Positive allosteric modulation of µ-opioid receptor - A new possible approach in the pain management?

The antinociceptive effect of the opioid drugs is achieved through activation of the µ-opioid receptor (MOP). The orthosteric and allosteric sites of opioid receptors may be modulated, orthosteric site by endogenous i.e.β-endorphin and exogenous opioids (morphine, oxycodone, fentanyl); whereas BMS-986121, BMS-986122, Comp5, MS1, Ignavine or even oxytocin act on the allosteric site of the MOP. Opioid therapy is associated with numerous side effects, such as: respiratory depression, sedation, constipation, and importantly, prolonged therapy can influence the development of tolerance, overdose, a

Structures of the entire human opioid receptor family.

Opioids are effective analgesics, but their use is beset by serious side effects, including addiction and respiratory depression, which contribute to the ongoing opioid crisis. The human opioid system contains four opioid receptors (μOR, δOR, κOR, and NOPR) and a set of related endogenous opioid peptides (EOPs), which show distinct selectivity toward their respective opioid receptors (ORs). Despite being key to the development of safer analgesics, the mechanisms of molecular recognition and selectivity of EOPs to ORs remain unclear. Here, we systematically characterize the binding of EOPs to O

Molecular cloning and tissue distribution of Crh and Pomc mRNA in the fat-tailed dunnart (Sminthopsis crassicaudata), an Australian marsupial.

Like all vertebrates, marsupials respond to stressors with the activation of the hypothalamo-pituitary-adrenal axis. However, peptides operating at the higher regulatory levels of this hormonal system, i.e. corticotropin-releasing hormone (CRH) and adrenocorticotropic hormone (ACTH), have not been investigated in marsupials. Here we report the molecular cloning of the precursor cDNAs of CRH (prepro-CRH) and of ACTH (proopiomelanocortin; POMC) in an Australian marsupial, the fat-tailed dunnart (Sminthopsis crassicaudata). Dunnart POMC and prepro-CRH are predicted to be peptides of 399 and 200 a

Opioids and skin homeostasis, regeneration and ageing - What's the evidence?

What has the opioid receptor system, known for beneficial as well as disastrous effects in the central nervous system, to do with skin? The question is appropriate considering the fact that the nervous system and the skin both derive from the ectoderm. As part of the skin neuroendocrine system, the opioid receptor system exemplifies the closeness between the nervous system and the skin. Overexpression of the δ-opioid receptor in keratinocytes yields dysregulation of involucrin, loricrin, and filaggrin, proteins essential to the integrity of the skin barrier. The μ-opioid receptor ligand β-endo

Effect of epidural and intravenous clonidine on the neuro-endocrine and immune stress response in patients undergoing lung surgery.

The effects of intravenous and epidural clonidine, 4 microg kg-1, combined with epidural morphine, 40 microg kg-1, on the neuro-endocrine and immune stress responses to thoracic surgery are reported. A control group received only epidural morphine. Anaesthesia was induced and maintained with propofol. Catecholamines, vasopressin, cortisol, beta-endorphin concentrations and leucocyte counts were measured before drug administration, immediately after intubation of the trachea, after thoracotomy and at the end of surgery. Catecholamines did not change in any of the groups. The other stress hormon

Pharmacologic manipulation of the respiratory control center in the infant.

The respiratory control center receives afferent stimuli from mechanical and neuromechanical sources. Information from both these sources, combined with voluntary and involuntary CNS control, effects stimulation of the respiratory muscles. In the infant, insufficiency of one or more of these elements of the respiratory control center is associated with considerable morbidity and mortality. Pharmacologic manipulation may provide a means of intervention. The xanthine derivative theophylline has been successfully used in the treatment of bronchopulmonary dysplasia and apnea in the infant. Naloxon

Brain opioid peptides may participate in the reversal of pentylenetetrazol-induced amnesia.

Post-training administration of pentylenetetrazol (PTZ, 45 mg/kg, i.p.) disrupted 48-h retention, in mice, of an inhibitory avoidance response. The effect was reversed by any of the following treatments given 1 h prior to testing: a) a beta-endorphin injection (0.1 microgram/kg, i.p.), b) PTZ injection, or c) exposure to a novel experience (10 min in a stainless steel box with a wire mesh top). All treatments had a similar time course of effectiveness (up to at least 3 h) and their effects were blocked by naltrexone (0.1 mg/kg, i.p.) but not by naltrexone methyl bromide (10 mg/kg, i.p.). These

[Hemothorax disclosing hemorrhagic necrosis of a pheochromocytoma: circumstance for detection of multiple endocrine neoplasia type IIa. Detection of 5 familial cases].

Whilst investigating 26 members of the same family, we discovered 5 cases of multiple endocrine neoplasia type II a. The present report demonstrates the diagnostic value of basal plasma thyrocalcitonin (TCT) assays, before and after stimulation with pentagastrin, and of plasma carcinoembryonic antigen (CEA) assays. Some of the clinical features encountered were novel--e.g. in one patient the phaeochromocytoma was revealed by a haemothorax with cardiovascular collapse--and others were peculiar; thus, in 4 cases the medullary thyroid carcinoma (MTC) was less than 2 cm in diameter and without lym

Effects of eight-hour naloxone infusions on human subjects.

Twelve normal male volunteers received saline control, low-dose naloxone, and high-dose naloxone infusions during three weekly sessions. The sessions were 16 hr long: 1 hr for predrug assessments, 8 hr during which either naloxone or saline was infused in a double-blind procedure, and a 7-hr postdrug observation period. The 8-hr infusions of naloxone had no effect on experimental ischemic arm pain. In addition, the ischemic arm pain procedure did not significantly increase either plasma levels of cortisol or immunoreactive beta-endorphin, suggesting that the procedure was not stressful. The hi

Role of pro-opiomelanocortin-derived peptides in the regulation of steroid production by human fetal adrenal cells in culture.

Synthetic (1-39)ACTH, (1-24)ACTH, (18-39)ACTH, alpha-MSH, met-enkephalin and alpha-, beta- and gamma-endorphin were tested for their ability to stimulate steroidogenesis by human fetal adrenal cells in culture. Adrenal cells were incubated with peptide hormones for two periods of 24h. On the third day of the experiment the cells were incubated with progesterone (4 micrograms/2 ml) for 8 h. At the doses tested only (1-39)ACTH, (1-24)ACTH and alpha-MSH stimulated steroidogenesis. None of the other peptides had any corticotrophic effect on the formation of cortisol, corticosterone or dehydroepian

Hypothalamic-pituitary-adrenal axis, neuroendocrine factors and stress.

The stress system coordinates the adaptive responses of the organism to stressors of any kind.(1). The main components of the stress system are the corticotropin-releasing hormone (CRH) and locus ceruleus-norepinephrine (LC/NE)-autonomic systems and their peripheral effectors, the pituitary-adrenal axis, and the limbs of the autonomic system. Activation of the stress system leads to behavioral and peripheral changes that improve the ability of the organism to adjust homeostasis and increase its chances for survival. The CRH and LC/NE systems stimulate arousal and attention, as well as the meso

Role of thymic peptides as transmitters between the neuroendocrine and immune systems.

Thymic peptides, a heterogenous family of polypeptidic hormones synthesized within the thymus, not only exert important regulatory effects within both the immune and neuroendocrine systems but are also themselves subject to control by hormones derived from the hypothalamic-pituitary-adrenal axis (HPA) and other endocrine glands. Regarding thymic hormonal function, thymulin production is up-regulated by several hormones, including prolactin, growth hormone and thyroid hormones. Other aspects of the physiology of thymic epithelial cells can also be modulated by hormones and neuropeptides, partic

Alcohol, the reward system and dependence.

Evidence is presented to show that multiple neurotransmitter systems of the brain reward systems including GABA, glutamate, dopamine, serotonin and opioid peptides are involved in alcohol reinforcement. Dependence is associated with changes in many of these same systems, but also with changes in other neurotransmitters, such as brain corticotropin releasing factor. A midbrain forebrain circuitry that involves parts of the nucleus accumbens and amygdala is hypothesized to be the focus for the neuropharmacology of alcohol reinforcement.

Melatonin treatment delays reproductive aging of female rat via the opiatergic system.

In female rat age-related reproductive decline is accompanied by progressive impairment of the neuroendocrine mechanisms that regulate LH secretion. The biosynthetic activity of the pineal gland is markedly depressed and the nocturnal secretion of melatonin decreases significantly. The aim of the present study was to evaluate whether the nocturnal administration of melatonin via the drinking water (0.4 micrograms/ml) throughout the course of aging from 14 to 24 months of age could (1) influence the age-related changes that occur in basal serum levels of LH and in the LH response to GnRH or to

Determinants of the nature of environmentally induced hypoalgesia.

Factors that determine whether naltrexone and pentobarbital anesthesia block the hypoalgesia produced by electric shock were investigated. The number of shocks delivered, the time since the last shock, and whether the subject is removed from the shock environment for testing were all found to be critical. One shock was followed by an initial hypoalgesia that was reversed by naltrexone but was not affected by pentobarbital. Moreover, this initial hypoalgesia was not eliminated by removing the subject from the shock apparatus. When testing was conducted in the shock context, this early hypoalges

The role of neurotransmitters and opioids in polycystic ovarian syndrome.

In this article, the characteristic gonadotropin dysfunction of PCOD is discussed. The potential role of modulators such as the neurotransmitters dopamine and norepinephrine as well as the role of endogenous opiates are considered. A review of the effect of these neuromodulators is followed by speculation as to whether disordered secretion of these substances may be operative in the pathophysiology of PCOD.

[Studies of immunosuppressive substances in human seminal plasma].

Endogenous opioids, which have been shown to exhibit pharmacological and physiological activity on the immune system, have recently been reported to inhibit rosette formation between human T lymphocytes and sheep red blood cells. Because those opioids were found at a higher concentration in seminal plasma than in plasma, we studied whether seminal plasma could influence E rosette formation in 23 patients who complained of infertility. Seminal plasma significantly inhibited E rosette formation. With the dilution of seminal plasma, the percentage of E rosette formation increased. The incubation

Opioid modulation of baroreceptor reflex sensitivity in dogs.

Opioid peptides can modulate cardiovascular function both directly and by modifying specific responses. The action of opioids on baroreceptors was evaluated in anesthetized dogs. Baroreceptor responsiveness was determined by observing the peak heart rate and blood pressure responses as well as by evaluating the slope of a line relating heart period to blood pressure while pharmacologically altering blood pressure with doses of phenylephrine hydrochloride and sodium nitroprusside. These tests were performed before and after administration of naloxone (40 micrograms/kg). Naloxone significantly b
