Entamoeba histolytica is a protozoan parasite responsible for invasive intestinal and extraintestinal amebiasis. The pathology of amebiasis is still poorly understood, which can be largely attributed to lack of molecular tools. Here we present the optimization of SNAP-tag technology via codon optimization specific for E. histolytica. The resultant SNAP protein is highly expressed in amebic trophozoites, and shows proper localization when tagged with an endoplasmic reticulum retention signal. Amebiasis is a rare condition in industrialised countries but is epidemiologically growing. Clinical manifestations may range from asymptomatic to invasive disease. With the purpose to determine the duration of serum antibodies against E. histolytica by means of the contraimmunoelectrophoresis technique, 31 patients with the diagnosis of intestinal or liver invasive amebiasis were included in this study. A case report of a 36-year-old black homosexual man with right upper- and lower-quadrant abdominal pain is presented. When exploratory surgery for appendicitis was performed, an abscess, consistent with an amebic abscess, was encountered in the right lobe of the liver. We present a patient with an acute amebic liver abscess with nonreactive serologic tests. Motile hematophagous trophozoites of Entamoeba histolytica were seen microscopically in scrapings from the wall of the abscess.
