Presence and effects of melatonin in Trypanosoma cruzi.

The unicellular organism Trypanosoma cruzi is an eukaryote whose cell cycle mainly occurs under darkness in the insect gut. The unique external phase corresponds to the metacyclic forms, the forms that are able to infect humans, which appear within the insect deyections. Thus, light may be a powerful stressor in this unicell. Epimastigote forms (the parasite forms that grow and transform to metacyclic forms in the insect gut) of Trypanosoma cruzi grow normally when cultured in a LD cycle of 0:24 hr, reaching exponential growth by the 7th day. A pulse of 2 hr of light (LD 2:22) was enough to bl…

The interaction of Trypanosoma congolense with endothelial cells.

Factors which affect adhesion of cultured Trypanosoma congolense bloodstream forms to mammalian feeder cells have been examined. Using an in vitro binding assay, the initial events following interaction of trypanosomes with bovine aorta endothelial (BAE) cells were monitored by both light- and electron microscopy. Metabolic inhibitors and other biochemicals were incubated with either cells or parasites, to test whether any inhibited the process. Our findings suggest that adhesion of the parasites is an active process requiring metabolic energy from the trypanosomes, but not from endothelial ce…

[Polyneuritis and myositis in Trypanosoma gambiense infection].

During a four-week trip to Nigeria a 54-year-old German developed a fever of 39 degrees C. Later on he had lymphadenopathy, pretibial oedema … ught to be due to polyneuritis and myositis. At this stage blood smear and CSF examination revealed Trypanosoma. He thereupon received suramin (1.0 g) and prednisolone (120 mg down to 40 mg) daily, to which melarsoprol was added after 6 days (0.5 ml up to 5.0 ml daily for 36 days). Almost all symptoms then regressed within 6 weeks.

Morphological changes of Trypanosoma vivax in mice.

Morphological changes were observed in the blood forms of Trypanosoma vivax strain Y486 in mice on Days 12-13 of infection, following the peak parasitaemia. During this period elongate trypomastigotes, 25-40 micron long, were observed, most showing an anterior movement of the kinetoplast towards the nucleus and some having a blunt posterior end. In a few parasites a complete transformation to the epimastigote form, 40-42 micron long, was observed. Small sphaeromastigotes were also present, especially in the fine capillaries of various organs. Smears and electron microscopy suggested the presen…

Effect of theophylline on differentiation of Trypanosoma brucei.

Differentiation of Trypanosoma brucei in the mammal limits the degree of parasitemia and prepares the trypanosome for passage back into the tsetse fly. In an attempt to define the signals that control differentiation, we found that theophylline, in contrast to indomethacin, blocked differentiation, prolonged parasitemia, elevated prostaglandin and cyclic AMP concentrations of rat plasma, and depressed intratrypanosomal cyclic AMP. Relatively nontoxic drugs that alter differentiation are powerful tools for elucidating the events that control this important process.

Mitogenic activity of an extract of Trypanosoma gambiense.

An extract obtained by repeated freeze/thawing of a freshly isolated strain of Trypanosoma gambiense caused stimulation of lymphocytes obtained from healthy adult donors and from cord blood. The active factor had some stimulatory effect on purified B and T lymphocytes and in two of five subjects increased IgM production by lymphocytes cultured in vitro. A trypanosome-derived mitogenic factor may play some part in the pathogenesis of the hypergammaglobulinaemia characteristic of African trypanosomiasis.

Bioinformatics and expression analysis of the Xeroderma Pigmentosum complementation group C (XPC) of Trypanosoma evansi in Trypanosoma cruzi cells.

Nucleotide excision repair (NER) acts repairing damages in DNA, such as lesions caused by cisplatin. Xeroderma Pigmentosum complementation group C (XPC) protein is involved in recognition of global genome DNA damages during NER (GG-NER) and it has been studied in different organisms due to its importance in other cellular processes. In this work, we studied NER proteins in Trypanosoma cruzi and Trypanosoma evansi, parasites of humans and animals respectively. We performed three-dimensional models of XPC proteins from T. cruzi and T. evansi and observed few structural differences between these …

Infestation of Mauritia flexuosa palms by triatomines (Hemiptera: Reduviidae), vectors of Trypanosoma cruzi and Trypanosoma rangeli in the Brazilian savanna.

To determine the infestation and trypanosome infection of triatomines captured in Mauritia flexuosa palm trees across its geographic distrib … bai (n=1) occurred in only one location. From 537 R. neglectus examined, 44 were infected (8%) with Trypanosoma rangeli and/or Trypanosoma cruzi (Tc Id). M. flexuosa was previously recognized as a suitable breeding ecotope for R. neglectus in the Brazilian states of Minas Gerais, Goiás, Tocantins and the Federal District. Our results expand this distribution …

Response of Trypanosoma vivax and Trypanosoma congolense in zebu cattle in North Cameroon to prophylactic treatment with two formulations of isometamidium.

We tested the efficacy of two formulations of isometamidium in a tsetse-infested farm in North Cameroon from 20 August 2000 to 5 January 2001. A total of 90 adult cattle were used in three groups of 30 each corresponding to two treated and one untreated control. Drug efficacies were evaluated in terms of reduction of parasite incidence in the host's blood, maintenance of packed-cell volume (PCV) and weight gains. Both drugs reduced the incidence of parasites even though re-infections 2 weeks after treatment were common. PCV values were similar in both treated groups but higher than in the untr

Cure of Trypanosoma brucei brucei and Trypanosoma brucei rhodesiense infections in mice with an irreversible inhibitor of S-adenosylmethionine decarboxylase.

A structural analog, 5'-([(Z)-4-amino-2-butenyl]methylamino)-5'-deoxy adenosine (MDL 73811), of decarboxy S-adenosyl-L-methionine, the product of the reaction catalyzed by S-adenosyl-L-methionine (AdoMet) decarboxylase (DC), was found to inhibit Trypanosoma brucei brucei AdoMet DC. The inhibition was time dependent (tau 50, 0.3 min), exhibited pseudo-first-order kinetics (Ki, 1.5 microM), and was apparently irreversible. The natural substrate of the reaction, AdoMet, protected the enzyme from inactivation, suggesting that MDL 73811 was directed at the enzyme active site and was probably cataly…

Co-infection with Trypanosoma congolense and Trypanosoma brucei is a significant risk factor for cerebral trypanosomosis in the equid population of the Gambia.

Trypanosomosis is a major cause of morbidity and mortality in working equids in The Gambia. Recently, a progressive, severe neurological syndrome characterised by a diffuse lymphoplasmacytic meningoencephalitis has been identified and associated with Trypanosoma brucei infection of the central nervous system. The pathogenesis of cerebral trypanosomosis is unclear and the clinical syndrome not well described. This observational cross-sectional study aimed to identify host and parasite related risk factors associated with the development of cerebral trypanosomosis and to describe the neurologica…

Observation on drug resistance of Trypanosoma (Nannomonas) congolense and Trypanosoma (Duttonella) vivax in cattle at a feedlot in the Northern Ivory Coast.

Studies carried out on feedlot cattle show Berenil at 7 mg/kg to be the treatment of choice in trypanosomiasis although repeated doses may be necessary. Resistant strains of Trypanosoma (Duttonella) vivax and Trypanosomas (Nannomonas) congolense survived dosages of between 0.5 mg/kg and 1.0 mg/kg of Trypamidium and were cross resistant to Homidium bromide.

Trypanosoma evansi infection in Tunisia: current situation.

Surra is a vector-borne disease, caused by a flagellate protozoan, Trypanosoma evansi, infecting all domestic mammals, including herbivores and dogs, and, very rarely, humans. In Tunisia, it affects mainly dromedaries (Camelus dromedarius) in the southern part of the country, causing heavy economic losses due to high morbidity, abortions and mortality. Trypanosoma evansi is mainly transmitted by mechanical vectors (Stomoxyine flies and tabanids), but also vertically, orally (to carnivores) and iatrogenically. In the present paper, we review and discuss the studies published on surra in Tunisia…

Global Protein Interaction Network for Trypanosoma cruzi.

Trypanosoma cruzi, the causative agent of Chagas disease, poses a significant health challenge due to limited therapeutic options and an incomplete understanding of its biology. Approximately half of the genome encodes hypothetical proteins with unknown functions, underscoring the need for systematic functional annotation. Protein-protein interactions (PPIs) underpin essential cellular processes, yet no large-scale PPI map has been developed for T. cruzi ─a critical gap that impedes both functional annotation of its proteome and drug discovery. This study presents the first comprehensive PPI n…
