Nanotip acetylcholine biosensor reveals cholinergic differentiated SH-SY5Y cells release partial vesicle content during exocytosis.

Acetylcholine (ACh) is a central neurotransmitter in cognitive function, motor control, and synaptic modulation, yet its electrochemical inactivity and the rapid kinetics of exocytosis have hindered real-time quantal measurements. Micrometer-scale enzymatic ACh biosensors previously enabled sub-millisecond extracellular recordings but were too large for synaptic positioning and intracellular recordings. Here we present a short, ultrafast and low-noise amperometric ACh biosensor based on a needle-shaped carbon fiber nanotip electrode functionalized with gold nanoparticles and enzymes. The minia…

Pan-cancer expression, methylation, and prognostic significance of α7 nicotinic acetylcholine receptor in tumor immunology.

The α7 nicotinic acetylcholine receptor (α7nAChR), functioning as a critical ligand-gated ion channel in the nervous system, has recently been implicated to play a role in various non-neuronal tissues. Nonetheless, a comprehensive understanding of its expression patterns, mechanisms of epigenetic control, and functions within the tumor microenvironment (TME) across various malignancies is still lacking. To address this gap, we performed an extensive multi-cancer investigation of CHRNA7 spanning 31 tumor types, leveraging datasets from The Cancer Genome Atlas and Genotype-Tissue Expression data…

Development of a double-pharmacophore workflow for screening subtype-selective M3 muscarinic acetylcholine receptor antagonists.

M3 subtype of muscarinic acetylcholine receptors (mAChRs) is a very important drug target validated for irritable bowel syndrome (IBS), over active bladder (OAB), and chronic obstructive pulmonary disease (COPD). The M2 subtype has high sequence and structural similarity with M3. This makes it difficult to design selective inhibitors for M3 subtype. To address this problem, a three-dimensional quantitative structure-activity relationship (3D-QSAR) pharmacophore model was generated to estimate M2 specific inhibitory constant (Ki). The predictions were compared with a reported 3D-QSAR M3 pharmac…

Screening, synthesis and structural characterization of a novel bicyclic peptide inhibiting α7 nicotinic acetylcholine receptor.

Peptides targeting the α7 nicotinic acetylcholine receptor (α7-nAChR) represent promising candidates for therapeutic and chemical probe development. Here, we identified a novel bicyclic peptide KP1877 via phage-displayed peptide library screening followed by chemical synthesis of active analogs. Structural characterization reveals KP1877 adopts a unique bicyclic conformation distinct from known natural α7-nAChR-targeting peptides, and exerts potent inhibitory effect by stabilizing the closed, non-conducting state of the receptor via orthosteric binding.

Design, synthesis and preliminary evaluation of mosquitoicidal activity of arecoline derivatives targeting muscarinic acetylcholine receptors.

To develop novel mosquito control agents, we designed and synthesized a series of derivatives based on the natural alkaloid arecoline as a lead compound, targeting muscarinic acetylcholine receptors (mAChRs). Using computer-aided drug design and docking techniques focused on mAChRs, we designed piperidine derivatives and screened for synthetically accessible candidates. The compounds were synthesized, structurally characterized, and evaluated for activity via a Fluo-4 calcium fluorescence assay using Chinese Hamster Ovary-K1 (CHO-K1) cells expressing the muscarinic acetylcholine receptor M1. M…

A high-throughput screening platform for acetylcholinesterase inhibitors using a genetically encoded acetylcholine fluorescent sensor.

Acetylcholinesterase (AChE) is a crucial hydrolytic enzyme in the central nervous system, responsible for the rapid degradation of the neurotransmitter acetylcholine (ACh) in the synaptic cleft, thereby maintaining the balance between neuronal excitation and inhibition. AChE is not only the primary target of neurotoxic agents and organophosphorus pesticides but its aberrant activity is also closely associated with various neurodegenerative diseases such as Alzheimer's disease (AD) and myasthenia gravis. The efficient and rapid discovery and screening of AChE inhibitors hold urgent and signific…

Adult-onset hippocampal α7 nicotinic acetylcholine receptor loss rapidly drives Alzheimer-like neuropathology in mice.

INTRODUCTION: The reduction of α7 nicotinic acetylcholine receptor (nAChR) in the hippocampus con-stitutes one of the neuropathological features observed in the brains of individuals with Alzheimer's disease (AD), and α7 nAChR is crucial to cognitive process. However, studies involving α7 nAChR knockout mice have not demonstrated any abnormal brain structure or damage to hippocampal neurons, a discrepancy likely driven by widespread developmental compensatory signaling reprogramming triggered by permanent whole-body α7 nAChR deletion from embryonic stages. This limitation renders traditional k…

Alcohol-induced liver injury is mediated via α4-containing nicotinic acetylcholine receptors expressed in hepatocytes.

BACKGROUND: Our previous study demonstrated that alcohol induced the expression of the α4 subunit of nicotinic acetylcholine receptors (nAChRs) in the livers of wild type mice (WT), and that whole-body α4 nAChR knockout mice (α4KO) showed protection against alcohol-induced steatosis, inflammation, and injury. Based on these findings, we hypothesized that hepatocyte-specific α4 nAChRs may directly contribute to the detrimental effects of alcohol on the liver. METHODS: Hepatocyte-specific α4 knockout mice (α4HepKO) were generated, and the absence of α4 nAChR was confirmed through PCR of genomic…

Serum cystatin C as a potential biomarker for generalized acetylcholine receptor antibody-positive myasthenia gravis.

OBJECTIVE: To identify new metabolic biomarkers associated with myasthenia gravis (MG). METHODS: We analyzed 285 potential metabolic molecules from UK Biobank (UKB) for MG patients and identified elevated serum cystatin C (Cys-C). Validation was performed using laboratory data, ELISA, and clinical information from Chinese (CHN) acetylcholine receptor antibody (AChR-Ab) positive generalized MG (gMG) cohorts. We assessed cytokines/chemokines/complements and peripheral blood T lymphocytes using Luminex assays and flow cytometry. MG-relevant scores including myasthenia gravis activities of daily …

Decreased non-neurogenic acetylcholine in bone marrow triggers age-related defective stem/progenitor cell homing.

Age-related decline in the ability of bone marrow (BM) to recruit transplanted hematopoietic stem and progenitor cells (HSPCs) limits the potential of HSPC-based medicine. Using in vivo imaging and manipulation combined with integrative metabolomic analyses, we show that, with aging, degradation of non-neurogenic acetylcholine disrupts the local Chrm5-eNOS-nitric oxide signaling, reducing arterial dilation and decreasing both BM blood flow and sinusoidal wall shear stress. Consequently, aging BM microenvironment impairs transendothelial migration of transplanted HSPCs, and their BM homing effi…

Targeting α7 Nicotinic Acetylcholine Receptor for Modulating the Neuroinflammation of Dry Eye Disease Via Macrophages.

PURPOSE: Patients with dry eye disease (DED) often exhibit neurological abnormalities and may even suffer from neuropathic pain and pain-related anxiety or depression. The α-7 nicotinic acetylcholine receptor (α7nAChR) is a pivotal regulator in the anti-inflammatory pathway connecting the nervous and immune systems, Here, we investigate the potential of α7nAChR agonist as a novel treatment for DED. METHODS: We induced DED model by unilateral excision of extraorbital lachrymal gland in C57/BL6 mice. After seven days of treatment, RNA sequencing was performed to identify differentially expresse…
