Cardiovascular tolerance of mianserin and interactions of mianserin with other drugs.

The presented data clearly show that mianserin, which is therapeutically equal to other antidepressants, is a drug with a new chemical structure, with a most probably different mechanism of action and with distinct advantages above comparative drugs. Mianserin has not only pronouncedly fewer anticholinergic and central side-effects but: a) apparently has no cardiotoxic effects neither on healthy subjects nor on depressive individuals nor on cardiac patients; b) doses not cause orthostatic hypotension; c) is devoid of adverse drug-drug interactions with coumarin-like anticoagulants, with sympat…

[The interaction between drugs and oral anticoagulants of the coumarin group].

The problem of drug interactions during prolonged use of coumarin anticoagulants is important for most patients receiving anticoagulants take several other drugs. Frequent interaction causes either potentialisation of the anticoagulant effect with risk of hemorrhage or inhibition of the anticoagulant effect with risk of thrombosis. The various mechanisms which may explain most interactions are reviewed and the authors propose a list of drugs which should not be administered with coumarin.

Nefopam HCl interaction study with eight other drugs.

A controlled clinical trial was undertaken to determine potential for drug interaction of nefopam HCl, a new analgesic, with eight other widely used compounds. Nefopam HCl was administered in combination with these drugs and placebo to forty-five healthy volunteer subjects in nine groups of five subjects each. Possible drug interactions were detected by the occurrence of side-effects and interference with bioavailability of the new analgesic, also by changes in vital signs or in various laboratory tests. Results indicated no statistically significant differences in these parameters between the…

THE INTERACTION OF SOME STIMULANT AND DEPRESSANT DRUGS ON THE FROG HEART.

THE ACTIVITY OF FROG ISOLATED HEARTS WAS DEPRESSED BY ALTERING THE PERFUSING RINGER SOLUTION IN FIVE DIFFERENT WAYS: by reducing the calcium content, by increasing the potassium content, and by adding ether, thiopentone or acetylcholine. Depressed hearts were perfused with Ringer solution containing the following stimulant drugs: paullinia tannin, tannic acid, hydrogen peroxide, sodium oleate, sodium caprylate and ouabain. All these stimulant drugs had similar actions on hearts depressed by calcium lack, ether and thiopentone; hearts depressed by acetylcholine were, however, only weakly stimul

Detection of Clinically Significant Drug-Drug Interactions in Fatal Torsades de Pointes: Disproportionality Analysis of the Food and Drug Administration Adverse Event Reporting System.

BACKGROUND: Torsades de pointes (TdP) is a rare yet potentially fatal cardiac arrhythmia that is often drug-induced. Drug-drug interactions (DDIs) are a major risk factor for TdP development, but the specific drug combinations that increase this risk have not been extensively studied. OBJECTIVE: This study aims to identify clinically significant, high-priority DDIs to provide a foundation to minimize the risk of TdP and effectively manage DDI risks in the future. METHODS: We used the following 4 frequency statistical models to detect DDI signals using the Food and Drug Administration Adverse…

High prevalence of recreational and illicit drug use in German people living with HIV with a potential for drug-drug interactions with antiretroviral therapy.

Recreational drug use is higher in people living with HIV (PLHIV) than in the general population in Europe. This use increases the risk for drug-drug interactions (DDIs) and adverse events. We assessed the prevalence and clinical consequences of substance abuse among PLHIV. BESIDE was a cross-sectional, multi-center study in 2016/18, evaluating comorbidities, polypharmacy and recreational/illicit drug use in PLHIV on antiretroviral therapy (ART) in Germany. Legal and illicit drug use was recorded using two anonymous patient questionnaires one year apart (Q1 and Q2). The BESIDE study population…

HyHO-DDI: A hypergraph neural network for high-order Drug-Drug interaction prediction.

OBJECTIVE: Drug-drug interactions (DDIs) are a major cause of adverse drug reactions (ADRs), particularly in patients receiving multiple medications. Existing computational methods predominantly focus on pairwise interactions, limiting their ability to capture the complex effects of high-order (multi-drug) therapies. This study proposes a computational framework for predicting myopathy-associated high-order DDIs. METHODS: We propose HyHO-DDIs, a hypergraph neural network model for predicting myopathy-associated high-order drug combinations using a dataset curated from the FDA Adverse Event Re…

Asymmetric drug-drug interaction prediction based on diffusion-augmented graph attention network.

The prediction of potential Drug-Drug Interactions (DDIs) is crucial to reduce the harmful side effects from the combined use of multiple drugs. However, existing methods cannot comprehensively capture the DDI asymmetric information, resulting in inaccurate predictions, which increases the risk of harmful side effects. To address this, inspired by the advantages of directed graphs in representing asymmetric relationships, we propose a novel framework called Diffusion Graph Attention DDI (DiffGAT-DDI), designed to effectively capture the DDI asymmetric information to improve DDI prediction accu…

Evaluation of drug-drug interaction between rosuvastatin and tacrolimus and the risk of hepatic injury in rats.

Multimorbidity, therapeutic complexity, and polypharmacy, which greatly increases the risk of drug-drug interactions (DDIs) and adverse medical outcomes, have become important and growing challenges in clinical practice. Statins are frequently prescribed to manage post-transplant dyslipidemia and reduce overall cardiovascular risk in solid organ transplant recipients. This study aimed to determine whether rosuvastatin has significant DDIs with tacrolimus (the first-line immunosuppressant) and to evaluate the risk of hepatotoxicity associated with concomitant therapy. We first studied whether a…

Evaluation of the Cytochrome P450 3A and P-glycoprotein Drug-Drug Interaction Potential of Futibatinib.

Futibatinib, a selective, irreversible fibroblast growth factor receptor 1-4 inhibitor, is being investigated for tumors harboring FGFR aberrations and was recently approved for the treatment of FGFR2 fusion/rearrangement-positive intrahepatic cholangiocarcinoma. In vitro studies identified cytochrome P450 (CYP) 3A as the major CYP isoform in futibatinib metabolism and indicated that futibatinib is likely a P-glycoprotein (P-gp) substrate and inhibitor. Futibatinib also showed time-dependent inhibition of CYP3A in vitro. Phase I studies investigated the drug-drug interactions of futibatinib wi…

Impact of valproic acid on busulfan pharmacokinetics: In vitro assessment of potential drug-drug interaction.

Busulfan (Bu) is an alkylating agent commonly used at high doses in the preparative regimens of hematopoietic stem cell transplantation (HSCT). It has been shown that such high doses of Bu are associated with generalized seizures which are usually managed by prophylactic antiepileptic drugs (AEDs) such as valproic acid (VPA). Being a strong enzyme inhibitor, VPA may inhibit Bu metabolism and thus increase its potential toxicity. Despite its clinical relevance, the potential interaction between Bu and VPA has not yet been evaluated. The aim of the present study was to assess and evaluate the po

Drug-drug interaction and acute kidney injury development: A correlation-based network analysis.

BACKGROUND: Drug-induced nephrotoxicity is a relatively common preventable cause of acute kidney injury (AKI), providing early recognition and management. The pharmacokinetics or pharmacodynamics of drug-drug interactions may lead to additive or synergistic toxicity. The influx of new medications or off-label use of medications in the critical care setting can lead to additional nephrotoxicities, often challenging to predict or detect. This study evaluates the patterns of medication utilization, their combinations, and the related associations with AKI. METHODS: We utilized correlation-based …
