The virology and developments toward control of human enterovirus 71.

Enterovirus 71 (EV71), a member of the Enterovirus genus in the Picornaviridae family, was first recognized as a dermotrophic virus that usually cause mild, self-limiting hand-foot-and-mouth disease (HFMD). However, EV71 infection can sometimes induce a variety of severe neurological complications and even death. Current large outbreaks of EV71 make this virus being a major public health issue. Intense effort has been made to address its underlying pathogenesis and to develop effective means for combating EV71 infections. Here, we aimed to provide an overview of cellular mechanisms underlying

HIV virology and pathogenetic mechanisms of infection: a brief overview.

Studies on HIV virology and pathogenesis address the complex mechanisms that result in the HIV infection of the cell and destruction of the immune system. These studies are focused on both the structure and the replication characteristics of HIV and on the interaction of the virus with the host. Continuous updating of knowledge on structure, variability and replication of HIV, as well as the characteristics of the host immune response, are essential to refine virological and immunological mechanisms associated with the viral infection and allow us to identify key molecules in the virus life cy…

Constantin Levaditi (1874-1953): a pioneer in Immunology and Virology.

The eminent doctor Constantin Levaditi represents one of the most important researchers in the field of medicine in the 20th century. Although he was engaged in many areas of the rapidly growing field of immunology, his name is associated mainly with research in poliomyelitis. His laboratory research contributed decisively to the clarification of the epidemiology of this dreadful disease that claimed thousands of victims. Moreover, his experimental work constituted the basis for the development of the vaccine against poliomyelitis, initially in 1955 by Jonas Salk (1914-95) using inactivated vi

Virology of infantile chronic recurrent parotitis in Santiago de Chile.

Infantile chronic recurrent parotitis (ICRP) has been attributed to multiple causes, including viral infections, and therefore its treatment remains empirical. Our aim was to evaluate the involvement of respiratory and oropharyngeal viruses in acute episodes of ICRP. Seventy children were studied, 50 patients and 20 age-matched controls, in a 2-year follow-up study. Saliva samples were taken from the parotid duct and analyzed by viral isolation and immunofluorescence for adenovirus (Ad), respiratory sincitial virus (RSV), parainfluenza virus (PI), influenza virus (Flu), Cytomegalovirus (CMV),

The Laboratory Virology and Serology Reporting Scheme, 1991 to 2000.

Between 1991 and 2000, the Laboratory Virology and Serology Surveillance Scheme (LabVISE) received 340,730 laboratory reports of viral and non-viral pathogen identifications. In this report, data on 136 viruses and 31 non-viral pathogens is analysed. The age and sex distribution and seasonal fluctuations in infections are described. The major clinical diseases associated with LabVISE pathogens are reviewed together with a survey of recent activity reported in Australia. The contribution of LabVISE over the 10-year period to surveillance of poliomyelitis in Australia, up to and beyond the eradi…

A combined pseudoreplica-immunochemical technique for research and diagnostic virology.

Pseudoreplica and immunochemical techniques were combined in a single protocol for identification of virus in research and clinical specimens. Stock preparations of vesicular stomatitis virus (VSV), cytomegalovirus (CMV), and herpes simplex virus (HSV) were used to develop the technique. Traditional pseudoreplicas of viral stock solutions were prepared but not negatively stained. The virus was then immunolabeled in two stages. Virus-specific polyclonal antisera were used in the first stage; colloidal gold conjugated antibodies were used as indicator antibody in the second stage. After immunola

[Virologic diagnosis of meningoencephalitis].

Many viruses can be responsible for meningoencephalitis. The principal ones are listed and the main features of their virological diagnosis are described. It must be borne in mind that a virological diagnosis must always be guided by the clinical context and that viral meningoencephalitis, relatively uncommon in Europe, is much more frequent in tropical countries.

Crimean Congo Hemorrhagic Fever Virus for Clinicians-Virology, Pathogenesis, and Pathology.

Crimean-Congo hemorrhagic fever (CCHF), caused by CCHF virus, is a tickborne disease that can cause a range of illness outcomes, from asymptomatic infection to fatal viral hemorrhagic fever; the disease has been described in >30 countries. We conducted a literature review to provide an overview of the virology, pathogenesis, and pathology of CCHF for clinicians. The virus life cycle and molecular interactions are complex and not fully described. Although pathogenesis and immunobiology are not yet fully understood, it is clear that multiple processes contribute to viral entry, replication, and …

Coronavirus Infections in Companion Animals: Virology, Epidemiology, Clinical and Pathologic Features.

Coronaviruses are enveloped RNA viruses capable of causing respiratory, enteric, or systemic diseases in a variety of mammalian hosts that vary in clinical severity from subclinical to fatal. The host range and tissue tropism are largely determined by the coronaviral spike protein, which initiates cellular infection by promoting fusion of the viral and host cell membranes. Companion animal coronaviruses responsible for causing enteric infection include feline enteric coronavirus, ferret enteric coronavirus, canine enteric coronavirus, equine coronavirus, and alpaca enteric coronavirus, while c

Epidemiological Surveillance of Viral Hemorrhagic Fevers With Emphasis on Clinical Virology.

This article will outline surveillance approaches for viral hemorrhagic fevers. Specific methods for surveillance of clinical samples will be emphasized. Separate articles will describe methods for surveillance of rodent-borne viruses (roboviruses) and arthropod-borne viruses (arboviruses). Since the appearance of hantaviruses and arenaviruses in the Americas, more than 30 different species in each group have been established, and therefore they have become the most frequently emerging viruses. Flaviviruses such as yellow fever and dengue viruses, although easier to recognize, are also more wi

Diagnostic virology and patient care: from vaguely interesting to vitally important.

The existence of pathogenic viruses was inferred by experiments at the turn of the twentieth century. Key developments in detection of viruses, including electron microscopy and monolayer cell culture, were made in the middle of that century. However, in terms of patient care, the results from the virology laboratory often arrived the patient was 'better or dead'. The advent of molecular techniques, particularly polymerase chain reaction and more recently whole genome sequencing made timely and accurate diagnosis of viral infections feasible. A range of approaches have been taken to identify a…

Evolution in Our Understanding of Hepatitis B Virus Virology and Immunology.

Hepatitis B virus (HBV) infection is a major global health challenge. HBV can cause significant morbidity and mortality by establishing acute and chronic hepatitis. Approximately 250 million people worldwide are chronically infected, and more than 2 billion people have been exposed to HBV. Since the discovery of HBV, the advances in our understanding of HBV virology and immunology have translated into effective vaccines and therapies for HBV infection. Although current therapies successfully suppress viral replication but rarely succeed in viral eradication, recent discoveries in HBV virology …

Molecular virology of hepatitis B virus and targets for antiviral intervention.

The members of the viral family Hepadnaviridae comprise one of the smallest enveloped DNA viruses and cause acute and chronic infections in mammals and birds, leading to large virus and antigen loads in the blood. They have a restricted host range and depend on differentiated hepatocytes for replication. Hepatitis B virus (HBV) is the prototype of the Hepadnaviridae. HBV can persist in infected hepatocytes and has evolved elaborate strategies to evade the immune system. HBV replicates like HIV (family of Retroviridae) via reverse transcription. Drugs licensed for inhibition of HIV reverse tran
