INTRODUCTION: Xanthine oxidase (XO) is a molybdoflavoprotein that catalyzes the oxidative hydroxylation of purines to produce uric acid and reactive oxygen species. These reaction products can cause severe disease conditions like hyperuricemia which makes XO enzyme, an important therapeutic target in diseases like gout.
AREAS COVERED: Herein, patents from 2015 to 2020 are discussed to disclose the synthetic, as well as natural compounds, claimed to inhibit XO enzyme. The article also presents the last five years of clinical progression of some prominent XO inhibitors.

A 30-month-old girl presented with gastrointestinal symptoms and a febrile urinary tract infection. Sonographic and radiographic imaging demonstrated left hydronephrosis due to a radiolucent ureteral stone. Surgical exploration identified an aberrant, patent left umbilical artery causing ureteral obstruction. Stone and urine analyses revealed hereditary xanthinuria that had not previously been recognized in this child and her family.

Xanthine oxidase catalyzes the biotransformation of many drugs, including the thiopurines and methyl-xanthines. Relaxant effects and adenosine-antagonism of 3-propyl-xanthine (enprofylline) and 10 different methyl-xanthines were examined in isolated guinea-pig tracheas. The chemical structural requirements for tracheal relaxation were found to be different from those for adenosine antagonism by the xanthine derivatives. The xanthine oxidase (XOD) inhibitory activity of propolis from China and Brazil was measured. The propolis from both place were seen to have XOD inhibitory activity. However, a stronger tendency was shown in the propolis from China. The compounds in each the propolis were measured quantitatively. Precise localization of xanthine oxidase activity might elucidate physiological functions of the enzyme, which have not been established so far.
