Ecological Correlates of Large-Scale Turnover in the Dominant Members of Pseudacris crucifer Skin Bacterial Communities.

Animals host a wide diversity of symbiotic microorganisms that contribute important functions to host health, and our knowledge of what drives variation in the composition of these complex communities continues to grow. Microbiome studies at larger spatial scales present opportunities to evaluate the contribution of large-scale factors to variation in the microbiome. We conducted a large-scale field study to assess variation in the bacterial symbiont communities on adult frog skin (Pseudacris crucifer), characterized using 16S rRNA gene amplicon sequencing. We found that skin bacterial communi

Modeling Progressive Pseudorheumatoid Arthropathy of Childhood: Disease Progression and Cartilage Pathology in CCN6 Knockout Sheep.

OBJECTIVE: Progressive Pseudorheumatoid Arthropathy of Childhood (PPAC), also known as Progressive Pseudorheumatoid Dysplasia (PPRD), is an autosomal recessive disorder caused by loss-of-function variants in Cellular Communication Network Factor 6 (CCN6, aka, WISP3). Patients with PPAC are asymptomatic at birth but develop polyarticular arthropathy in childhood, requiring multiple arthroplasties before 30 years of age. Currently, no therapy prevents or delays PPAC progression. Furthermore, how loss of CCN6 affects joint development or homeostasis is unknown due to the unavailability of pre-sym

Nutrition and gut-brain axis: opposing effects of dietary fiber and Western-style diets on Alzheimer's disease.

PURPOSE OF REVIEW: This review summarizes how diet shapes the gut-brain axis and contributes to Alzheimer's disease (AD) pathology, emphasizing the contrasting effects of Western-style diets and dietary fiber. RECENT FINDINGS: Western diets rich in sugar and saturated fat disrupt gut microbial balance, increase intestinal permeability, and promote systemic inflammation, oxidative stress, and lipid metabolic imbalance, all of which accelerate neurodegeneration. In contrast, dietary fiber supports microbial diversity, improves lipid and glucose metabolism, and reduces neuroinflammation through

Environmental Enrichment Exerts a Selective, Stage-Dependent Modulation on the Autism-like Behavioral Phenotype Induced by Embryonic Valproic Acid in Zebrafish.

Autism Spectrum Disorder (ASD) is characterized by deficits in communication, socialization, and restricted behavioral patterns. Valproic acid (VPA) is a teratogen that, through epigenetic mechanisms, induces ASD-like phenotypes in animal models. The zebrafish (Danio rerio) represents an ideal model for evaluating these alterations due to its complex behavioral repertoire. This study analyzed the longitudinal effect of an enriched environment (EE) on VPA-induced behavioral alterations in juvenile and adult stages (n = 72; n = 18 per group: CTRL-SE, CTRL-EE, VPA-SE, and VPA-EE). Subjects were e

Ingested microplastics and the gut-brain axis: A systematic review of mechanistic pathways and neurobiological impacts in human and preclinical models.

Miroplastic (MP) pollution which affects both humans and animals, has emerged as a global health concern. Evidence from in vivo rodent studies, in vitro human cell models, and multi-omics analyses indicates that ingested and inhaled MPs can accumulate in the gastrointestinal tract, cross epithelial barriers, and alter host-microbe interactions. Nanoplastics (NPs, <1 μm), owing to their small size and high surface-area-to-volume ratio, may exhibit greater cellular uptake, tissue translocation, and neurotoxic potential than larger MPs. These processes contribute to systemic effects, including im

Anesthetic-Induced Developmental Neurotoxicity: Cognitive Sequelae of Early Exposure.

Preclinical studies showing widespread neuronal apoptosis and long-term behavioral deficits when exposed to common anesthetics raised concerns for anesthetic-induced neurotoxicity in pediatric patients during periods of rapid synaptogenesis. N-methyl-D-aspartate receptor antagonists have been shown in animal studies to disrupt activity-dependent plasticity, hinder mitochondrial function, and induce epigenetic dysfunction. These studies have led to regulatory warnings and investigation into the potential long-term cognitive and neurodevelopmental consequences in children who have been exposed t

CHD8 dysregulation in neurodevelopment: Emerging insights into autism pathophysiology.

Autism is considered a multifaceted neurodevelopmental condition, marked by impairments in social interaction, communication, and repetitive behaviours. Recent genetic research has highlighted mutations in the CHD8 (Chromodomain Helicase DNA-binding Protein 8) gene as one of the most significant risk factors associated with ASD. CHD8 encodes a chromatin remodeler protein that regulates gene expression by altering chromatin accessibility. This review aims to comprehensively review current findings on CHD8 mutations, with a focus on the gene's role in brain development, synaptic function, and AS

A consensus roadmap for post-traumatic epilepsy: Clinical biomarkers, research priorities, policy barriers, and pathways to interventional trials.

Understanding the mechanisms underlying post-traumatic epilepsy (PTE) following traumatic brain injury (TBI), and developing strategies to prevent or modify its progression, has been the focus of large collaborative efforts within the epilepsy and TBI research communities for over a decade. However, to date, preclinical and clinical researchers with expertise in these areas have not formally convened to discuss ways to move the field forward. To enable communication and collaboration, scientific experts as well as individuals with lived experience of PTE were gathered during the inaugural Inte

Sex-specific placental adaptations associated with fetal growth restriction: a review.

Fetal growth restriction (FGR) is a major obstetric complication linked to placental insufficiency, increasing the risk of perinatal morbidity, mortality, and long-term health issues. The placenta is central to fetal development and maternal-fetal communication; its dysfunction leads to impaired fetal development. Numerous experimental FGR models exist, from rodents to large animals, but most studies, despite changes to some federal funding requirements, have historically overlooked the impact of fetal sex on placental development, function, and adaptation. Recent evidence indicates that male

Oral delivery of insulin via bitter melon-derived extracellular vesicles: a novel strategy for effective hypoglycemia.

Bitter melon is an edible crop, widely recognized for its hypoglycemic effects. Extracellular vesicles, as key signaling mediators produced by living cells, retain the bioactive components of their parent plants and possess the ability to traverse biological barriers to facilitate intercellular communication. On this basis, bitter melon-derived extracellular vesicles (BMEVs) were isolated. Subsequent mechanistic investigations revealed that BMEVs promote glycogen synthesis and glucose consumption via the Akt-FoxO1-PEPCK pathway. Subsequently, insulin (INS) was loaded into BMEVs to develop an o

Linking Ecosystem-Service Perceptions and Wool Valorisation in Alpine Sheep Farming Systems.

Wool is often regarded as a low-value by-product of Alpine sheep farming, despite its potential to convey the ecosystem services (ESs) generated by these systems. This study examines how awareness and perception of these services are associated with consumer attitudes towards locally produced wool in the Lanzo Valleys in Northwestern Italy. A convenience sample of 400 adults familiar with the study area completed an online questionnaire. Principal Component Analysis identified two orientations towards wool products-Sustainability Orientation and Product-Value Orientation-which together explain

Immune-brain plasticity underpins stress and affective behaviors.

The lifetime prevalence of mood disorders such as major depressive disorder (MDD) is thought to approach up to 50% of the world's population. Traditionally, research into the mechanisms of these disorders has focused on neurotransmission, but emerging evidence highlights neuroimmune interactions-the molecular signaling between immune and brain cells-as key regulators of brain plasticity, affective behavior, and potential vulnerability to mood disorders. Chronic stress models have unearthed how immune cell responses modify neural circuit activity, synaptic connectivity, and behaviors relevant f

The age-associated decline in neuroplasticity and its implications for post-stroke recovery in animal models of cerebral ischemia: The therapeutic role of extracellular vesicles.

Older individuals are typically more susceptible to stroke, and age-related differences in brain plasticity significantly affect recovery and treatment responses following cerebral ischemia and traumatic brain injury. Extracellular vesicles (EVs) have emerged as promising diagnostic and therapeutic tools due to their role in intercellular communication and ability to cross the blood-brain barrier. While EVs hold potential in promoting brain repair, their efficacy is influenced by donor age-those derived from young stem cells exhibit more regenerative profiles, whereas aged donor EVs may carry

Advances in mitochondria-nucleus crosstalk in septic cardiomyopathy.

Sepsis-induced cardiomyopathy (SICM), a critical contributor to the high mortality rate associated with sepsis, involves complex pathophysiological mechanisms that remain incompletely elucidated. In recent years, dysregulation of bidirectional signaling communication between mitochondria and the nucleus has been recognized as a pivotal factor in the pathogenesis of SICM. The anterograde signaling pathways-including the PGC-1α/NRF1/NRF2 axis, SIRT3-mediated deacetylation, and TFAM-dependent mitochondrial DNA (mtDNA) maintenance-are suppressed by inflammation and metabolic disturbances. Comparative assisted reproductive technologies in human and veterinary medicine: implications for reproductive success.

Assisted reproductive technologies (ART) have been widely and successfully used in both humans and livestock. However, only in humans and cattle have in vitro fertilization (IVF), in vitro embryo culture (IVC), and embryo transfer (ET) developed into large commercial sectors. The major differences between human and animal ART include the rationale of the treatment and the patient groups. While ART is used to treat infertility in humans, veterinary ART aims to maximize genetic gain and minimize generation intervals. Human ART is filled with societal, cultural, and emotional challenges, whereas

Intracellular and extracellular redox signals during exercise and aging.

Regular physical activity enhances systemic health and resilience, partly through the generation of reactive oxygen species (ROS) that serve as key modulators of redox-sensitive signaling pathways. This review explores how redox signaling mediates both local and systemic responses to exercise, with particular focus on skeletal muscle and aging. We first examine the compartmentalized generation of ROS within myofibers, highlighting the distinct contributions of mitochondrial and NADPH oxidase systems and the context-dependent nature of oxidative eustress versus distress. We then detail how redo
