Huntington's disease: pathogenesis to animal models.

Huntington's disease (HD) is an inherited genetic disorder, characterized by cognitive dysfunction and abnormal body movements called chorea. George Huntington, an Ohio physician, described the disease precisely in 1872. HD is a dominantly inherited disorder, characterized by progressive neurodegeneration of the striatum but also involves other regions, primarily the cerebral cortex. The mutation responsible for this fatal disease is an abnormally expanded and unstable CAG repeat within the coding region of the gene encoding the huntingtin protein. Various hypotheses have been put forward to e

The role of the striatum in sentence processing: evidence from a priming study in early stages of Huntington's disease.

The role of sub-cortical structures such as the striatum in language remains a controversial issue. Based on linguistic claims that language … f declarative memory, and that grammatical rules are processed by the procedural system. Journal of Cognitive Neuroscience, 9(2), 266-276). Here we bolstered the striatum-rule hypothesis by investigating lexical abilities and rule application at the phrasal level. Both processing aspects were assessed in a model of striatal dysfunction, namely Huntington's disease (HD). …

Language learning impairments: integrating basic science, technology, and remediation.

One of the fundamental goals of the modern field of neuroscience is to understand how neuronal activity gives rise to higher cortical function. However, to bridge the gap between neurobiology and behavior, we must understand higher cortical functions at the behavioral level at least as well as we have come to understand neurobiological processes at the cellular and molecular levels. This is certainly the case in the study of speech processing, where critical studies of behavioral dysfunction have provided key insights into the basic neurobiological mechanisms relevant to speech perception and

The Smart Therapy Model: a Fourth Wave framework for assessment-driven, integrative multimodal biopsychosocial psychotherapy.

INTRODUCTION: Despite decades of psychotherapy research yielding over 100 empirically supported treatment protocols, clinical practice continues to face persistent challenges, including high dropout rates averaging 15-20% in controlled settings and widespread treatment resistance. The central tension is the "research-practice gap": whereas research validates singular, manualized protocols, clinicians predominantly operate eclectically. Critically, the field lacks a standardized framework to guide how clinicians should select and sequence interventions-not merely which protocols exist. Emerging

The trans-species core SELF: the emergence of active cultural and neuro-ecological agents through self-related processing within subcortical-cortical midline networks.

The nature of "the self" has been one of the central problems in philosophy and more recently in neuroscience. This raises various questions: (i) Can we attribute a self to animals? (ii) Do animals and humans share certain aspects of their core selves, yielding a trans-species concept of self? (iii) What are the neural processes that underlie a possible trans-species concept of self? (iv) What are the developmental aspects and do they result in various levels of self-representation? Drawing on recent literature from both human and animal research, we suggest a trans-species concept of self tha

Mouse plasmacytoma-expressed transcript 1 knock out induced 5-HT disruption results in a lack of cognitive deficits and an anxiety phenotype complicated by hypoactivity and defensiveness.

Serotonin (5-HT) is involved in many developmental processes and influences behaviors including anxiety, aggression, and cognition. Disruption of the serotonergic system has been implicated in human disorders including autism, depression, schizophrenia, and ADHD. Although pharmacological, neurotoxin, and dietary manipulation of 5-HT and tryptophan hydroxylase has added to our understanding of the serotonergic system, the results are complicated by multiple factors. A newly identified ETS domain transcription factor, Pet-1, has direct control of major aspects of 5-HT neuronal development. Pet-1

Multimodal data processing for affective perception in AI-supported developmental bibliotherapy: a mixed-methods feasibility study in university libraries.

INTRODUCTION: Multimodal data processing is increasingly important for understanding human affective experience in naturalistic settings. In this study, "multimodal" refers to the integration of heterogeneous human-centered evidence streams rather than synchronized perceptual or physiological sensing. We evaluated a low-threshold university-library support model integrating developmental bibliotherapy salons with AI chatbot-supported emotional expression. METHODS: A convergent mixed-methods feasibility and acceptability design was conducted at a Chinese university. Of 180 baseline respondents

The Tumor Neuroimmune Niche: A Unifying Framework for Cancer Progression and Its Neuropsychiatric Burden.

BACKGROUND: Cancer has traditionally been framed as a cell-intrinsic genetic disease and, more recently, as a disorder of the tumor microenvironment (TME). An emerging third view reframes malignancy as a systems-level neuroimmune disorder, in which neurons, glia, immune cells, and tumor cells form a continuously communicating network, the tumor neuroimmune niche. SUMMARY: This review synthesizes evidence that the nervous system actively regulates tumor initiation, proliferation, angiogenesis, invasion, and metastasis through autonomic, sensory, and neuroendocrine inputs, while tumors in turn

A Multi-Subject Tractography Atlas of Human Cerebellar Connections.

The cerebellum is a highly interconnected structure involved in motor coordination and balance as well as cognitive, sensory, autonomic, and emotional functions. Its connectivity relies on complex afferent and efferent white matter pathways linking it with the brainstem, spinal cord, thalamus, cerebral cortex, vestibular system, and limbic-related structures. Recent advances in diffusion MRI (dMRI) and tractography have enabled the in vivo reconstruction of these pathways, offering new opportunities to study cerebellar anatomy and connectivity. However, compared with supratentorial white matte

Multimodal subspace independent vector analysis effectively captures latent relationships between brain structure and function.

A key challenge in neuroscience is inferring relationships between brain structure and function from high-dimensional, multimodal neuroimaging data. While conventional multivariate approaches often simplify statistical assumptions and estimate one-dimensional independent sources shared across modalities, the true relationships between latent sources are likely more complex-statistical dependence may exist both within and between modalities and span more than one dimension per modality. Here, we introduce Multimodal Subspace Independent Vector Analysis (MSIVA), a method for capturing both joint

Nrf2/Keap1 Signaling Axis in the Brain: Master Regulator of Oxidative Stress in Neurodegenerative and Psychiatric Disorders.

Oxidative stress is a crucial factor in the development of CNS disorders, including neurodegenerative and psychiatric conditions. The Nrf2/Keap1 signaling axis plays a central role in defending against oxidative damage by regulating antioxidant and cytoprotective gene expression. Beyond its antioxidant function, Nrf2 influences neurogenesis, synaptic plasticity, mitochondrial bioenergetics, and glial neuronal interactions, all of which are vital for maintaining neural integrity and cognitive performance. Dysregulation of this pathway through altered dimerization, post-translational modificatio

Intracranial EEG referencing for large-scale category-selective mapping in the human ventral occipito-temporal cortex.

Intracranial EEG (iEEG) is increasingly used in many fields of human cognitive neuroscience since it offers a unique opportunity to directly record brain activity from awake humans at a high spatial and temporal resolution. However, little is known about the influence of the reference montage on the spatial and temporal characteristics of iEEG activity. Here, we compare the spatial and temporal profiles of neural activity for five reference montages (scalp reference, common average, zero reference, local Bipolar, and Laplacian) applied to a large dataset of depth electrodes (StereoElectroEncep…

From Synaptic Plasticity to Neurodegeneration: BDNF as a Transformative Target in Medicine.

The brain-derived neurotrophic factor (BDNF) has become one of the cornerstones of neuropathology, influencing synaptic plasticity, cognitive resilience, and neuronal survival. Apart from its molecular biology, BDNF is a powerful target for transformative benefit in precision medicine, leading to innovative therapeutic approaches for neurodegenerative and psychiatric diseases like Alzheimer's disease (AD), Parkinson's disease (PD), major depressive disorder (MDD), and post-traumatic stress disorder (PTSD). Nevertheless, clinical applicability is obstructed by hurdles in delivery, patient-speci

Prenatal stress, hormones, and fetal brain development: gender differences.

Prenatal stress is increasingly recognized as a critical factor influencing fetal brain development, with potential long-term risks for neurodevelopmental and psychiatric disorders. This review explores the effects of prenatal stress on the fetal brain, with particular emphasis on sex differences and the interplay between genetic, epigenetic, and environmental factors. The activation of the hypothalamic-pituitary-adrenal (HPA) axis in response to maternal stress leads to increased levels of glucocorticoids, which can cross the placenta and alter fetal brain structure and function. Key regions
