Molecular detection of Sodalis glossinidius, Spiroplasma and Wolbachia endosymbionts in wild population of tsetse flies collected in Cameroon, Chad and Nigeria.

Background Tsetse flies are cyclical vectors of African trypanosomiasis. They have established symbiotic associations with different bacteria, which influence certain aspects of their physiology. The vector competence of tsetse flies for different trypanosome species is highly variable and is suggested to be affected by various factors, amongst which are bacterial endosymbionts. Symbiotic interactions may provide an avenue for the disease control. The current study provided the prevalence of 3 tsetse symbionts in Glossina species from Cameroon, Chad and Nigeria. Results Tsetse flies were colle…

'Where are the dead flies!': perceptions of local communities towards the deployment of Tiny Targets to control tsetse in the Democratic Republic of the Congo.

The National Programme for the control of human African trypanosomiasis in Democratic Republic of Congo includes a large-scale vector control operation using Tiny Targets. These are small panels of insecticide-impregnated cloth that are deployed in riverine habitat where tsetse flies concentrate. The effectiveness of Tiny Targets depends partly on acceptance by local communities. In 2018, we conducted research to explore the perception and acceptability of Tiny Targets in two different village clusters where Tiny Targets had been deployed by the local community or external teams. We conducted …

Extrachromosomal inheritance of susceptibility to trypanosome infection in tsetse flies. I. Selection of susceptible and refractory lines of Glossina morsitans morsitans.

Differences in susceptibility to infection with Trypanosoma congolense between F1 families of Glossina morsitans morsitans indicated that susceptibility is maternally inherited in this species of tsetse fly. Twelve F1 families, six selected for susceptibility and six selected for refractoriness to infection, have been bred for up to 13 generations. The reciprocal differences demonstrated in the F1 generation persisted in these selected families over many generations, indicating that susceptibility/refractoriness to T. congolense infection is extrachromosomally inherited in G. m. morsitans. Parasitological investigation of bovine Trypanosomosis, vector distribution and tsetse flies infection rate study, Dabo Hana District, Buno Bedelle Zone, Southwest Ethiopia.

A cross-sectional study was conducted to explore the prevalence of Trypanosome infections in cattle and within the tsetse flies from December 2020 to May 2021 in Dabo Hana district, Buno Bedelle Zone, Southwest Ethiopia. A total of 415 blood samples were examined utilizing Buffy coat and Giemsa-stained thin blood smear techniques. Vector distribution and tsetse fly infection rate were studied by deploying 60 traps in four purposively chosen villages of the district. The prevalence of Trypanosomes was 10.6% and 6.5% in cattle and in tsetse flies, respectively. Trypanosoma congolense (59.1%) in …

Trypanosomes infection, endosymbionts, and host preferences in tsetse flies (Glossina spp.) collected from Akagera park region, Rwanda: A correlational xenomonitoring study.

Akagera National Park and its surroundings are home to tsetse flies and a number of their mammalian hosts in Rwanda. A One-health approach is being used in the control and surveillance of both animal and human trypanosomosis in Rwanda. Determination of the infection level in tsetse flies, species of trypanosomes circulating in vectors, the source of tsetse blood meal and endosymbionts is crucial in understanding the epidemiology of the disease in animals and humans in the region. Tsetse flies (n = 1101), comprising Glossina pallidipes (n = 771) and Glossina morsitans centralis (n = 330) were c…

A machine learning approach to integrating genetic and ecological data in tsetse flies (Glossina pallidipes) for spatially explicit vector control planning.

Vector control is an effective strategy for reducing vector-borne disease transmission, but requires knowledge of vector habitat use and dispersal patterns. Our goal was to improve this knowledge for the tsetse species Glossina pallidipes, a vector of human and animal African trypanosomiasis, which are diseases that pose serious health and socioeconomic burdens across sub-Saharan Africa. We used random forest regression to (i) build and integrate models of G. pallidipes habitat suitability and genetic connectivity across Kenya and northern Tanzania and (ii) provide novel vector control recomme

Tripartite interactions between tsetse flies, Sodalis glossinidius and trypanosomes--an epidemiological approach in two historical human African trypanosomiasis foci in Cameroon.

Epidemiological surveys were conducted in two historical human African trypanosomiasis foci in South Cameroon, Bipindi and Campo. In each focus, three sampling areas were defined. In Bipindi, only Glossina palpalis was identified, whereas four species were identified in Campo, G. palpalis being highly predominant (93%). For further analyses, 75 flies were randomly chosen among the flies trapped in each of the six villages. Large and statistically significant differences were recorded between both (1) the prevalence of Sodalis glossinidius (tsetse symbiont) and the prevalence of trypanosome inf

Protecting cows in small holder farms in East Africa from tsetse flies by mimicking the odor profile of a non-host bovid.

BACKGROUND: For the first time, differential attraction of pathogen vectors to vertebrate animals is investigated for novel repellents which when applied to preferred host animals turn them into non-hosts thereby providing a new paradigm for innovative vector control. For effectively controlling tsetse flies (Glossina spp.), vectors of African trypanosomosis, causing nagana, repellents more powerful than plant derived, from a non-host animal the waterbuck, Kobus ellipsiprymnus defassa, have recently been identified. Here we investigate these repellents in the field to protect cattle from nagan…

Molecular identification of diminazene aceturate resistant trypanosomes in tsetse flies from Yoko in the Centre region of Cameroon and its epidemiological implications.

African animal trypanosomiases are caused by trypanosomes cyclically or mechanically transmitted by tsetse and other biting flies. Although molecular tools have been developed to identify drug-resistant trypanosomes in mammals, little or no investigation on drug-resistance has been undertaken on trypanosomes harbored by tsetse flies. Moreover, no data on mechanical vectors of African trypanosomes is available in most endemic areas of Cameroon. This study was designed to update our knowledge on the cyclical and mechanical vectors of African trypanosomes, and using molecular tools to identify di…

Factors affecting the propensity of tsetse flies to enter houses and attack humans inside: increased risk of sleeping sickness in warmer climates.

BACKGROUND: Sleeping sickness, or human African trypanosomiasis, is caused by two species of Trypanosoma brucei that are transmitted to humans by tsetse flies (Glossina spp.) when these insects take a bloodmeal. It is commonly assumed that humans must enter the normal woodland habitat of the flies to become infected, but recent studies found that tsetse frequently attack humans inside buildings. Factors affecting human/tsetse contact in buildings need identification. METHODOLOGY/PRINCIPAL FINDINGS: In Zimbabwe, tsetse were allowed access to a house via an open door. Those in the house at suns…

Successful application of deltamethrin pour on to cattle in a campaign against tsetse flies (Glossina spp.) in the pastoral zone of Samorogouan, Burkina Faso.

1,500-2,000 head of cattle were treated with deltamethrin 1% Spot On in an area of high tsetse densities, notably of Glossina morsitans submorsitans. After four treatments at monthly intervals, the time between two treatments was increased to two months. 11 months after the commencement of the campaign the fly population had decreased from initially 54.2 flies/trap/day to densities varying between 0.06-2.0 flies/trap/day, mostly G. palpalis gambiensis. Blood-meal analysis showed that this species was surviving in limited areas, mainly feeding on monitor lizards; consequently it is unlikely tha

The effect of isometamidium chloride on insect forms of drug-sensitive and drug-resistant stocks of Trypanosoma vivax: studies in vitro and in tsetse flies.

Isometamidium chloride-resistant and -sensitive Trypanosoma vivax insect forms were continuously propagated in vitro without feeder-layer cells in a semi-defined liquid medium at 27 degrees C. The effect of isometamidium chloride (Samorin) on T. vivax was assessed by monitoring the viability of epimastigotes and the production of metacyclic forms. Populations of insect forms of T. vivax stock IL 1392 and clone IL 3185 showed reduced growth and died after 10 days when cultivated in the presence of 1 ng/ml isometamidium chloride and after 6 days in the presence of 10 ng/ml. Assessment of α-Cypermethrin Pour-On Application and Diminazene Aceturate for Treating Trypanosome-Related Diseases Caused by Tsetse Flies on Cattle in Mô, Togo.

The effects of tsetse-transmitted trypanosomosis control in high tsetse flies (Glossina spp.) challenge and trypanocidal drug resistance settings remain poorly understood in Togo owing to poor data coverage on the current disease impact. From March 2014 to November 2017, a database of zoo-sanitary surveys integrating the evolution of disease incidence and intervention coverage made it possible to quantify the apparent effects attributable to the control effort, focused on all sedentary cattle breeds in the 1,000 km² area of Mô in Togo. The strategy involved an initial phase with cross-sectiona…
