[Tissue engineering of vascularized myocardial prosthetic tissue. Biological and solid matrices].

Tissue engineering of bioartificial myocardial tissue will become an increasingly important therapeutic approach in the near future but supply of oxygen and nutrients as well as evacuation of metabolic products represent a critical obstacle in tissues with a thickness of 100 µm and above. Viability of seeded cells in the myocardial patch is positively correlated with its function and thus early sufficient vascularization is mandatory. The choice of substrate, structure of matrices, specific cellular seeding and addition of growth factors contribute to this necessary vascularization process.Thi…

Surface-directed engineering of tissue anisotropy in microphysiological models of musculoskeletal tissue.

Here, we present an approach to model and adapt the mechanical regulation of morphogenesis that uses contractile cells as sculptors of engineered tissue anisotropy in vitro. Our method uses heterobifunctional cross-linkers to create mechanical boundary constraints that guide surface-directed sculpting of cell-laden extracellular matrix hydrogel constructs. Using this approach, we engineered linearly aligned tissues with structural and mechanical anisotropy. A multiscale in silico model of the sculpting process was developed to reveal that cell contractility increases as a function of principal

Demineralization of Camel Dentin for Dental Tissue Engineering.

BACKGROUND: Demineralisation of dentin effectively removes the crystalline hydroxyapatite phase while preserving the collagen matrix and associated bioactive proteins. This process results in an osteoinductive scaffold with favourable biological properties, making demineralised dentin a promising naturally derived biomaterial for dentoalveolar bone and orthopaedic bone tissue regeneration. OBJECTIVE: This study aimed to evaluate the structural and elemental characteristics of camel dentin matrix after demineralisation using ethylenediaminetetraacetic acid (EDTA) and citric acid (CA) at variou

Harnessing Starch for Next-Generation Corneal Tissue Engineering.

Corneal transplantation is the gold-standard treatment for end-stage corneal disease. However, its clinical use is limited by the global shortage of donor tissue, risks of immune rejection, and postoperative complications. Corneal tissue engineering (CTE) has emerged as a promising alternative strategy, focusing on the development of biocompatible scaffolds that support cellular regeneration while maintaining the critical optical clarity and biomechanical properties of the native cornea. Starch (ST), a naturally derived and abundant polysaccharide, has garnered significant interest as a biomat…

Specific tissue engineering for temporomandibular joint disc perforation.

BACKGROUND: The temporomandibular joint (TMJ) disc is a critical fibrocartilaginous structure with limited regenerative capacity in the oral system. Perforation of the TMJ disc can lead to osteoarthritis and ankylosis of the TMJ because of the lack of disc protection. Clinical treatments for TMJ disc perforation, such as discectomy, hyaluronic acid injection, endoscopic surgery and high position arthroplasty of TMJ, are questionable with regard to long-term outcomes, and only three fourths of TMJ disc perforations are repairable by surgery, even in the short-term. Tissue engineering offers the…

Bone Mineralization in Electrospun-Based Bone Tissue Engineering.

Increasing the demand for bone substitutes in the management of bone fractures, including osteoporotic fractures, makes bone tissue engineering (BTE) an ideal strategy for solving the constant shortage of bone grafts. Electrospun-based scaffolds have gained popularity in BTE because of their unique features, such as high porosity, a large surface-area-to-volume ratio, and their structural similarity to the native bone extracellular matrix (ECM). To imitate native bone mineralization through which bone minerals are deposited onto the bone matrix, a simple but robust post-treatment using a simul…
