Defensive Effects of a Unique Polysaccharide, Sacran, to Protect Keratinocytes against Extracellular Stimuli and Its Possible Mechanism of Action.

Sacran, a polysaccharide isolated from the alga Aphanothece sacrum (Suizenji-nori), has unique physical and physiological characteristics. In a previous study, we reported that sacran improves skin conditions in individuals who suffer from atopic dermatitis (AD), focusing on its trapping function against extrinsic stimuli compared with hyaluronic acid (HA). First, we examined the penetration of sacran through stratum corneum (SC) with an impaired barrier function using immature reconstructed human epidermal equivalents. Sacran penetrates the SC to living cell layers of the epidermis, which sug

The drug ornidazole inhibits photosynthesis in a different mechanism described for protozoa and anaerobic bacteria.

Ornidazole of the 5-nitroimidazole drug family is used to treat protozoan and anaerobic bacterial infections via a mechanism that involves preactivation by reduction of the nitro group, and production of toxic derivatives and radicals. Metronidazole, another drug family member, has been suggested to affect photosynthesis by draining electrons from the electron carrier ferredoxin, thus inhibiting NADP+ reduction and stimulating radical and peroxide production. Here we show, however, that ornidazole inhibits photosynthesis via a different mechanism. While having a minute effect on the photosynth

Acute exposure to pure cylindrospermopsin results in oxidative stress and pathological alterations in tilapia (Oreochromis niloticus).

Cylindrospermopsin (CYN) is increasingly recognized as a potential threat to drinking water safety, due to its ubiquity. This cyanotoxin has been found to cause toxic effects in mammals, and although fish could be in contact with this toxin, acute toxicity studies on fish are nonexistent. This is the first study showing that single doses of CYN pure standard (200 or 400 μg CYN/kg fish bw) by oral route (gavage) generate histopathological effects in fish (Tilapia-Oreochromis niloticus) exposed to the toxin under laboratory condition. Among the morphological changes, disorganized parenchymal arc

Phosphonate utilization by the globally important marine diazotroph Trichodesmium.

The factors that control the growth and nitrogen fixation rates of marine diazotrophs such as Trichodesmium have been intensively studied be … rophs. The apparent absence of genes encoding a carbon-phosphorus lyase pathway in the other marine cyanobacterial genomes suggests that, relative to other phytoplankton, Trichodesmium is uniquely adapted for scavenging phosphorus from organic sources. This adaptation may help to explain the prevalence of Trichodesmium in low phosphate, oligotrophic systems.

Uncovering the cytotoxicity and growth inhibition of 14 Microcystis extracts on freshwater fish cell lines.

Freshwater cyanobacteria raise health and ecotoxicological concerns due to their production of secondary metabolites. The Microcystis genus synthesizes the well-known cyanotoxins, microcystins (MCs), but also various metabolites with documented sublethal and lethal effects. The toxic potential of such mixtures remains poorly characterized, although biota, including fish, are exposed to those metabolite cocktails through direct water uptake and/or trophic transfer. To provide preliminary answers to this complex issue, this study investigated the toxicity of 14 Microcystis extracts with distinct…

The small protein SbtC is a functional component of the CO2 concentrating mechanism in Synechocystis sp. PCC 6803.

Oxygenic phototrophs fix CO2 via the enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO), which shows relatively low CO2 affinity and specificity. To circumvent low and fluctuating CO2 concentrations in aquatic systems, cyanobacteria and algae have evolved sophisticated inorganic carbon (Ci) concentrating mechanisms (CCMs). Bicarbonate transporters such as SbtA play a crucial role in the cyanobacterial CCM and hence display multiple layers of tight regulation. Control of sbtA gene expression and corresponding transporter activity involves the PII-like protein SbtB, whose gene is f…

Environmental drivers of phytoplankton community structure and diatom size restructuring in the Yellow Sea.

The Yellow Sea (YS) is a productive marginal sea undergoing substantial environmental change associated with warming and hydrographic variability. We analyzed total and size-fractionated phytoplankton assemblages using high-performance liquid chromatography pigment analysis and microscopy across 12 seasonal surveys in 2020, 2022, and 2023. Diatoms dominated throughout the study period (56.7 ± 15.0%), with cryptophytes subdominant in winter, spring, and autumn and cyanobacteria and prymnesiophytes more prominent under warmer, nutrient-imbalanced summer conditions. Realized niche analysis separa…

In Vitro Evaluation of Virucidal Effect of Polysaccharides Extracted and Purified from Arthrospira platensis and Dunaliella salina on Human Adenovirus Type 5 in A549 Cells.

Polysaccharides derived from cyanobacteria and microalgae have attracted increasing interest as natural virucidal agents. Among them, polysaccharides from the cyanobacterium Arthrospira platensis (A. platensis), and the green microalgae Dunaliella salina (D. salina) have shown virucidal activities, mainly against enveloped viruses, while evidence on non-enveloped viruses is still limited. In this study, the virucidal activity of purified polysaccharides extracted from A. platensis (APPs) and from D. salina (DSPs) was evaluated in vitro against human adenovirus type 5 (HAdV5), a non-enveloped p…

Algal and Cyanobacterial Hydrolysates as Serum-Reducing Supplements Across Mammalian, Avian and Fish Cells with Identification of Bioactive Chromopeptides.

Maintaining sustainable production of healthy and nutritious food represents a major global challenge. Cultivated meat could reduce the environmental impact of livestock production, but it remains costly, largely due to the expense of fetal bovine serum, a common cell culture media supplement. Algal and cyanobacterial proteins, particularly phycobiliproteins, represent promising sustainable sources of potentially bioactive peptides that may contribute to the development of serum-reduced or serum-free culture media. This study investigated the potential of hydrolysates prepared using the protei…

Rescuing fertility: C-Phycocyanin prevents ovarian damage through NRF2-mediated ferroptosis pathways in polycystic ovary syndrome models.

Polycystic ovary syndrome (PCOS), a common female endocrine disorder marked by disrupted folliculogenesis and hyperandrogenism, is increasingly linked to oxidative stress. Despite this association, the mechanistic basis remains poorly characterized. C-Phycocyanin (C-PC), a cyanobacteria-derived protein with potent antioxidant properties, has demonstrated therapeutic potential for treating PCOS, though the molecular pathways mediating its effects have yet to be delineated. This study employed both a dehydroepiandrosterone (DHEA)-induced murine model and DHEA-challenged human granulosa cells (KG…

Combined effects of microcystin-LR and rice straw-derived biochar on the hepatic antioxidant capacity of zebrafish: Insights from LC-MS/MS-based metabolomics analysis.

Microcystin-LR (MC-LR) produced by cyanobacteria blooms poses a serious risk to aquatic organisms. Rice straw-derived biochar (BC) is gradually being utilized as an effective adsorbent to remove water pollutants. In the present study, the combined toxicity of MC-LR and BC on hepatic antioxidant capacity and metabolic phenotype of zebrafish (Danio rerio) were conducted due to the increasing concern of eutrophication in aquatic environments. Female zebrafish were exposed to solutions of MC-LR (10 μg/L) and BC (100 μg/L) individually and in combination for 30 days. The results indicated that sub-…

α-LA attenuates microcystin-LR-induced hepatocellular oxidative stress in mice through Nrf2-mediated antioxidant and detoxifying enzymes.

Microcystins constitute a class of toxins synthesized by cyanobacteria and are known to inflict significant damage on the antioxidant defense system of living organisms, primarily targeting the liver. α-Lipoic acid (α-LA) is universally recognized as a potent antioxidant in biological systems. It exerts its beneficial effects through multiple mechanisms-directly neutralizing reactive oxygen species (ROS) and free radicals, and indirectly enhancing antioxidant defenses by facilitating the regeneration of glutathione (GSH). However, the precise modus operandi of α-LA's protective effect against …
