MeMLO12 Impairs Cassava Resistance to Xanthomonas axonopodis pv. manihotis by Repressing Reactive Oxygen Species, Callose Deposition, and Defense Hormone Pathways.

Cassava (Manihot esculenta Crantz) is a vital food and energy crop in tropical regions, yet its yield is significantly threatened by cassava bacterial blight, caused by Xanthomonas axonopodis pv. manihotis (Xpm). Mildew resistance locus O (MLO) genes are known negative regulators of plant immunity, but their roles in cassava disease susceptibility remain unclear. In this study, we identified the MeMLO12 gene from cassava and investigated its function in response to Xpm infection. Subcellular localization revealed that MeMLO12 is specifically localized in the nucleus. Functional analysis via tr…

NIT24 and NIT29-Dependent Auxin Biosynthesis Promotes Virulence and Auxin-SA Antagonism in Rice During Xanthomonas oryzae pv. oryzae Infection.

Auxin production by plant-associated bacteria is increasingly recognized as a determinant of host colonization, yet its integration into virulence networks remains incompletely understood. Nitrilase-dependent auxin (IAA) biosynthesis contributes to virulence in Xanthomonas oryzae pv. oryzicola (Xoc), but whether this mechanism operates in X. oryzae pv. oryzae (Xoo) has remained unclear. Here, we identify two conserved nitrilases, NIT24 and NIT29, in the Xoo strain PXO99A and examine their contributions to IAA production and virulence. Deletion of either gene reduces IAA accumulation, and under…

Overexpressing GH3.1 and GH3.1L reduces susceptibility to Xanthomonas citri subsp. citri by repressing auxin signaling in citrus (Citrus sinensis Osbeck).

The auxin early response gene Gretchen Hagen3 (GH3) plays dual roles in plant development and responses to biotic or abiotic stress. It functions in regulating hormone homeostasis through the conjugation of free auxin to amino acids. In citrus, GH3.1 and GH3.1L play important roles in responding to Xanthomonas citri subsp. citri (Xcc). Here, in Wanjingcheng orange (Citrus sinensis Osbeck), the overexpression of CsGH3.1 and CsGH3.1L caused increased branching and drooping dwarfism, as well as smaller, thinner and upward curling leaves compared with wild-type. Hormone determinations showed that …

In vitro susceptibility of 33 clinical case isolates of Xanthomonas maltophilia. Inconsistent correlation of agar dilution and of disk diffusion test results.

Xanthomonas maltophilia is an emerging nosocomial pathogen, possibly selected by a changing antimicrobial usage and patient population. In the present study, we tested the susceptibility of 33 recent clinical case isolates to 12 commonly employed antimicrobials. Trimethoprim-sulfamethoxazole (1:19 ratio) and doxycycline were uniformly the most active agents; ciprofloxacin and fleroxacin were slightly less active and, along with tetracycline and ceftazidime, more variable in their potency. Interestingly, the disk diffusion method routinely overstated the activity of ciprofloxacin (12% very majo…

The RpfF Acyl-ACP thioesterase of major rice pathogen Xanthomonas oryzae pv. oryzae is essential for maintenance of membrane function and virulence.

In Xanthomonas, RpfF is a bifunctional enzyme having both enoyl-ACP dehydratase and acyl-ACP thioesterase activities catalyzed by a single active site. While the hydratase activity is essential for producing quorum-sensing signals of the diffusible signal factor (DSF) family, the physiological role of the thioesterase activity remained unclear. Singh and coworkers have reported that deletion of the gene encoding RpfF (a ∆rpfF strain) of the rice pathogen X. oryzae pv. oryzae (Xoo) resulted in loss of membrane integrity (Singh et al, 2022). We report that the loss in membrane integrity is due t…
