To address the pharmacokinetic limitations of current selective estrogen receptor degraders (SERDs) and explore structurally novel ERα-targeted degraders, a series of coumarin derivatives were designed, synthesized, and biologically evaluated. Structure-activity relationship studies indicated that the phenoxy linker, 7- and 3-position bisphenolic hydroxyl groups, and R-configured basic side chain were critical for activity. Among them, compound 23g bearing an (R)-3-methylpyrrolidine moiety showed the most potent antiproliferative activity against MCF-7 cells, with an IC50 value of 0.015 μM. SP

Microplastics (MPs) accumulate in tissues, including kidney tissue, while Bisphenol A (BPA) is a plasticizer of particular concern. At present, the combined effects of MPs and BPA are unexplored in human renal cells. Therefore, we exposed a proximal tubular cell line (PTECs) to polyethylene (PE)-MPs and BPA, both separately and in combination. When co-exposed, cells showed a significantly reduced cell viability (MTT test) and a pronounced pro-oxidant (MDA levels, NRF2 and NOX4 expression by Western blot) and pro-inflammatory response (IL1β, CCL/CCR2 and CCL/CCR5 mRNAs by RT-PCR), compared to t

Calbindin D-28k (CB) is a calcium-binding protein widely distributed in living organisms that may act as a calcium buffer and sensory protein. CB is present in the enteric nervous system (ENS) situated in the gastrointestinal tract, which controls the majority of activities of the stomach and intestine. The influence of various doses of bisphenol A (BPA)-a chemical compound widely used in plastics production-on the number and distribution of CB-positive enteric neuronal cells in the porcine caecum was investigated with an immunofluorescence technique. The obtained results showed that low dosag

Environmental dermatology is the study of how environmental factors affect the integumentary system. The environment includes natural and built habitats, encompassing ambient exposure, occupational exposures, and lifestyle exposures secondary to dietary and personal care choices. This review explores common toxins found in personal care products and packaging, such as bisphenols, parabens, phthalates, per- and poly-fluoroalkyl substances, p-phenylenediamine, and formaldehyde. Exposure to these toxins has been associated with carcinogenic, obesogenic, or proinflammatory effects that can potenti

Bisphenol A (BPA), a well-known endocrine disruptor present in epoxy resins and polycarbonate plastics, negatively disturbs the male reproductive system affecting male fertility. In vivo studies showed that BPA exposure has deleterious effects on spermatogenesis by disturbing the hypothalamic-pituitary-gonadal axis and inducing oxidative stress in testis. This compound seems to disrupt hormone signalling even at low concentrations, modifying the levels of inhibin B, oestradiol, and testosterone. The adverse effects on seminal parameters are mainly supported by studies based on urinary BPA conc

BACKGROUND: Endocrine disrupting chemicals (EDCs) are xenobiotics with the ability to interfere with hormone action, even at low levels. Prior environmental epidemiology studies link numerous suspected EDCs, including phthalates and bisphenol A (BPA), to adverse neurodevelopmental outcomes. However, results for some chemicals were inconsistent and most assessed one chemical at a time.
OBJECTIVES: To evaluate the overall impact of prenatal exposure to an EDC mixture on neurodevelopment in school-aged children, and identify chemicals of concern while accounting for co-exposures.
METHODS: Among

BACKGROUND: Endocrine disrupting compounds (EDCs) such as phthalates and phenols can affect placental functioning and fetal health, potentially via epigenetic modifications. We investigated the associations between pregnancy exposure to synthetic phenols and phthalates estimated from repeated urine sampling and genome wide placental DNA methylation.
METHODS: The study is based on 387 women with placental DNA methylation assessed with Infinium MethylationEPIC arrays and with 7 phenols, 13 phthalates, and two non-phthalate plasticizer metabolites measured in pools of urine samples collected twi

Environmental pollution has emerged as a critical global health threat, with xenobiotic exposure increasingly linked to the pathogenesis of chronic liver diseases. The gut-liver axis has been proposed as a potential mediator in the body's response to these insults. This review evaluates how selected environmental pollutants-heavy metals, particulate matter (PM), and endocrine-disrupting chemicals (EDCs), particularly Bisphenol A (BPA)-disrupt this homeostatic balance. Evidence indicates that environmental toxins induce gut dysbiosis and impair intestinal barrier integrity. This "leaky gut" phe

Emerging contaminants of concern (CECs) are increasingly detected in tropical riverine systems, yet their prioritization remains challenging due to data gaps, complex mixtures, and limited integration of ecological and human health perspectives. This study proposes an integrated framework to identify priority CECs by combining occurrence data, ecological risk quotients (RQ), human health hazard quotients (HQ), and a novel prioritization index (PI). Thirty-four (34) multiclass CECs, including pharmaceuticals, hormones, and plasticizers, were quantified in four rivers in Johor, Malaysia using op

Bisphenol S (BPS), a widely utilized environmental endocrine disruptor, poses significant risks to environmental pollution and human health. While the reproductive toxicity of BPS is recognized, its transgenerational consequences, particularly from early-life exposure, remain poorly defined. In this study, we established a mouse model of early-life BPS exposure to assess its effects on male reproductive development in offspring. Our findings demonstrate that early-life BPS (0.2, 2, 20 mg/L) exposure impairs testicular architecture, diminishes sperm quantity, elevates sperm morphological abnorm

The aim of the present study was to investigate the role of endoplasmic reticulum (ER) stress in bisphenol A (BPA) - induced hepatic lipid accumulation as well as the protective effects of Sulforaphane (SFN) in this process. Human hepatocyte cell line (LO2) and C57/BL6J mice were used to examine BPA-triggered hepatic lipid accumulation and the underlying mechanism. Hepatic lipid accumulation, triglycerides (TGs) levels, the expression levels of lipogenesis-related genes and proteins in the ER stress pathway were measured. It was revealed that BPA treatment increased the number of lipid droplet

Selenium deficiency can lead to multiple tissue and organ damage in the body and could coexist with chronic toxic exposures. Contamination from Bisphenol A (BPA) exposure can induce the occurrence of various injuries including pyroptosis. However, it is not clear whether selenium deficiency and BPA exposure affect tracheal tissue pyroptosis in chickens. To investigate whether selenium deficiency and BPA exposure induce chicken tracheal tissue pyroptosis via the NF-κB/NLRP3/Caspase-1 pathway and the effect of their combined exposure on tissue injury, we developed a model of relevant chicken tra

Endometriosis is a hormone-dependent inflammatory gynecological disease of reproductive-age women. It is clinically and pathologically characterized by the presence of functional endometrium as heterogeneous lesions outside the uterine cavity. The two major symptoms are chronic pelvic pain and infertility, which profoundly affect women's reproductive health and quality of life. This significant individual and public health concerns underscore the importance of understanding the pathogenesis of endometriosis. The environmental endocrine-disrupting chemicals (EDCs) are exogenous agents that inte

Prenatal exposure to endocrine-disrupting chemicals has been linked to gestational hypertension (GH) and preeclampsia (PE). However, the results were conflicting and inconclusive. We conducted a systematic review and meta-analysis for an overview of these relationships. We searched PubMed, and Google Scholar for studies investigating bisphenol A, phthalates, and per or poly-fluoroalkyl substances and GH or PE. Pooled odds ratio (OR) with a 95% confidence interval (CI) were calculated for risk estimate using the generic inverse variance method. A total of 14 studies were included in the present

Ferrihydrite (Fh) and transition metal ions such as Cu(II) are ubiquitous in natural and engineered aquatic systems, influencing pollutant degradation through Fenton-like processes. In contrast to previous studies focusing exclusively on either Cu(II)-catalyzed homogeneous reactions or iron oxide-mediated heterogeneous reactions, this work presents an integrated view of Cu(II)-Fh interactions under environmentally relevant pH conditions. We demonstrate that Cu(II) and Fh jointly establish a dual-oxidant system capable of generating both hydroxyl radicals (•OH) and Cu(III). These reactive speci

To establish a novel in vivo test system for rapid detection of environmental estrogens, an ere-zvtg1: gfp transgenic zebrafish line has been generated. In this transgenic line, under control conditions, GFP was exclusively expressed in the liver of mature adult female fish. Male and larval transgenic fish did not express GFP but could be induced to express GFP in the liver after exposure to 17-alpha-ethynylestradiol (EE(2)). Concurrent accumulation of zvtg1 and gfp mRNAs in embryos and larvae after EE(2) exposure was observed, which indicated that the expression of gfp transgene was driven by

Prenatal exposure to bisphenol analogs is becoming increasingly common and may affect the development of myopia in children. However, human evidence and mechanistic studies remain limited. In this study, 370 mother-child pairs from the Shanghai-Minhang Birth Cohort Study (S-MBCS) were analyzed. We measured the concentrations of five bisphenol analogs in maternal urine, quantified genome-wide DNA methylation (DNAm) in placental samples, and assessed the myopia status and spherical equivalent (SE) of school-aged children. Multiple CpG sites with methylation levels significantly associated with S

BACKGROUND: Methylmalonic acid (MMA) has been associated with genetic factors and vitamin B12 status. However, the impact of environmental pollutant exposure on MMA levels and whether MMA statistically accounts for a portion of the pollutant-cardiovascular disease (CVD) risk remain unexplored.
AIM: To assess the association between bisphenol A (BPA) and MMA and quantify the extent to which MMA accounts for the BPA-CVD association among adults aged 60 and younger.
METHODS: Cross-sectional data from 2275 adults aged ≤ 60 years were obtained from the US National Health and Nutrition Examination
