Autophagy regulates cellular senescence by mediating the degradation of CDKN1A/p21 and CDKN2A/p16 through SQSTM1/p62-mediated selective autophagy in myxomatous mitral valve degeneration.

Myxomatous mitral valve degeneration (MMVD) is one of the most important age-dependent degenerative heart valve disorders in both humans and dogs. It is characterized by the aberrant remodeling of extracellular matrix (ECM), regulated by senescent myofibroblasts (aVICs) transitioning from quiescent valve interstitial cells (qVICs), primarily under TGFB1/TGF-β1 control. In the present study, we found senescent aVICs exhibited impaired macroautophagy/autophagy as evidenced by compromised autophagy flux and immature autophagosomes. MTOR-dependent autophagy induced by rapamycin and torin-1 attenua…

Dissection of autophagy in tobacco BY-2 cells under sucrose starvation conditions using the vacuolar H(+)-ATPase inhibitor concanamycin A and the autophagy-related protein Atg8.

Tobacco BY-2 cells undergo autophagy in sucrose-free culture medium, which is the process mostly responsible for intracellular protein degradation under these conditions. Autophagy was inhibited by the vacuolar H(+)-ATPase inhibitors concanamycin A and bafilomycin A1, which caused the accumulation of autophagic bodies in the central vacuoles. Such accumulation did not occur in the presence of the autophagy inhibitor 3-methyladenine, and concanamycin in turn inhibited the accumulation of autolysosomes in the presence of the cysteine protease inhibitor E-64c. Electron microscopy revealed not onl…

Autophagy as a Proposed Regulatory Interface in the Gut Microbiota-Cardiovascular Axis.

The gut-cardiovascular axis is increasingly recognized as a contributor to cardiovascular disease (CVD) risk; however, most available evidence remains associative, and the mechanistic links between gut dysbiosis, autophagy, and cardiovascular pathology require further validation. This review proposes a conceptual framework in which autophagy may serve as a regulatory interface linking gut microbiota-derived signals with vascular and cardiac dysfunction. We summarize evidence that microbial metabolites, including trimethylamine N-oxide (TMAO), short-chain fatty acids (SCFAs), bile acids, and dy…

Autophagy dependent HIF1α proteostasis is compromised in models of PEX1 deficiencies.

Peroxisomes, along with mitochondria, coordinate and compartmentalize oxidative metabolism in eukaryotic cells. Rare genetic disorders cause … that PEX1 deficiency alters HIF1α proteostasis by reducing degradation through a hydroxylation- and autophagy-dependent mechanism. Notably, enhancing autophagic capacity by ULK1 agonism was sufficient to reduce HIF1α levels in PEX1 deficient cells. Lastly, we demonstrate that upon hypoxia-reoxygenation, PEX1 deficient cells are slower to reset HIF1α levels. …

Chaperone-mediated autophagy is a tumor-suppressive mechanism in hepatocellular carcinoma.

Chaperone-mediated autophagy (CMA) is a selective lysosomal pathway essential for proteostasis and stress adaptation that declines with aging and metabolic disease, conditions closely linked to hepatocellular carcinoma (HCC). Using genetically engineered mouse models with systemic, hepatocyte-specific, or T cell-specific deletion of the CMA regulator LAMP2A in an MYC-driven, TP53-deficient HCC context, we demonstrate that CMA exerts cell-type-dependent tumor-suppressive functions. Hepatocyte-intrinsic CMA loss promotes early malignant transformation, whereas T cell-specific CMA deficiency impa…

Canonical pathways and selective mechanisms of autophagy in inflammatory bowel disease.

Inflammatory bowel disease (IBD), encompassing Crohn's disease (CD) and ulcerative colitis (UC), is a chronic inflammatory disorder of the gastrointestinal tract. Autophagy, an essential intracellular homeostatic process, plays a pivotal role in the pathogenesis and progression of IBD. This review systematically examines recent advances in understanding the involvement of autophagy in IBD, with a particular focus on the regulatory mechanisms governing its sequential phases-initiation, elongation, and termination-and their respective contributions to intestinal inflammation. We highlight how dy…

Magnetic fields as biophysical activators of autophagy: A preclinical systematic review.

While pharmacological inducers of autophagy have been extensively studied, their systemic adverse effects may limit clinical use. Increasing preclinical evidence suggests that magnetic fields (MFs) can represent a non-invasive alternative for autophagy modulation. In this systematic review, we sought to evaluate preclinical evidence on MF-mediated autophagy activation, including intervention parameters, mechanistic pathways, and therapeutic outcomes, to guide future translational research. Following PRISMA guidelines, we searched (January 2010-August 2025) four structured electronic databases …

Autophagy as a therapeutic linchpin in metabolic diseases and obesity-associated diabetes.

Autophagy, a conserved lysosomal degradation pathway, is increasingly recognized as a central regulator of metabolic health. Its impairment contributes directly to obesity and type 2 diabetes by disrupting nutrient sensing, stress adaptation, and organelle quality control. Hyperactivation of MTORC1 with insufficient AMPK and SIRT1 signaling suppresses autophagic flux, driving lipid accumulation, insulin resistance, and mitochondrial dysfunction. Clinically relevant consequences include adipose inflammation and hypertrophy, hepatic steatosis with impaired β-oxidation, pancreatic β-cell failure …

EPA restores neuroimmune homeostasis in Schizophrenia: evidence for the involvement of autophagy.

Immune dysregulation, particularly inflammation, may contribute significantly to the pathophysiology of schizophrenia (SZ). Autophagy is an essential cellular process that maintains neuroimmune homeostasis, but its relationship with inflammation in SZ remains unclear. Eicosapentaenoic acid (EPA), an omega (n)-3 polyunsaturated fatty acid (PUFA), was reported to exert therapeutic effects by modulating inflammation. Here, we hypothesized that EPA alleviates SZ-related behavioral and neurochemical abnormalities via autophagy regulation. Transcriptomic analysis of peripheral blood from SZ patients…

Autophagy modulation in ulcerative colitis with traditional Chinese medicine: A review.

BACKGROUND: Ulcerative colitis (UC) is a diffuse inflammatory disease that affects the rectum and colon. Management predominantly relies on pharmacotherapy, but challenges related to the side effects, hormone dependence, and drug resistance remain unresolved. Recent studies have identified autophagy as a promising therapeutic target in UC. Accumulating evidence indicates that Traditional Chinese Medicine (TCM) exerts therapeutic effects through the modulation of autophagy-related pathways. However, the underlying mechanisms remain fragmented and have not been systematically integrated. Therefo…

Autophagy as a mechanistic link between physical exercise and Alzheimer's disease.

Alzheimer's Disease (AD), the most prevalent cause of dementia worldwide, is a neurodegenerative disorder that currently has no cure. A growing body of evidence suggests that physical exercise is a potential non-pharmacological strategy in the treatment of AD. Recent findings highlight the involvement of autophagy in the modulatory actions of physical exercise for AD. Here, we present a narrative review of the current knowledge on how exercise impacts AD, specifically focusing on its regulation of autophagic activity in animal models of AD. Evidence from rodent studies further demonstrates tha…

The pleiotropic impact of chaperone-mediated autophagy on skeletal muscle integrity.

Skeletal muscle is a fundamental tissue as it is found throughout the body, sustains posture, and produces movement. Yet, skeletal muscle disorders, such as myopathies, affect a large percentage of the population, degrading an individual's quality of life. A recent study links myopathy progression to the decline in chaperone-mediated autophagy that occurs during aging. Underscoring the importance of a balanced CMA pathway in maintaining skeletal muscle function and integrity, the study also provides mechanistic insights into the pathways that are dysregulated due to defective CMA and presents …

Autophagy and Lysosomal Dysregulation in Endometriosis: Therapeutic Opportunities and Molecular Insights.

Endometriosis is a chronic estrogen-dependent inflammatory condition caused by the presence of endometrium-like tissue outside the uterus. It can cause pelvic pain, infertility, and a reduction in quality of life. Despite being a very common disorder, there is still a lack of understanding of the molecular pathogenesis of endometriosis, which has hampered the development of effective targeted therapies. Recent studies have revealed that dysregulation of autophagy and lysosomal function are critical drivers of disease pathogenesis. Autophagy is a highly conserved, highly regulated process of in…

Autophagy in endothelial cells: A gatekeeper of immune cell trafficking and function.

Endothelial cells (ECs) are increasingly recognised as dynamic regulators of immunity rather than passive vascular barriers. By integrating inflammatory, metabolic, and mechanical cues, ECs actively shape leukocyte trafficking and determine inflammatory outcomes. Autophagy, a conserved lysosomal degradation pathway essential for cellular homeostasis, has emerged as a key determinant of endothelial function in health and disease. Recent work identifies EC autophagy as a context-dependent determinant of leukocyte trafficking, vascular inflammation, antigen presentation, and endothelial phagocyti…

The autophagy-senescence-inflammasome axis: A novel triad in neurodegenerative diseases?

Chronic neuroinflammation is a defining feature of brain ageing and neurodegenerative disorders, yet the molecular mechanisms responsible for its persistence remain incompletely understood. Although autophagy dysfunction, glial senescence, and inflammasome activation are well-established contributors to progressive neurodegeneration, these processes are often analysed independently or through pairwise interactions, leaving their collective contribution to persistent neuroinflammation and disease progression insufficiently defined. Here, we synthesise emerging evidence supporting an integrated …
