Current pharmacotherapy for obesity.

More than one-third of adults in the USA have obesity, which causes, exacerbates or adversely impacts numerous medical comorbidities, including diabetes mellitus and cardiovascular disease. Despite intensive lifestyle modifications, the disease severity warrants further aggressive intervention, including pharmacotherapy, medical devices and bariatric surgery. Noninvasive anti-obesity drugs have thus now resurfaced as targeted adjunctive therapeutic approaches to intensive lifestyle intervention, bridging the gap between lifestyle and bariatric surgery. In this Review, we discuss FDA-approved a

Evidence for differential top-down and bottom-up suppression in posterior parietal cortex.

When searching for an object, we usually avoid items that are visually different from the target and objects or places that have been searched already. Previous studies have shown that neural activity in the lateral intraparietal area (LIP) can be used to guide this behaviour; responses to task irrelevant stimuli or to stimuli that have been fixated previously in the trial are reduced compared with responses to potential targets. Here, we test the hypothesis that these reduced responses have a different genesis. Two animals were trained on a visual foraging task, in which they had to find a ta

Studies on the anorectic effect of N-acylphosphatidylethanolamine and phosphatidylethanolamine in mice.

N-acyl-phosphatidylethanolamine is a precursor phospholipid for anandamide, oleoylethanolamide, and other N-acylethanolamines, and it may in itself have biological functions in cell membranes. Recently, N-palmitoyl-phosphatidylethanolamine (NAPE) has been reported to function as an anorectic hormone secreted from the gut and acting on the brain (Gillum et al., [5]). In the current study, two of our laboratories independently investigated whether NAPE metabolites may be involved in mediating the anorectic action of NAPE i.p. injected in mice. Thus, the anorectic activity of a non-hydrolysable N

Enhanced effect of neuropeptide Y on food intake caused by blockade of the V(1A) vasopressin receptor.

Food intake is regulated by various factors such as neuropeptide Y. Neuropeptide Y potently induces an increase in food intake, and simultaneously stimulates arginine-vasopressin (AVP) secretion in the brain. Recently, we reported that V(1A) vasopressin receptor-deficient (V(1A)R(-/-)) mice exhibited altered daily food intake accompanied with hyperglycemia and hyperleptinemia. Here, we further study the involvement of the AVP/V(1A) receptor in the appetite regulation of neuropeptide Y with V(1A)R(-/-) mice and antagonists for the AVP receptor. The intra-cerebral-ventricle administration of neu

Appetite control and energy balance regulation in the modern world: reward-driven brain overrides repletion signals.

Powerful biological mechanisms evolved to defend adequate nutrient supply and optimal levels of body weight/adiposity. Low levels of leptin indicating food deprivation and depleted fat stores have been identified as the strongest signals to induce adaptive biological actions such as increased energy intake and reduced energy expenditure. In concert with other signals from the gut and metabolically active tissues, low leptin levels trigger powerful activation of multiple peripheral and brain systems to restore energy balance. It is not just neurons in the arcuate nucleus, but many other brain s

Morphine withdrawal decreases responding reinforced by sucrose self-administration in progressive ratio.

Previous studies have shown that withdrawal from psychostimulant drugs such as d-amphetamine or methamphetamine decreases motivation to work for a natural reinforcement, which is thought to be associated with the withdrawal-induced depressive state and hypofunction of the mesolimbic dopamine system. However, to our knowledge, studies exploring the effect of morphine withdrawal on motivation for a natural reinforcement are lacking. The purpose of the present study was to examine whether motivation to work for a natural reinforcement changes during morphine withdrawal. Three groups of male Sprag

Antifeedants in the feces of the pine weevil Hylobius abietis: identification and biological activity.

Egg-laying females of the pine weevil, Hylobius abietis (L.), regularly deposit feces adjacent to each egg. Egg cavities are gnawed in the bark of roots of recently dead conifer trees. After egg deposition, the cavity is sealed by feces and a plug of bark fragments. Root bark containing egg cavities with feces is avoided as food by pine weevils, which indicates the presence of natural antifeedants. Here we present the first results of the isolation and chemical analyses of antifeedant compounds in the feces of H. abietis. In feeding bioassays, methanol extracts of the feces revealed strong ant

Pattern generation for walking and searching movements of a stick insect leg. II. Control of motoneuronal activity.

In the stick insect, Cuniculina impigra, intracellular recordings from mesothoracic motoneurons that control flexion and extension of the tibia and depression and levation of the trochantero-femur were made while the leg performed walking-like movements on a treadband or stereotyped rhythmic searching movements. We were interested in how synaptic input and intrinsic properties contribute to form the activity pattern of motoneurons during rhythmic leg movements without sensory feedback from other legs. During searching and walking, motoneurons expressed a rhythmic bursting pattern that was form

Ethopharmacological analysis of the effects of putative 'anxiogenic' agents in the mouse elevated plus-maze.

The literature on the effects of anxiety-provoking agents in humans and animals is replete with inconsistent and contradictory findings as well as data that may have alternate explanations. To further our understanding in this area, ethological methods were used to assess in detail the effects of four putative anxiogenic agents in the murine elevated plus-maze test. Compounds studied were FG 7142 (1.25-10.0 mg/kg), pentylenetetrazol (PTZ; 1.875-30.0 mg/kg), isoproterenol (0.125-1.0 mg/kg), and sodium lactate (32.75-262.0 mg/kg). FG 7142 produced an anxiogenic-like profile at 10 mg/kg, an effec

Pharmacological antagonism of lipoprivic feeding induced by sodium mercaptoacetate.

Drugs, such as sodium mercaptoacetate and methylpalmoxirate, which block fatty acid oxidation at different levels in the metabolic pathway, stimulate feeding. It is well known that selective centrally induced stimulation of dopamine, serotonin (5-hydroxytryptamine, 5-HT) and beta-adrenoceptors, or inhibition of the opiatergic system substantially decrease food intake in rats trained to eat 4 h a day. The results of the present study show that centrally acting dopaminergic and serotoninergic anorectic drugs, the opiate receptor antagonist naloxone, the alpha-adrenoceptor blocking drug phentolam

Cyclo(His-Pro) potentiates the reduction of food intake induced by amphetamine, fenfluramine, or serotonin.

Electrophysiological and pharmacological evidence suggests that cyclo(His-Pro) (cHP) could reduce food intake by modulating the actions of relevant neurotransmitters. We tested this hypothesis by giving rats a combination of cHP or its analogs centrally and an anorectic, amphetamine or fenfluramine, systemically. Compared to saline control, cHP at doses too low to affect food intake by itself significantly potentiated the reduction of food intake by amphetamine. This potentiation is thought to be due to cHP modulation of norepinephrine (NE) action, because at the low dose used amphetamine acts

Role of the posterior parietal association cortex in the processing of spatial event information.

Humans and monkeys with damage to the parietal association cortex (PC) exhibit deficits on visual-spatial tasks that are often dependent upon the attentional requirements of the task at hand. In order to test whether there is a correspondence between humans and rats in terms of the function of PC in the processing of spatial information, separate groups of rats were trained in two list-learning tasks, a win-stay task and a win-shift task, on an eight-arm radial maze. It is assumed that these two tasks vary in degree of required attention effort, because the win-shift rule is considerably easie

A comparison of the effects of phenylpropanolamine, d-amphetamine and d-norpseudoephedrine on open-field locomotion and food intake in the rat.

d,l-Norephedrine (PPA) is available as an over-the-counter appetite suppressant and nasal decongestant in the U.S.A. The pseudoisomer d-norpseudoephedrine, is available as an appetite suppressant in Europe, and has been isolated as one of the stimulatory components (cathine) of the Khat plant. Some authors have misidentified cathine as PPA and this confusion in the literature has resulted. PPA and d-norpseudoephedrine possess significantly different pharmacological properties despite having identical structural formulae. Anorectic activity was determined in a food-deprived rat model. PPA and d

(S)-3-[(benzyloxy)methyl]morpholine hydrochloride: a nonstimulant appetite suppressant without conventional neurotransmitter releasing properties.

The synthesis and appetite-suppressant activity of (S)-3-[(benzyloxy)methyl]morpholine hydrochloride in dogs are reported. The oral ED50 for appetite suppression in dogs of 3 was 12 mg/kg, and it was tolerated up to 200 mg/kg. 3 had no inhibitory effect on the release or uptake of noradrenaline, dopamine, or serotonin at 10(-5) M. The (R) enantiomer of 3 was not anorexiant.

A comprehensive psychological approach to obesity.

Many different treatments for obesity have proven somewhat successful, with none emerging as clearly superior to the others. Surgical approaches, although usually successful at achieving and maintaining weight loss, are accompanied by varied and often harmful side effects. Unless new, safer procedures are developed, treatment may continue to be as hazardous as the obesity itself. Because medical treatment alone has not achieved beneficial, lasting results, research aimed at finding new procedures should be encouraged. Our five-year experience in working with obese patients has led us to the fo

CERVICAL ABSCESSES CAUSED BY GEMELLA CUNICULI IN A DOMESTIC RABBIT (ORYCTOLAGUS CUNICULUS)-DIAGNOSIS AND SURGICAL THERAPY.

Gemella spp. is considered an emerging pathogen that causes endocarditis, sepsis, intracranial abscessation, and other infections in humans. A four-year-old female spayed lop-eared rabbit (Oryctolagus cuniculus) was presented for hyporexia and decreased water consumption. Physical examination revealed a lobulated right cervical mass-like lesion. Gemella cuniculi was cultured from the lesion and confirmed via whole-blood PCR and direct sequencing. CT revealed a right cervical mass involving the right salivary gland with a suspected draining tract and consolidation in the right caudal lung lobe.
