteine. Sulfhydryl alkylated derivatives of cysteine were ineffective. The effects observed with the active thiols were easily reversed and could not be attributed to an action in the bioassay or on SRS-A itself. The physicoc

Endogenous SLPI contributes to the regulation of inflammatory responses in peritoneal macrophages by modulating MMP-9 production.

Secretory leukocyte protease inhibitor (SLPI) is described as a potent regulator of inflammation and tissue homeostasis with pleiotropic functions. It has been shown to inhibit pro-inflammatory responses in myeloid cells. However, its expression patterns and specific functions in different monocyte and macrophage populations remain poorly understood. Therefore, we investigated its expression patterns in murine tissue macrophage populations by analysis of publicly available datasets and flow cytometry. Among various tissues, peritoneal macrophages were identified as a major source of SLPI, sugg

Effective removal of textile dye via synergy of adsorption and photocatalysis over ZnS nanoparticles: Synthesis, modeling, and mechanism.

In this work, we prepared sulfur-zinc nanoparticles (ZnS-TGA) functionalized with thioglycolic acid by a hydrothermal method and tested their photodegradation ability by solar irradiation. ZnS-TGA were characterized by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), high-resolution transmission electron microscope (HR-TEM), UV-Vis spectrophotometer and photoluminescence spectroscopy. In the characterization of these nanoparticles, thioglycolic acid proved to be a strong capping ligand, with a specific surface area of 36.82 m2/g and an average size of 7.15 nm. To test t

The effect of tissue type-plasminogen activator deletion and associated fibrin(ogen) deposition on macrophage localization in peritoneal inflammation.

There are two plasminogen activators (PAs), urokinase type-PA (u-PA) and tissue type-PA (t-PA). While u-PA is considered to be involved in cellular migration and tissue remodeling and t-PA in fibrinolysis, this distinction is not always clear-cut. With the use of u-PA and t-PA gene deficient mice (u-PA-/- and t-PA-/- mice, respectively) we have assessed the role of each PA in acute peritonitis. The cellular infiltrate in both thioglycolate- and antigen-induced peritoneal exudates was unaffected in u-PA-/- mice; in contrast, in t-PA-/- mice, the macrophage numbers, particularly of the Mac-1(hi)

Role of cannabinoid receptors in inhibiting macrophage costimulatory activity.

Delta(9)-tetrahydrocannabinol (THC) inhibits several immunologic functions of macrophages. THC's impact on peritoneal macrophages to deliver costimulatory signals to a helper T cell hybridoma was investigated by T cell interleukin-2 production stimulated with immobilized anti-CD3 antibody. The drug's inhibition of costimulatory activity depended on the macrophages. THC decreased costimulation provided by peritoneal cells elicited with polystyrene beads and thioglycollate, but the drug had no influence with macrophages elicited with thioglycollate alone. Bead administration induced CB2 mRNA exp

BMS-813160: A Potent CCR2 and CCR5 Dual Antagonist Selected as a Clinical Candidate.

BMS-813160 (compound 3) was identified as a potent and selective CCR2/5 dual antagonist. Compound 3 displayed good permeability at pH = 7.4 in PAMPA experiments and demonstrated excellent human liver microsome stability. Pharmacokinetic studies established that 3 had excellent oral bioavailability and exhibited low clearance in dog and cyno. Compound 3 was also studied in the mouse thioglycollate-induced peritonitis model, which confirmed its ability to inhibit the migration of inflammatory monocytes and macrophages. As a result of this profile, compound 3 was selected as a clinical candidate.

[Study on the estrogen derivative marked by CdTe quantum dots].

Water-soluble CdTe quantum dots (QDs) stabilized by thioglycolic acid (TGA) with high quantum yield were synthesized. And the small biomolecule 17beta-amino-estradiol was marked successfully by CdTe QDs. The analysis of UV-Vis spectroscopy, fluorescence spectroscopy, infrared spectroscopy and inverted microscope photographs showed that the surface carboxyl of CdTe quantum dots conjugated with the amino of 17beta-amino-estradiol through the activation of N-hydroxysuccinimide. 17beta-amino-estradiol marked by CdTe QDs successfully provided an experimental basis for new drug screening model.

Lesinurad for the treatment of hyperuricaemia in people with gout.

Gout is a common form of inflammatory arthritis caused by deposition of monosodium urate crystals. The central strategy for effective long-term management of gout is serum urate lowering. Current urate-lowering drugs include both xanthine oxidase inhibitors and uricosuric agents. Lesinurad is a URAT1 inhibitor that selectively inhibits urate rebsorption at the proximal renal tubule. Lesinurad 200mg daily in combination with a xanthine oxidase is approved for urate-lowering therapy in patients with gout. Areas covered: The published literature was searched using Pubmed and additional informatio

Norepinephrine and its metabolites are involved in the synthesis of neuromelanin derived from the locus coeruleus.

In order to elucidate the chemical structure of black to brown pigments, neuromelanins (NMs), in the substantia nigra (SN) and the locus coeruleus (LC) in the central nervous system of humans and other mammalian species during aging, chemical degradative methods are powerful tools. HPLC analysis after hydroiodic acid hydrolysis detected aminohydroxyphenylethylamines, aminohydroxyphenylacetic acids, and aminohydroxyethylbenzenes, which confirmed that SN-NM and LC-NM contain melanin derived not only from dopamine and norepinephrine (NE) but also from several other catecholic metabolites, such as
