Modulation of circadian rhythms in articular cartilage by heat pulse.

OBJECTIVE: Disruption of the circadian clock, such as during ageing, is linked to cartilage degeneration in mice and increased osteoarthritis (OA) risk in humans. This study investigated whether heat pulses can resynchronise dampened circadian rhythms in articular cartilage. METHODS: Femoral head cartilage explants were isolated from PER2::LUC circadian reporter mice. Tissues were exposed to heat shock at various temperatures and durations. Circadian bioluminescence from explants was recorded in real-time over several days. Pharmacological compounds were used to investigate the pathways invol…

Surface-based treatment options for cartilage lesions of the knee.

Articular cartilage lesions of the knee pose a significant clinical challenge due to the limited regenerative capacity of hyaline cartilage and the wide variability in patient presentation and lesion characteristics. Surface-based cartilage restoration techniques have evolved substantially, shifting from isolated marrow stimulation toward biologically enhanced strategies designed to improve repair tissue quality, durability, and clinical outcomes while preserving native joint anatomy. These techniques are primarily indicated for contained chondral defects without substantial subchondral bone c…

The Prospects of Electromagnetic Stimulation in Cartilage and Bone Tissue Engineering.

The achievements of regenerative medicine are based on methods of controlling stem cell division and differentiation. Electromagnetic fields stimulate cell differentiation by means of affecting calcium channels and cellular signaling. However, only a small part of the mechanisms underlying electromagnetic field effect on cells has been studied. The prospect of their use in tissue engineering as an addition or alternative to biochemical effects becomes clear in the course of numerous experiments. Electromagnetic stimulation enhances the effect of biochemical differentiation inducers and can cau

Mechanisms of resveratrol in cartilage injury repair and protection: A review.

Cartilage injury is a central pathological feature of degenerative joint diseases such as osteoarthritis. Due to its avascular, aneural, and low-cell-density nature, cartilage has a very limited capacity for self-repair. Consequently, identifying agents capable of delaying cartilage degeneration and promoting repair remains a major research focus. Resveratrol, a natural polyphenol, has garnered significant attention for its potent antioxidant, anti-inflammatory, and cytoprotective properties. Based on recent research advances, this narrative review summarizes the multifaceted protective roles …

The role of Pth1r in posterior cranium cartilage regulation and craniosynostosis.

Craniosynostosis is a major craniofacial congenital disorder that causes developmental complications. During normal cranial development, intramembranous ossification forms the flat bones and sutures, while cartilage appears transiently in the posterior calvarial region. However, in craniosynostosis, premature suture fusion disturbs normal calvarial morphogenesis. To clarify the role of transient cartilage in this morphogenetic disruption, we investigated its molecular regulation and pathology in mice, focusing on parathyroid hormone 1 receptor (Pth1r) signaling. Conditional deletion of Pth1r i…

Artificial Chlorella Biohybrids for Alleviation of Cartilage Degeneration in Osteoarthritis.

Osteoarthritis (OA), one of the most common chronic joint diseases, is characterised by cartilage imbalance and disruption of the cartilage extracellular matrix. In this study, we observed impaired mitophagy and glucose metabolism disorders in OA chondrocytes, which exacerbated cartilage degeneration. And we unveil Chlorella, a natural microorganism that appeared more than two billion years ago, as an efficient regulator of both mitophagy and glucose metabolism in OA chondrocytes. Chlorella activates mitophagy and rescues mitochondrial function. More importantly, Chlorella inhibits glycolysis …

Effect of Malnutrition on Femoral Cartilage Thickness in Pediatric Patients.

Background/Objectives: Malnutrition is an imbalance of nutrients required for growth, development, and organ function. Its impact on bone development is known, but its effects on cartilage remain unclear. This study aimed to evaluate the femoral cartilage thickness in children with primary malnutrition. Methods: In this cross-sectional observational study, 83 children with primary malnutrition and 62 age- and sex-matched healthy controls were included. Patients with primary malnutrition were classified as mild, moderate and severe. Femoral cartilage thickness measurements of all children were …

Assessment of CA4+ Impact on Mechanical Properties of Articular Cartilage.

X-ray imaging of articular cartilage could be a breakthrough for the early diagnosis of tissue degeneration. This approach relies on radiopaque contrast agents to enhance the visualization of soft tissues. The potential impact of contrast agents on the mechanical response of articular cartilage should be considered in the frame of both clinical and research applications. Attention has been drawn to a solution containing molecules with six iodine atoms and four positive charges (CA4+), which has been shown to improve the X-ray visibility of articular cartilage. This study aimed to determine the…

Cartilage Cap in Scaphoid Nonunions: An Indication of Stability and Biology?

PURPOSE: Scaphoid nonunion management is controversial and is dependent upon many factors without a clear algorithm. Traditionally, open approaches involved corticocancellous, vascularized, or nonvascularized structural bone grafting with internal fixation. Recent arthroscopic techniques have improved the ability to assess the stability of fractures and the need for bone grafting while preserving blood supply. We present our experience with arthroscopic-assisted scaphoid nonunion repairs that were discovered to have an intact cartilage cap at the nonunion site and thus were treated with screw …

The Protective Activity of Apigenin Against Bone and Cartilage Diseases.

Bone and cartilage diseases have become the leading causes of joint disability due to the destruction of bone and cartilage. No effective drugs are available to cure bone and cartilage diseases. Exploring natural compounds as therapeutic alternatives shows promise. Apigenin, a naturally occurring flavonoid, exhibits various pharmacological activities, such as anti-inflammation, antioxidant, and immune modulation. Apigenin acts as a phytoestrogen and mediates the balance of bone remodeling by inducing osteogenic differentiation, promoting osteoblast-associated bone formation, and inhibiting ost…

Cartilage degeneration in hemophilic arthropathy: to focus on prevention or regeneration?

INTRODUCTION: One of the problems that remain unsolved in people with hemophilia (PwH) is how to regenerate articular cartilage. AREAS COVERED: In this article, we have reviewed the existing information on articular cartilage regeneration in the general population and in PwH. In the general population, it has been reported that the clinical results of different types of commercial products are better than those of microfractures and that the results of products that combined microfractures and allograft products are better than those of autologous chondrocyte products. In PwH iron chelators, …

Osteochondral solutions for cartilage lesions of the knee: a narrative review.

BACKGROUND AND OBJECTIVE: Cartilage lesions of the knee occur frequently and can cause debilitating symptoms such as pain, swelling, locking, clicking, or catching. In some lesions, both the cartilage and underlying subchondral bone are affected, which may require treatment of both the osseous and cartilaginous units. The objective of this review was to assess the available evidence of advanced treatments for osteochondral lesions in the knee. METHODS: A literature search was conducted in the electronic databases of PubMed, Embase and Cochrane Library for a narrative review on studies reporti…
