The effects of low-intensity ultrasound on peripheral nerve regeneration in poly(DL-lactic acid-co-glycolic acid) conduits seeded with Schwann cells.

This study attempted to improve the efficacy of peripheral nerve regeneration, using the stimulus of low-intensity ultrasound (US) on poly(DL-lactic acid-co-glycolic acid) (PLGA) nerve guidance conduits seeded with Schwann cells. The possible differences between the ultrasonic effects of biodegradable and nonbiodegradable materials used as conduits were also investigated, by comparison with a group of silicone conduits. The PLGA conduits were seeded with or without Schwann cells (6 x 10(3) cells). All conduits were implanted 10 mm into right sciatic nerve defects in rats and underwent 12 ultra

AIDS-related Kaposi's sarcoma cells rapidly internalize endostatin, which co-localizes to tropomysin microfilaments and inhibits cytokine-mediated migration and invasion.

AIDS-related Kaposi's sarcoma (KS) is the most common HIV-related malignancy. In some respects, KS is analogous to other angioproliferative diseases, in that KS lesions are highly vascularized and promoted by inflammatory cytokines. However, unlike other cancers or inflammatory mediated vascular diseases, KS is unique in that the KS lesional cells both express and respond to the complete angiogenic cytokines vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF). Therefore, the angiogenic phenotype, which is crucial for cancer progression, is inherent to KS tumor c

O(2) modulates large-conductance Ca(2+)-dependent K(+) channels of rat chemoreceptor cells by a membrane-restricted and CO-sensitive mechanism.

Hypoxic inhibition of large-conductance Ca(2+)-dependent K(+) channels (maxiK) of rat carotid body type I cells is a well-established fact. However, the molecular mechanisms of such inhibition and the role of these channels in the process of hypoxic transduction remain unclear. We have examined the mechanisms of interaction of O(2) with maxiK channels exploring the effect of hypoxia on maxiK currents recorded with the whole-cell and the inside-out configuration of the patch-clamp technique. Hypoxia inhibits channel activity both in whole-cell and in excised membrane patches. This effect is str

The expression of co-stimulatory and accessory molecules on cultured human muscle cells is not dependent on stimulus by pro-inflammatory cytokines: relevance for the pathogenesis of inflammatory myopathy.

Specific activation of naive T cells requires TCR engagement plus interaction of CD28 on T cells with co-stimulatory B7-1/B7-2 on APCs. Since muscle cells may be directly involved in activating muscle-infiltrating T lymphocytes in polymyositis and inclusion body myositis, we analyzed B7 expression on myoblasts before and after treatment with pro-inflammatory cytokines. We found no expression of B7-1/B7-2, either constitutively or after stimulus with cytokines. Furthermore, myoblasts failed to stimulate alloreactive peripheral blood lymphocytes in mixed lymphocyte reactions. Lack of B7 expressi

Th0-like CD4+ T cells protect mice with murine retrovirus-induced immunodeficiency syndrome (MAIDS) against co-infection with Listeria monocytogenes.

We examined the host defence mechanism against infection with Listeria monocytogenes, a facultative intracellular bacterium, in mice with murine acquired immunodeficiency syndrome (MAIDS) caused by LP-BM5 murine leukaemia virus (MuLv) infection. Although LP-BM5 MuLV infection in C57BL/6 mice leads to a stage of immunodeficiency characterized by severe compromise of cell-mediated immunity, the mice with established MAIDS infected with LP-BM5 8 weeks previously, showed resistance to an intraperitoneal infection with Listeria monocytogenes. These MAIDS mice also showed resistance to a lethal dose

Selective Absence of Co-Inhibitory Receptor-Expressing CD4+ T Cells in Lungs of Obese House Dust Mite-Allergic C57BL/6 Mice.

BACKGROUND: Asthma and obesity are highly prevalent chronic inflammatory diseases, the mutual pathomechanistic relationships of which have not been fully elucidated yet. We hypothesized that inflammatory mechanisms in obese adipose tissue influence the phenotype of an induced allergic airway inflammation (AAI) with altered activation processes in CD4+ T cells being involved in this interaction. METHODS: C57BL/6 and BALB/c mice were fed high-fat (HFD) or normal (ND) diet for several weeks and subjected to a house dust mite (HDM)-induced model of AAI mimicking main features of human asthma. Bimetallic Cu-NC/Co-NC nanocomposite from metal-organic framework precursors as a cathodic electrocatalyst for enhanced microbial fuel cell performance.

Developing an affordable catalyst for microbial fuel cells (MFCs) is essential for advancing alternative energy sources and promoting environmentally sustainable power generation. Herein, an active bimetallic Cu-NC/Co-NC nanocomposite was developed as a novel cathodic electrocatalyst using copper-1,3,5-benzene tricarboxylic acid and zeolitic imidazole framework-67 through controlled pyrolysis under nitrogen atmosphere. The synergistic interaction between Cu and Co active centers is specifically designed to enhance oxygen reduction reaction in a two-chamber MFC. Structural and morphological cha

Effects of yttrium doping on oxygen conductivity in Ba(Fe, Co, Zr, Y)O3-δ cathode materials for proton ceramic fuel cells.

Proton ceramic fuel cells (PCFCs) operate efficiently at lower operating temperatures than solid oxide fuel cells, but their performance is often limited by cathodic oxygen reduction reaction (ORR) kinetics. BaCoxFeyZrzY1-x-y-zO3-δ (BCFZY) perovskites are promising triple-conducting cathodes for PCFCs, although the role of acceptor dopants such as Y on oxygen conductivity remains unclear. In this study, we investigate the effects of Y doping on oxygen conductivity in BaFe0.4Co0.4Zr0.2O3-δ (BCFZ), BaFe0.4Co0.4Zr0.1Y0.1O3-δ (BCFZY0.1) and BaFe0.4Co0.4Y0.2O3-δ (BCFY). Multi-level evidence for Pb-Cd co-exposure nephrotoxicity: An integrated analysis of NHANES, outcome-oriented network toxicology, and cell-based validation.

BACKGROUND: Lead (Pb) and cadmium (Cd) remain widespread environmental toxicants, and the kidney is a major target organ. However, reliance on a single biomarker matrix or renal endpoint may incompletely characterize Pb-Cd-associated renal abnormalities. Whether blood- and urine-based Pb/Cd biomarkers exhibit differential association profiles across renal outcomes, and whether outcome-oriented networks converge on shared candidate pathways, remains unclear. METHODS: We analyzed NHANES 2011-2018 data from 5049 adults. Blood and urinary Pb and Cd biomarkers were evaluated in relation to estimat

Tissue-specific tolerance mechanisms and lymph node co-drainage shape T cell immunity in the upper digestive system and pancreatic cancer progression.

The liver, pancreas, and duodenum share lymph nodes (LNs), providing a unique system to examine how tissue origin of self-antigens shapes T cell fate. Comparing mice expressing ovalbumin (OVA) from distinct subcellular compartments, we found that cytosolic OVA from the liver or pancreas, but not gut, was immunologically ignored. High-dose hepatic-secreted OVA triggered antigen-specific T cell deletion, whereas secreted pancreatic and intestinal OVA induced regulatory T (Treg) cells, revealing immunological ignorance, clonal deletion, and Treg cell generation as tissue-specific tolerance mechan

Co-transplantation of pancreatic islets with adipose-derived mesenchymal stem cells in vascularized kidney ECM scaffolds to restore type 1 diabetes mellitus.

Type 1 diabetes mellitus (T1DM) requires innovative strategies to restore insulin production while overcoming immune rejection and limited donor availability. In this study, we developed a bioengineered insulin-producing tissue (IPT) by combining isolated pancreatic islets with adipose-derived stem cells (ADSCs) on a decellularized kidney extracellular matrix (k-ECM). Instead of discarding the substandard conditions islets, we demonstrate that they provide valuable bioactive signals like cytokines, growth factors, and extracellular matrix cues, which promote ADSCs differentiation into function

Shiga Toxin Induces Apoptosis via ROS-Caspase Activation in Human Cerebral Endothelial Cell Line hCMEC/D3 and Astrocyte Co-Culture.

Hemolytic uremic syndrome (HUS), a fatal complication of Shiga toxin-producing Escherichia coli (STEC) infection, is classically characterized by acute renal failure, but frequently accompanied by central nervous system (CNS) dysfunction. Because the CNS is normally protected by the blood-brain barrier (BBB), the toxin-mediated BBB injury is considered to be a major cause of neurologic sequelae in STEC infection. Here, we delineate how Shiga toxin type 1a (Stx1a) and Shiga toxin type 2a (Stx2a) compromise BBB-like endothelial barrier integrity with the human brain microvascular endothelial cel

Use of the J774A.1 Cell Line as a Model in the In Vitro Study of Extracellular Vesicle Secretion from Histiocytic Sarcoma in Patients with Bacterial Co-Infections.

Histiocytic sarcoma (HS) is an aggressive hematological malignancy whose transformed cells exhibit morphological and immunophenotypic characteristics similar to macrophages, and arises de novo or as part of a clonal 'evolution' of other pre-existing hematological neoplasms. This study investigates the potential use of the J774A.1 cell line (a cell line derived from murine tumor cells, commonly used in macrophage research) as a research model to study the role of polydisperse extracellular vesicles (PEVs) secreted by the HS cells, considering that bacterial infections are common in patients wit

Iron oxide nanoparticles co-conjugated with hyaluronic acid and limonene trigger oxidative stress and cell cycle arrest in triple-negative breast cancer.

The hyaluronic acid receptor is typically overexpressed in cancer cells, and hyaluronic acid targeting has gained attention in cancer treatment. In this work, cytotoxic potential of magnetic iron oxide nanoparticles co-conjugated with hyaluronic acid and limonene in the MDA-MB-231 cell line was characterized. Physicochemical characteristics of the nanoparticles were studied by FT-IR, XRD, TEM, SEM-EDS, DLS, zeta potential and VSM analyses. Cytotoxic effects of Fe3O4@Glu-HA-LIM NPs in cancer and normal cells were studied by MTT assay. Flow cytometry and AO/PI staining were employed to study cel

Chlorpyrifos and tamoxifen co-pretreatment promotes stem-like phenotype and upregulation of anti-estrogen therapy resistance markers in ERα breast cancer cells.

Endocrine therapy (ET) is the standard first-line treatment for hormone receptor positive breast cancer (BC) but about 30 % of patients develop resistance. Chlorpyrifos (CPF), is an endocrine disruptor pesticide that promotes a stemness phenotype and triggers molecular mechanisms involved in ET resistance. The goal of this study is to assess whether CPF induces tamoxifen (TAM) resistance by modulating estrogen receptor α (ERα) expression through histone deacetylase 1 (HDAC1) regulation. We analyzed public databases of BC patients and performed in vitro experiments using the MCF-7 cell line. We

Chrysin-loaded poly (lactic-co-glycolic acid) nanoparticles alleviate sepsis-induced splenic injury by regulating myeloid-derived suppressor cells.

Sepsis is a serious condition resulting from a dysregulated immune-mediated response to an infection. Myeloid-derived suppressor cells (MDSCs) aggregate and serve a protective function in the pathophysiology of lipopolysaccharide (LPS) shock. However, the regulation of MDSCs by chrysin-loaded poly (lactic-co-glycolic acid) (PLGA) nanoparticles (CHR-NP) remains unexplored. CHR-NP was synthesized and characterized by Fourier transform infrared (FTIR) spectroscopy, transmission electron microscopy (TEM), zeta potential, and dynamic light scattering (DLS). High-index facet-rich quaternary PtCuFeCo octopods as anti-CO poisoning bifunctional electrocatalysts for direct methanol/ethylene glycol fuel cells.

High-index facets and doping strategies can generate unanticipated effects for Pt-based nanomaterials, but there is still a tremendous challenge to integrate the two advantages to construct advanced bifunctional electrocatalysts for direct proton/anion exchange membrane alcohol fuel cells. Herein, we successfully synthesized quaternary Pt41.8Cu51.6Fe5.0Co1.6 octopod nanocrystals (ODNs) with high-index facets through a double active auxiliary doping strategy. Electrochemical activity analysis reveals that Pt41.8Cu51.6Fe5.0Co1.6 ODNs/C could serve as an alluring bifunctional electrocatalyst for

The Combination of Recombinant Methioninase and Low-dose Chloroquine Selectively Eradicates Colon-Cancer Cells Without Apparent Toxicity on Co-cultured Normal Fibroblasts.

BACKGROUND/AIM: Methionine addiction is a metabolic hallmark of cancer. Recombinant methioninase (rMETase) targets methionine addiction and effectively depletes methionine. rMETase has shown synergy with chemotherapeutic agents on numerous types of cancer cells. Chloroquine (CQ), an anti-autophagy agent, has demonstrated anti-cancer efficacy in pre-clinical studies. The present study aimed to evaluate the cancer selectivity and synergistic efficacy of rMETase and CQ in a co-culture model of colon-cancer cells and normal fibroblasts.
