Correlation between caspase-3 activation and three different markers of DNA damage in neonatal cerebral hypoxia-ischemia.

Caspase-3 has been identified as a key protease that, by targeting a limited number of proteins, can disrupt essential homeostatic processes and initiate an orderly disassembly of cells, including degradation of genomic DNA. We demonstrate the usefulness of an antibody specific for activated caspase-3 in a model of neonatal rat hypoxia-ischemia (Hl) and correlate the spatial and temporal activation of caspase-3 with three different markers of DNA damage and with the loss of a neuronal marker [microtubule-associated protein 2 (MAP 2)]. An oligonucleotide hairpin probe (HPP) with one base overha

Ectromelia virus virulence factor p28 acts upstream of caspase-3 in response to UV light-induced apoptosis.

Ectromelia virus (EV) virulence factor p28 (EVp28) is a member of a family of poxvirus proteins that are defined largely by the presence of a C-terminal RING finger motif and localization to virus factories within the cytoplasm of infected cells. Previously, overexpression of the Shope fibroma virus (SFV) homologue, N1R, in vaccinia virus (VV)-infected BGMK cells was found to inhibit virus-induced apoptosis. Here, we report that both EVp28 and overexpression of SFV N1R in poxvirus-infected HeLa cells protect specifically from UV light-induced apoptosis, but not from apoptosis induced by Fas or

Proteolytic processing of the Alzheimer's disease amyloid precursor protein within its cytoplasmic domain by caspase-like proteases.

Alzheimer's disease is characterized by neurodegeneration and deposition of betaA4, a peptide that is proteolytically released from the amyloid precursor protein (APP). Missense mutations in the genes coding for APP and for the polytopic membrane proteins presenilin (PS) 1 and PS2 have been linked to familial forms of early-onset Alzheimer's disease. Overexpression of presenilins, especially that of PS2, induces increased susceptibility for apoptosis that is even more pronounced in cells expressing presenilin mutants. Additionally, presenilins themselves are targets for activated caspases in a…

Modulation of caspase-3 activity and Fas ligand mRNA expression in rat liver cells in vivo by alcohol and lipopolysaccharide.

The purpose of this study was to determine if exacerbation of apoptosis precedes liver injury during chronic exposure of rats to alcohol. After 7 weeks of feeding an alcohol- or dextrin-containing liquid diet, the animals were treated with gram-negative bacterial lipopolysaccharide (1 mg x kg(-1) body weight, intravenously) or sterile saline and sacrificed 3 hr after the treatment. Alanine:2-oxoglutarate aminotransferase (ALT) and lactate:NAD oxidoreductase [lactate dehydrogenase (LDH)] were measured in plasma. The caudate lobe of the liver was resected for histology, while the rest of the org

Apoptosis Repressor with Caspase Recruitment Domain in Uncommon Epidermal Growth Factor Receptor-Mutant Lung Adenocarcinoma: An Aging Hypothesis.

Uncommon epidermal growth factor receptor (EGFR)-mutant lung adenocarcinoma comprises clinically and biologically heterogeneous subgroups with variable sensitivity to targeted therapy. Incomplete responses and eventual progression suggest that factors beyond receptor inhibition influence disease persistence during therapy. This hypothesis-generating narrative review examines whether apoptosis repressor with caspase recruitment domain (ARC), encoded by nucleolar protein 3 (NOL3), warrants investigation as a downstream node in the regulation of apoptosis within an aging-informed framework. The a

FANCD2 knockdown suppresses malignant phenotypes and is associated with caspase/PARP-mediated apoptosis in NCI-H2052 pleural mesothelioma cells.

BACKGROUND: Malignant pleural mesothelioma (PM) is a highly aggressive thoracic malignancy with an extremely poor prognosis and limited effective therapeutic strategies, representing a critical unmet clinical need. Fanconi anemia complementation group D2 (FANCD2) is a critical effector of the cellular DNA damage response system and plays a central role in maintaining genomic integrity. While the genoprotective functions of FANCD2 have been extensively characterized, its specific biological roles and molecular regulatory mechanisms in the malignant progression of PM remain poorly understood. Cardiotoxin from Naja atra Activates the NLRP3/Caspase-1/GSDMD Pyroptosis Pathway to Induce Skin Tissue Injury.

BACKGROUND: Cardiotoxin (CTX) from Naja atra venom is a principal virulence factor responsible for progressive local tissue necrosis and systemic inflammation following snakebite. Despite its clinical importance, the molecular mechanisms underlying CTX-induced skin injury remain poorly defined. METHODS: We employed a two-arm strategy combining transcriptome-guided discovery with mechanistic functional validation. In the transcriptomic arm, C57BL/6 mice received intradermal CTX injection (120 μg/50 μL), and skin tissues were harvested at 6, 12, and 24 h post-injection for RNA-seq analysis. Dif

Targeting the NLRP3/Caspase-1 pyroptotic pathway exacerbates insulin resistance upon exposure to nanoplastics with different surface chemistries.

The surface chemical properties of nanoplastics (NPs), such as surface charge and functional groups, can markedly influence their toxicological effects. Recent studies have shown that NPs can disrupt glucose homeostasis. However, under metabolically susceptible conditions, such as hyperglycemia and metabolic dysfunction, differences in metabolic toxicity and the underlying mechanisms among surface-modified NPs remain unclear. In this study, zebrafish larvae and Hep G2 cells under high-glucose conditions were used to systematically compare the effects of PS-NPs, NPs-NH₂, and NPs-COOH on insulin

Functional analysis of Caspase-1 in Cotesia chilonis via pupal microinjection and larval nanomaterial immersion-mediated RNA interference.

Cotesia chilonis, a dominant endoparasitoid of the striped stem borer, Chilo suppressalis, plays a crucial role in regulating the population of C. suppressalis in the field. RNA interference (RNAi) in C. chilonis is challenging due to the unique biological traits of endoparasitoids. In this study, dsRNA was delivered to C. chilonis by microinjection of pupae within cocoons (delivery outside the host) and nanomaterial-mediated immersion, which delivered dsRNA to larvae inside the host. Both systems were effective in silencing the target gene, Cccaspase-1, with microinjection achieving 41.34% RN

Dual-Enzyme Responsive Polymeric Micelles with Cascade Targeting and Caspase-3 Activated Drug Release for Tumor-Specific Chemotherapy.

Cancer, as a global health crisis, continues to threaten human life and health. Chemotherapy occupies a central position in cancer treatment, but conventional chemotherapeutic agents have some limitations, such as low solubility, lack of specific targeting, insufficient bioavailability and high toxicity to normal tissues. However, the use of DOX is associated with serious adverse effects, including cardiotoxicity and myelosuppression, which limit its effectiveness in antitumor therapy. To overcome these challenges, drug delivery systems (DDSs) have been developed, with polymeric micelles emerg

Oncolytic virus OHSV2 induces pyroptosis in bladder cancer cells via the TLR4/NLRP3/Caspase-1/GSDMD pathway.

Bladder cancer remains a significant clinical challenge due to high recurrence and progression rates, necessitating novel therapeutic strategies. Oncolytic viruses, such as the herpes simplex virus type 2-based OHSV2, have demonstrated promising antitumor effects through direct oncolysis and immune activation. This study investigated the efficacy, safety, and molecular mechanisms of OHSV2 in the treatment of bladder cancer. In vitro and in vivo experiments demonstrated that OHSV2 potently inhibited bladder cancer cell proliferation, migration, and clonogenicity in a dose-dependent manner, whil

N-stearoyltyrosine attenuates ischemia-reperfusion injury by inhibiting caspase-dependent apoptosis and PARP-1/AIF-mediated parthanatos.

Cerebral ischemia‑reperfusion (I/R) injury induces neuronal death via caspase-dependent apoptosis and parthanatos, a poly (ADP-ribose) polymerase 1 (PARP-1)/apoptosis-inducing factor (AIF)-mediated caspase-independent pathway. Simultaneous inhibition of these pathways may provide enhanced neuroprotection. N-Stearoyltyrosine (NsTyr), a synthetic analogue of the endocannabinoid anandamide (AEA), exhibits neuroprotective effects in various models, however, its efficacy and mechanism in middle cerebral artery occlusion (MCAO) remain unclear. Neuronal oxygen-glucose deprivation/ reoxygenation (OGD/

Pseudomonas aeruginosa exoenzyme Y activity inhibits exoenzyme S-induced caspase activation in pulmonary microvascular endothelial cells.

Pseudomonas aeruginosa is the most common cause of ventilator-associated pneumonia in intensive care unit (ICU) patients. Virulent P. aeruginosa strains utilize a type III secretion system that injects exoenzymes S, T, U, and/or Y (ExoS, T, U, and/or Y, respectively) into the host cell cytosol. Most clinically isolated P. aeruginosa strains encode ExoY, often in combination with ExoU and ExoT, or ExoS and ExoT. The effects of each individual exoenzyme on host cell signaling have been extensively studied, but how these signaling mechanisms are altered during co-intoxication is poorly understood

Therapeutic Exosomes Carrying VEGFA siRNA Inhibit Pathological Corneal Angiogenesis via PI3K-Akt-Caspase-3 Signaling.

Background/Objectives: Neovascularization, defined as the sprouting of new blood vessels from pre-existing vasculature, is a critical pathological feature in ocular diseases such as pathological myopia and represents a leading cause of corneal vision loss. Vascular endothelial growth factor A (VEGFA) plays a pivotal role in endothelial cell proliferation, migration, survival by anti-apoptotic signaling, and vascular permeability. Dysregulation of VEGFA is closely linked to pathological neovascularization. Exosomes, nanosized phospholipid bilayer vesicles ranging from 30 to 150 nm, have emerged

Inhibition of the Caspase-9/GSDME axis by Auranofin: a potential therapeutic strategy for bacterial toxin-mediated severe systemic disease.

Bacterial toxin-mediated severe systemic diseases, such as Shiga toxin-induced hemolytic uremic syndrome (HUS), are associated with an exceptionally high mortality rate due to life-threatening multi-organ failure and a profound inflammatory surge. Despite this severe clinical burden, targeted therapeutic drugs remain unavailable. Here, we investigated the therapeutic potential of Auranofin (AUR), an FDA-approved compound, in mitigating systemic lethality by targeting the Caspase-9/GSDME-mediated cell death axis. Utilizing a high-throughput screening of 2819 FDA-approved drugs, we identified AU

CpG-ODN from Streptococcus thermophilus as a tool for inducing extrinsic apoptosis via caspase-8 activation in mouse splenocytes.

Synthetic oligodeoxynucleotides (ODNs) containing unmethylated CpG motifs activate Toll-like receptor 9 (TLR9), thereby triggering a diverse range of immunomodulatory responses. Although the immune-stimulating properties of CpG-ODNs have been extensively studied, their potential application as apoptosis-inducing tools remains underexplored, particularly for probiotic-derived sequences. In this study, we established a method for inducing extrinsic apoptosis in mouse splenocytes using MsST, a CpG-ODN identified from the lacZ gene of Streptococcus thermophilus ATCC19258.
